Populations of F6 recombinant inbred lines, generated by single seed descent from a half diallel among eight bread wheat lines adapted to the East African highlands, were used to identify those crosses that were more likely to produce cultivars which combined resistance to yellow rust with improved yield. Crosses having the most resistant line as one parent offered the best prospect of success, particularly those which produced F1hybrids exhibiting better parent heterosis. For plot grain yield there was a highly significant correlation between the observed and predicted rankings of the recombinant inbred line populations for the proportion of individual lines equalling or surpassing the target value. For yellow rust severity, however, this correlation was non-significant when a target value of zero was used. Adopting a slightly less stringent target of 0.25, coupled with the omission of two aberrant populations, increased this correlation significantly. Oxidative autoactivation of latent collagenase by human neutrophils   总被引:32,自引:0,他引:32  
The pathological destruction of collagen plays a key role in the development of inflammatory disease states affecting every organ system in the human body. Neutrophils localized at inflammatory sites can potentially degrade collagen by releasing a metalloenzyme, collagenase, which is stored in a latent inactive form. Triggered human neutrophils were shown to release and simultaneously activate their latent collagenase. The activation of the latent enzyme was coupled to an oxidative process that required the generation of a highly reactive oxygen metabolite, hypochlorous acid. Oxidative regulation of latent collagenase activity may be important in the pathogenesis of connective tissue damage in vivo.  相似文献

Two methods for the determination of iron in olive oil by spectrophotometry and by adsorptive stripping square wave voltammetry (Ad-SSWV) have been developed. These two methods are based on the formation of a 5,5-dimethylcyclohexane-1,2,3-trione 1,2-dioxime 3-thiosemicarbazone (DCDT)-iron(II) complex in strongly acid media. In both, iron is extracted from the olive oil by using HCl. Spectrophotometric determination of iron with DCDT is based on the feature that the DCDT-Fe complex shows an absorbance maximum at 550 nm. A calibration graph has been constructed from 0 to 4000 ng mL(-)(1), and the detection limit was 115 ng mL(-)(1) (57 ng g(-)(1) in olive oil). On the other hand, the voltammetric determination of the metal is based on the appearance of a peak due to an adsorptive reductive process of the complex that it is observed when the Ad-SSWV technique is used. A calibration graph has been constructed from 0 to 30 ng mL(-)(1), and the detection limit was 0.55 ng mL(-)(1) (13.75 ng g(-)(1) in olive oil according to the proposed procedure).  相似文献

The retention walls in a pond containing the residues from the pyrite mine of Aznalcóllar (southern Spain) broke open on 25 April 1998, spilling approximately 6 × 106 m3 of polluted water and toxic tailings, which affected some 55 km2. Drying and aeration of the tailings resulted in oxidation, forming an acidic solution with high pollutant contents, the effects of which were studied in a calcareous soil. The infiltration of this solution markedly affected only the first 12 mm of the soil, where strong acidification caused the weathering of the carbonates, and where the fine mineral particles were hydrolysed. The SO42− ions in the acidic solution precipitated almost entirely at this depth, forming gypsum, hydroxysulphates and complex sulphates. The Fe3+ ions also precipitated there, mainly in amorphous or poorly crystallized forms, adsorbing to As, Sb, Tl and Pb dissolved in the acidic solution. The Al3+ ions, though partly precipitating in the acidic layer, accumulated mostly where the soil pH exceeded 5.5 (12–14 mm in depth). They did so primarily as amorphous or poorly crystallized forms, adsorbing to Cu dissolved in the acidic solution. The Zn2+ and Cd2+ ions accumulated mainly at pH > 7.0 (19–21 mm in depth), being adsorbed chiefly by clay mineral. After 15 months, only the first 20 mm of the soil were acidified by the oxidation of the tailings and most of the pollutants did not penetrate deeper than 100 mm. Consequently, the speed of the cleanup of the toxic spill is not as important as a thorough removal of tailings together with the upper 10 cm of the soil.  相似文献

Five tetraploid clones of potato were crossed to each of eight diploid first division restitution (FDR) 2n pollen producers and four tetraploid males using a line × tester mating design. A total of 59 families were obtained and evaluated under long days at Rhinelander (USA). A subset of 49 families were grown under short days at four Peruvian locations. Tuber yield (kg per plant) of 4x-2x families was similar to, or greater than, that of 4x-4x families in short day environments. The diploid clones had breeding values greater than or equal to those of the tetraploid clones. Only families derived from FDR 2n pollinators combined high tuber yield with stability and high dry matter content. Reciprocal recurrent selection would be the best breeding scheme, in which the diploids are testers of the tetraploids and vice-versa. Intra-population improvement could be achieved through phenotypic recurrent selection. The best materials from both breeding pools could then be crossed to produce tetraploid hybrids through unilateral sexual polyploidization (4x-2x crosses).  相似文献

