Toxoplasma gondii, a zoonotic protozoal parasite, is well-known for its global distribution and its ability to infect virtually all warm-blooded vertebrates. Nonetheless, attempts to describe the population structure of T. gondii have been primarily limited to samples isolated from humans and domesticated animals. More recent studies, however, have made efforts to characterize T. gondii isolates from a wider range of host species and geographic locales. These findings have dramatically changed our perception of the extent of genetic diversity in T. gondii and the relative roles of sexual recombination and clonal propagation in the parasite's lifecycle. In particular, identification of novel, disease-causing T. gondii strains in wildlife has raised concerns from both a conservation and public health perspective as to whether distinct domestic and sylvatic parasite gene pools exist. If so, overlap of these cycles may represent regions of high probability of disease emergence. Here, we attempt to answer these key questions by reviewing recent studies of T. gondii infections in wildlife, highlighting those which have advanced our understanding of the genetic diversity and population biology of this important zoonotic pathogen.  相似文献

Real-time PCR directed to intergenic spacer (IGS) noncoding region between 16S and 23S rRNA genes was used for species specific detection of Mycoplasma felis in conjunctival scrapings. Samples were collected from 57 cats suffering from chronic conjunctivitis in 2008-2010 (Wroc?aw, Poland). Samples from 36 cats (63.2%) were shown to be positive for Mycoplasma felis. Our research gives a first insight in the occurrence of Mycoplasma felis among domestic cats in Poland suggesting that this pathogen may constitute an underestimated cause of chronic conjunctivitis.  相似文献

Several toy breed dogs are predisposed to syringomyelia (SM), a spinal cord disorder, characterised by fluid-filled cavitation. SM is a complex trait with a moderately high heritability. Selective breeding against SM is confounded by its complex inheritance, its late onset nature and high prevalence in some breeds. This study investigated the early outcome of existing SM breeding guidelines. Six hundred and forty-three dogs, 550 Cavalier King Charles spaniels (CKCS) and 93 Griffon Bruxellois (GB), were identified as having either one (454 dogs) or both parents (189 dogs) with MRI-determined SM status. Offspring without SM were more common when the parents were both clear of SM (SM-free; CKCS 70 per cent, GB 73 per cent). Conversely, offspring with SM were more likely when both parents had SM (SM-affected; CKCS 92 per cent, GB 100 per cent). A mating of one SM-free parent with an SM-affected parent was risky for SM affectedness with 77 per cent of CKCS and 46 per cent of GB offspring being SM-affected. It is recommended that all breeding dogs from breeds susceptible to SM be MRI screened; that the SM status at five years old is established; and all results submitted to a central database that can be used by dog breeders to better enable mate selection based on estimated breeding values.  相似文献

Blood was obtained from 61 neonatal Holstein calves originating from a farm in Germany with a high incidence of bovine neonatal pancytopenia (BNP). In order to detect alterations that might be related to BNP, selected haematological analytes were determined. Haematological examinations demonstrated alterations in at least two of the three cell lineages in 10 calves (16.39 per cent). Six animals (9.84 per cent) developed a bleeding disorder indicative of BNP at approximately two weeks of age. None of these animals showed alterations in complete blood cell count at sampling in the first week of life. In weeks when calves with BNP were born, an increase in the number of apparently healthy calves demonstrating decreases in blood cell counts was observed.  相似文献

Background: Diagnosis of canine idiopathic pulmonary fibrosis (IPF) is challenging. Endothelin‐1 (ET1) is a biomarker of IPF in humans, but whether ET1 can detect and differentiate IPF from other canine respiratory diseases is unknown. Objective: To evaluate whether measurement of the concentration of ET1 in serum and bronchoalveolar lavage fluid (BALF) can be used to distinguish canine IPF from chronic bronchitis (CB) and eosinophilic bronchopneumopathy (EBP). Animals: Twelve dogs with IPF, 10 dogs with CB, 6 dogs with EBP, 13 privately owned healthy West Highland White Terriers (WHWT), and 9 healthy Beagle dogs. Methods: Prospective, case control study. ET1 concentration was determined by ELISA in serum and in BALF. Results: No significant difference in serum ET1 concentration was detected between healthy Beagle dogs and WHWT. Serum ET1 concentration was higher in dogs with IPF (median interquartile range; 2.32 pg/mL, 2.05–3.38) than healthy Beagle dogs (1.28, 1.07–1.53; P < .001), healthy WHWT (1.56, 1.25–1.85; P < .001), dogs with EBP (0.94 0.68–1.01; P = .001), and dogs with CB (1.54 0.74–1.82; P = .005). BALF ET1 concentration was below the detection limit in healthy WHWT and in dogs with CB, whereas it was measurable in all dogs with IPF. A cut‐off serum concentration of 1.8 pg/mL had a sensitivity of 100% and a specificity of 81.2% for detection of IPF, with an area under the receiver operating characteristic curve of 0.818. Conclusions and Clinical Importance: Serum ET1 can differentiate dogs with IPF from dogs with EBP or CB. ET1 can be detected in BALF of dogs with IPF.  相似文献

The aim of this study was to compare genetic gain for a traditional aquaculture sib breeding scheme with breeding values based on phenotypic data (TBLUP) with a breeding scheme with genome-wide (GW) breeding values. Both breeding schemes were closed nuclei with discrete generations modeled by stochastic simulation. Optimum contribution selection was applied to restrict pedigree-based inbreeding to either 0.5 or 1% per generation. There were 1,000 selection candidates and a sib test group of either 4,000 or 8,000 fish. The number of selected dams and sires to create full sib families in each generation was determined from the optimum contribution selection method. True breeding values for a trait were simulated by summing the number of each QTL allele and the true effect of each of the 1,000 simulated QTL. Breeding values in TBLUP were predicted from phenotypic and pedigree information, whereas genomic breeding values were computed from genetic markers whose effects were estimated using a genomic BLUP model. In generation 5, genetic gain was 70 and 74% greater for the GW scheme than for the TBLUP scheme for inbreeding rates of 0.5 and 1%. The reduction in genetic variance was, however, greater for the GW scheme than for the TBLUP scheme due to fixation of some QTL. As expected, accuracy of selection increased with increasing heritability (e.g., from 0.77 with a heritability of 0.2 to 0.87 with a heritability of 0.6 for GW, and from 0.53 and 0.58 for TBLUP in generation 5 with sib information only). When the trait was measured on the selection candidate compared with only on sibs and the heritability was 0.4, accuracy increased from 0.55 to 0.69 for TBLUP and from 0.83 to 0.86 for GW. The number of selected sires to get the desired rate of inbreeding was in general less in GW than in TBLUP and was 33 for GW and 83 for TBLUP (rate of inbreeding 1% and heritability 0.4). With truncation selection, genetic gain for the scheme with GW breeding values was nearly twice as large as a scheme with traditional BLUP breeding values. The results indicate that the benefits of applying GW breeding values compared with TBLUP are reduced when contributions are optimized. In conclusion, genetic gain in aquaculture breeding schemes with optimized contributions can increase by as much as 81% by applying genome-wide breeding values compared with traditional BLUP breeding values.  相似文献

The experimental materials comprised 44 hybrid [female (Polish Large White x Polish Landrace) x male Duroc] growing-finishing pigs. The animals were randomly divided into two groups: 24 pigs were slaughtered immediately after transport and 20 pigs were slaughtered after a 24-hour rest period in the lairage. The meat content of pork carcasses, carcass dressing percentage, the proximate chemical composition, physicochemical and sensory properties of meat and shear force values were determined. Serum creatine kinase activity and cortisol levels were determined in blood samples collected before transport and during carcass bleeding. Pigs slaughtered immediately after transport, compared with those slaughtered after a 24-hour rest period, were characterized by a higher meat content of the carcass and a higher carcass dressing percentage. Pre-slaughter handling had no effect on pork quality. The incidence of normal-quality meat, partially PSE (pale, soft, exudative) meat and PSE meat was similar in both groups. Chemical analysis showed that the content of dry matter, total protein, fat and minerals in meat was comparable in both groups. As regards the functional properties of the pork, samples from the carcasses of pigs that had rested before slaughter had a higher contribution of the red color component. Meat from pigs slaughtered immediately after transport had more desirable sensory properties. Pre-slaughter resting had a significant effect on those analyzed physiological parameters which were found to be good indicators of pre-slaughter stress. Serum creatine kinase activity and cortisol levels were higher in blood samples collected after transport (during carcass bleeding) than in samples collected before transport, pointing to a strong stress response of animals to pre-slaughter treatment. The decrease in serum cortisol levels in blood samples collected during bleeding from the carcasses of pigs slaughtered after a 24-hour rest period, compared with samples collected from animals slaughtered immediately after transport, suggests that rest before slaughter alleviated stress induced by pre-slaughter handling operations.  相似文献