Accurate assessments of genetic gains ensuing from plant breeding for the most important agronomic characteristics in Nordic spring barley (Hordeum vulgareL.) are not available. Hence this research was aimed to determine the rate of genetic improvement in the Nordic barley breeding pool. This study included 90, 2-row spring barley cultivars released (1942–1988) and29, 6-row spring barley cultivars released(1930–1991) adopted by Nordic farmers that were tested in four Nordic locations for three consecutive years. Relative genetic gain owing to plant breeding was 13% in2-row barley and 34% in 6-row barley for grain yield. The absolute gain for this characteristic was 13 ± 3 kg ha-1 year -1 in 2-row barley, and22 ± 3 kg ha-1 year-1 in6-row barley. Improved yield was achieved in Nordic barley by reducing plant height(0.20 ± 0.04 cm year-1 for 2-rowbarley and 0.16 ± 0.06 cm year-1for 6-row cultivars), thereby reducing significantly lodging (0.5 ± 0.1%year-1 and 0.4 ± 0.1year-1), and increasing significantly the harvest index (0.0008 ± 0.0002year-1 and 0.0018 ± 0.0002year-1). Additionally, in 2-row spring barley cultivars resistance to powdery mildew (0.19 ± 0.08% year-1)and thousand-kernel weight (0.07 ±0.03 g year-1) were also significantly enhanced, whereas hectoliter weight was improved (0.06 ± 0.02 kg year-1)in 6-row barley cultivars in the period investigated.   1. Ecological stability based on statistical variability was contrasted with the asymptotical local stability of transient states during successional changes in community structure in kelp forests and barren grounds.
  2. Simplified semi-quantitative ecological models with different levels of complexity were constructed to represent transient successional states in kelp and barrens, and Routh–Hurwitz's criteria and Levins' criterion of local stability were also estimated.
  3. The results suggest that ecological stability based on statistical variation does not match local (mathematical) stability, since although successional changes in kelp exhibited less variability (measured as percentage coefficient of variation), barren beds were more locally stable. The local stability criteria used in the current work could be a suitable technique for evaluating the likelihood of transient ecological states to resist or change, since it considers states at equilibrium to maintain their properties only against small disturbances.
  4. These outcomes are relevant because the natural alternation between kelp and barrens could be disrupted by the intensive harvest of kelp species carried out over the last 10 years on the central-north Chilean coast and in other temperate ecosystems.
  5. Additionally, the negative effects of harvesting kelp species could have a synergetic relationship with other drivers such as climate change and coastal pollution. Thus, strict policy managements to limit the kelp harvest are deeply encouraged. A monitoring programme for assessing the current conditions of benthic–pelagic ecosystems along the Chilean coast is also required.

  • 1. A dynamical and spatial simulation model of a harvested benthic ecosystem of central northern Chile (Tongoy Bay) was constructed using the ECOSPACE software package.
  • 2. In this system the red alga (Chondrocanthus chamissoi), the scallop (Argopecten pupuratus), the gastropod (Xanthochorus cassidiformis) and the crab (Cancer polyodon) are harvested intensively. The impacts of harvesting these resources exclusively in the seagrass, sand‐gravel, and in the sand habitats, as well as, in the seagrass and sand‐gravel and in all habitats were assessed. The goal was to explore policies of sustainable exploitation of the benthic systems.
  • 3. The most important findings were: (a) Fishing exclusively in either the seagrass or sand habitats produces a population increase in the sea star Luidia magallanica, in the seagrass Heterozostera tasmanica, and in the crab Paraxanthus barbiger. (b) Exclusive fishing in the sand‐gravel habitat causes only small effects on the species and groups, which suggests that this habitat is the most resistant to harvest. (c) The simultaneous fishing on two or three habitats would produce the largest negative effect on the entire system. Therefore, a habitat rotation fishery is recommended.
  • 4. Our study suggests that trophic‐spatially explicit models offer great possibilities for the screening and planning of effective interventions or manipulations of natural systems.

Wheat cultivars differ widely in manganese (Mn) efficiency. To investigate the reasons for different Mn efficiencies, a pot experiment with soil, a solution‐culture experiment, and model calculations were carried out. The pot experiment was conducted with wheat (Triticum aestivum L. cvs. PBW 373, PBW 154, PBW 343, PBW 138, and Triticum durum L. cvs. PBW 34 and PDW 233) grown in a screen house in India. The soil was a loamy sand with pH 8.1, DTPA‐extractable Mn 1.62 mg (kg soil)–1, and initial soil solution Mn concentration (CLi) of 0.19 μM. When fertilized with 50 mg Mn (kg soil)–1, CLi increased to 0.32 μM. At CLi 0.19 μM, wheat cv. PBW 373 produced 74% of its maximum shoot dry weight (SDW) with 64% of its maximum root length (RL), while cv. PDW 233 produced only 25% of its maximum SDW with 11% of its maximum RL. The other wheat cultivars were between these extremes. Manganese deficiency caused a reduction in shoot growth, but more strongly reduced root growth. The low Mn efficiency of T. durum cv. PDW 233 was related to a strong depression of its root growth. Manganese influx was similar for all cultivars. In solution culture below 1 μM Mn, under controlled climate‐chamber conditions, Mn influx was linearly related to Mn concentration. Both the efficient cv. PBW 343 and the inefficient cv. PDW 233 had a similar influx. Uptake kinetic parameters from the solution experiment together with soil and plant parameters from the pot experiment were used in a mechanistic nutrient‐uptake model. Calculated values of Mn influx for wheat grown in soil were 55% to 74% of measured values. A sensitivity analysis showed that increasing CLi or the slope of the uptake isotherm by about 30% would be enough to reach the observed influx. The results of this research indicate that an increase of Mn solubility by microbial or chemical mobilization would increase Mn uptake. But on the other hand, no chemical mobilization would be required to increase Mn uptake if the plant improved its uptake kinetics. Low Mn efficiency of some wheat cultivars was related to their reduced root growth at low soil Mn supply.  相似文献