Twelve ruminally cannulated crossbred Angus steers were used to evaluate ruminal fermentation characteristics and diet digestibility when 30% (DM) corn dried distillers grains with solubles (DDGS) containing 0.42 or 0.65% (DM) of dietary S was incorporated into finishing diets based on steam-flaked corn (SFC) or dry-rolled corn (DRC). The study was a replicated, balanced randomized incomplete block design with a 2 × 2 factorial arrangement of treatments. Factors consisted of dietary S concentration (0.42 and 0.65% of DM; 0.42S and 0.65S, respectively) and grain processing method (SFC or DRC). The 0.65S concentration was achieved by adding H(2)SO(4) to DDGS before mixing rations. Steers were assigned randomly to diets and individual, slatted-floor pens, and fed once daily for ad libitum intake. Two 15-d experimental periods were used, each consisting of a 12-d diet adaptation phase and a 3-d sample collection phase. Samples were collected at 2-h intervals postfeeding during the collection phase. Ruminal pH was measured immediately after sampling, and concentrations of ruminal ammonia and VFA were determined. Fecal samples were composited by steer within period and used to determine apparent total tract digestibilities of DM, OM, NDF, CP, starch, and ether extract. Feeding 0.65S tended (P = 0.08) to decrease DMI but resulted in greater apparent total tract digestibilities of DM (P = 0.04) and ether extract (P = 0.03). Ruminal pH increased (P < 0.05) in steers fed 0.65S diets, which may be attributable, in part, to decreased (P = 0.05) VFA concentrations and greater (P < 0.01) ruminal ammonia concentrations when 0.65S was fed, compared with feeding 0.42S. These effects were more exaggerated in steers fed DRC (interaction, P < 0.01), compared with steers fed SFC. Steers fed DRC-0.65S had greater (P < 0.01) acetate concentration than steers fed DRC-0.42S, but acetate concentration was not affected by S concentration when SFC was fed. Propionate concentration was decreased (P < 0.01) in steers fed SFC-0.65S compared with steers fed SFC-0.42S, but dietary S concentration had no effect on propionate concentration when DRC was fed. Butyrate concentration was less (P < 0.01) in steers fed 0.65S diets than in steers fed 0.42S. Lactate concentrations tended (P = 0.06) to decrease in steers fed 0.65S diets. Feeding DDGS with increased S concentration may decrease feed intake and ruminal VFA concentration but increase ruminal ammonia concentration.  相似文献

Reasons for performing study: To describe the clinical symptoms, treatment, and outcome of meniscal cysts in horses. These structures have not been previously described in the literature as a potential cause of lameness in the horse. Hypothesis: Meniscal cysts are an uncommon condition of the femorotibial joint but can be a significant cause of lameness. Symptoms can be resolved by arthroscopic excision. Methods: Records of horses diagnosed with meniscal cysts and treated by cyst excision and meniscal debridement at 2 surgical practices were reviewed. Clinical outcome was determined by repeat veterinary examination and contact with owner. Results: Seven cases of meniscal cyst were treated with arthroscopic cyst excision and meniscal debridement. Five of 7 horses had lameness attributable to femorotibial joint pathology, while the remaining 2 horses had meniscal cysts found incidentally during diagnostic arthroscopy for the treatment of osteochondritis dissecans of the lateral trochlear ridge of the femur. Five of 6 horses with long‐term follow‐up were sound and a 7th horse was improved 11 months after surgery. Conclusions and potential relevance: Meniscal cysts, while uncommon, can be associated with progressive lameness in the horse. Surgical excision of the cysts results in resolution or improvement of symptoms, without evidence of recurrence on follow‐up examination.  相似文献

The objective of this study was to evaluate the effects of substituting wheat dried distillers grains with solubles (DDGS) for barley grain and barley silage on intake, digestibility, and ruminal fermentation in feedlot beef cattle. Eight ruminally cannulated Angus heifers (initial BW 455 ± 10.8 kg) were assigned to a replicated 4 × 4 Latin square design with 4 treatments: control, low (25%), medium (30%), and high (35%) wheat DDGS (DM basis). The diets consisted of barley silage, barley concentrate, and wheat DDGS in ratios of 15:85:0 (CON), 10:65:25 (25DDGS), 5:65:30 (30DDGS), and 0:65:35 (35DDGS; DM basis), respectively. The diets were formulated such that wheat DDGS was substituted for both barley grain and barley silage to evaluate whether wheat DDGS can be fed as a source of both energy (grain) and fiber in feedlot finishing diets. Intakes (kg/d) of DM and OM were not different, whereas those of CP, NDF, ADF, and ether extract (EE) were greater (P < 0.01) and intake of starch was less (P < 0.01) for the 25DDGS compared with the CON diet. The digestibilities of CP, NDF, ADF, and EE in the total digestive tract were greater (P < 0.05) for 25DDGS vs. CON. Ruminal pH and total VFA concentrations were not different (P > 0.15) between 25DDGS and CON diets. Replacing barley silage with increasing amounts of wheat DDGS (i.e., from 25DDGS to 35DDGS) linearly reduced (P < 0.05) intakes of DM and other nutrients without altering (P=0.40) CP intake. In contrast, digestibilities of DM and other nutrients in the total digestive tract linearly increased (P < 0.05) with increasing wheat DDGS except for that of EE. Additionally, with increasing amounts of wheat DDGS, mean ruminal pH tended (P=0.10) to linearly decrease, and ruminal pH status decreased with longer (P=0.04) duration of pH <5.5 and <5.2, and greater (P=0.01) curve area under pH <5.8 and <5.5 without altering (P > 0.19) ruminal VFA and NH(3)-N concentrations. Results indicated that wheat DDGS can be effectively used to replace both barley grain and silage at a moderate amount to meet energy and fiber requirements of finishing cattle. However, when silage content of the diet is low (<10%), wheat DDGS is not an effective fiber source, so replacing forage fiber with wheat DDGS in finishing diets decreases overall ruminal pH status even though the rapidly fermentable starch content of the diet is considerably reduced.  相似文献
