Canine oral melanoma is the first malignancy of the oral cavity in dogs and is characterized by a local invasiveness and a high metastatic propensity. A better knowledge of genetic alterations is expected to improve management of this tumour. Copy number alterations are known characteristics of mucosal melanomas both in dogs and humans. The goal of this study was to explore the prognostic value of somatic focal amplifications on chromosomes (Canis Familiaris [CFA]) 10 and 30 in canine oral melanoma. The cohort included 73 dogs with oral melanoma confirmed by histology, removed surgically without adjuvant therapy and with a minimal follow‐up of 6 months. Epidemiological, clinical and histological data were collected and quantitative‐PCR were performed on formalin‐fixed paraffin‐embedded (FFPE) samples to identify specific focal amplifications. The 73 dogs included in the study had a median survival time of 220 days. Focal amplifications on CFA 10 and 30 were recurrent (49.3% and 50.7% of cases, respectively) and CFA 30 amplification was significantly associated with the amelanotic phenotype (P = .046) and high mitotic index (MI; P = .0039). CFA 30 amplification was also linked to poor prognosis (P = .0005). Other negative prognostic factors included gingiva location (P = .003), lymphadenomegaly (P = .026), tumour ulceration at diagnosis (P = .003), MI superior to 6 mitoses over 10 fields (P = .001) and amelanotic tumour (P = .029). In multivariate analyses using Cox proportional hazards regression, CFA 30 amplification (Hazard ratio [HR] = 2.08; P = .011), tumour location (HR = 2.20; P = .005) and histological pigmentation (HR = 1.87; P = .036) were significantly associated with shorter survival time. Focal amplification of CFA 30 is linked to an aggressive subset and constitutes a new prognostic factor.  相似文献

The objective of this study was to compare gelatinization properties and molecular composition of starches extracted from locally grown organic and conventional spelt using thermal, rheological, and SEC analyses, along with Concanavalin A method. Organic and conventional spelt was planted in six replicated plots, and the extracted starch was analyzed for their gelatinization properties. DSC showed that the gelatinization temperature ranged from 56.7 to 68.8 °C with an average peak of 62.4 °C, with no evidence for statistical difference in gelatinization properties between treatments. Rheological behavior variation among samples was more pronounced than that between the two growing conditions. The amylose content ranged from 23.0% to 29.8%. There was no significant difference in the molecular weight of amylose and amylopectin irrespective of the plot locations, although a significant difference was found between the amylopectin molecular weight of organic and conventional spelt starches when analyzed collectively. The organic spelt starch studied may substitute the conventional starch when gelatinization behavior is considered.  相似文献

Power law frequency-size distributions of forest fires have been observed in a range of environments. The scaling behaviour of fires, and more generally of landscape patterns related to recurring disturbance and recovery, have previously been explained in the frameworks of self-organized criticality (SOC) and highly optimized tolerance (HOT). In these frameworks the scaling behaviour of the fires is the global structure that either emerges spontaneously from locally operating processes (SOC) or is the product of a tuning process aimed at optimizing the trade-offs between system yield and tolerance to risks (HOT). Here, we argue that the dominant role of self-organized or optimised fuel patterns in constraining unplanned-fire sizes, implicit in the SOC and HOT frameworks, fails to recognise the strong exogenous controls of fire spread (i.e. by weather, terrain, and suppression) observed in many fire-prone landscapes. Using data from southern Australia we demonstrate that forest fire areas and the magnitudes of corresponding weather events have distributions with closely matching scaling exponents. We conclude that the spatial scale invariance of forest fires may also be a mapping of the meteorological forcing pattern. A computer program was used to map the distribution of Longidorus and Xiphinema nematodes in the British Isles and to analyse relationships between distribution and vegetation or soil factors. Most of the records were obtained by taking soil samples from five vegetation types in each 10 km square of the Universal Transverse Mercator (UTM) maps of the British National Grid and the Irish National Grid. Nematodes were extracted from aliquots of each soil sample by a sieving and decanting technique, then killed by heat and fixed in triethanolamine formalin. A mapping program was written in FORTRAN to produce crude line-printer maps for checking and for sets of standard 80-column cards as input for the Biological Records Centre where the final maps were produced. Analysis of relationships between nematode occurrence and various biotic and abiotic factors was carried out using a standard computer program designed for social survey work and available at the Edinburgh Regional Computing Centre. The maps produced from survey data clearly defined the limits of distribution of Longidorus and Xiphinema species within the British Isles and analysis indicated factors such as soil type or vegetation which may be important in limiting their occurrence. It is suggested that similar survey and computer mapping procedures can be used to establish a European Survey of Plant Parasitic Nematodes. Un programme sur ordinateur a permis d'établir des cartes de distribution des especès de Longidorus et Xiphinema dans les Iles britanniques et danalyser les relations qui existent entre la distribution des nematodes et les facteurs biotiques et édaphiques. Les données proviennent pour la plupart d'échantillons de sol preleves par reference a cinq types de vegetation dans chacun des carres de 10 km formant le réseau cartographique de la Grande-Bretagne et de L'Irlande. Les nematodes sont extraits des echantillons de sol par une methode de tamisage, suivie d'une decantation; ils sont ensuite tues par la chaleur et fixes dans une solution de formaline-triethanolamine. Un programme cartographique a ete etabli en FORTRAN afin d'obtenir des cartes brutes imprimées qui permettent des verifications et le transfert des donnees sur cartes à 80 colonnes qui alimentent le systeme du « Biological Records Centre » dont la tiche consistera a produire les cartes definitives. Lanalyse des relations entre la distribution des nematodes et divers facteurs biotiques et abiotiques a été réalisée par un programme normalisé, en usage pour les enquctes toxicologiques du Centre régional dinformatique d'Edimbourg. Les mêmes methodes pourraient être utilisees pour L'etablissement dune etude de la distribution des nematodes au niveau europeen.  相似文献

Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum (Lib.) De Bary is a serious fungal disease of soybean. Senescing petals provide a starting nutrient source for the invasion of healthy tissue by the advancing oxalic acid secreting fungal hyphae. Since oxalic acid is a major pathogenicity factor of SSR, transgenic soybean capable of degrading oxalic acid may be resistant to the pathogen. Transgenic soybean plants were produced byAgrobacterium -mediated transformation with the wheat germin gene (gf-2.8) encoding an oligomeric protein, oxalate oxidase (OxO), which oxidizes oxalic acid to carbon dioxide and hydrogen peroxide (H2O2). Transgenic soybean homozygous for 35S- gf-2.8 produced an approx. 130 kDa protein indistinguishable from wheat germin, and with OxO activity. OxO activity was prominent in cell walls proximal to the site of pathogen attack. The transgenics had greatly reduced disease progression and lesion length following cotyledon and stem inoculation with S. sclerotiorum indicating that the germin gene product conferred resistance to SSR. This is the first report of plant resistance to the fungal pathogen S. sclerotiorum in transgenic plants expressing OxO.  相似文献

We examined relationships between stable isotopes of carbon (delta(13)C) and oxygen (delta(18)O) in tree rings of Callitris columellaris F. Muell in the semi-arid Pilbara region of north-western Australia. To test the hypothesis that stomatal control of photosynthesis decreases during drier periods, we developed delta(13)C and delta(18)O chronologies spanning 1919-1999, and used a permutation regression approach to relate a 21-year running correlation between delta(13)C and delta(18)O to rainfall and temperature at Marble Bar and our study site. The relationship between delta(13)C and delta(18)O switched from being always negative before 1955 to being consistently positive after 1976, suggesting an increase in stomatal control of photosynthesis in recent decades. Changes in the delta(13)C-delta(18)O relationship reflected changes in rainfall, which has increased in the region by 30% since 1976. The correlation between delta(13)C and delta(18)O was positively related to the 21-year running mean of normalized rainfall anomalies at both the study site (P = 0.045, Adj. r(2) = 0.47) and Marble Bar (P = 0.046, Adj. r(2) = 0.48). In addition, the delta(13)C-delta(18)O correlation was negatively related (P = 0.047, Adj. r(2) = 0.61) to temperatures at Marble Bar. Our interpretation of the role of changes in climate affecting the relationship between tree-ring delta(13)C and delta(18)O is supported by evidence from the isotope composition of foliage samples: foliar delta(13)C and delta(18)O were negatively correlated with log stomatal conductance (delta(13)C, r = -0.41; delta(18)O, r = -0.42), whereas the correlation between foliar delta(13)C and delta(18)O was positive (r = 0.63, P = 0.027) after the summer wet period. Our data indicate that stomatal control of photosynthesis in Callitris adjusts to region-wide changes in climate and that, in a warmer and drier world, trees might adapt by increasing non-stomatal control of photosynthesis.  相似文献

A survey on the occurrence of dermatoses in the Irish Water spaniel (IWS) was carried out in the United Kingdom. A group of 20 dogs was selected and examined clinically. All dogs had a nonpruritic, noninflammatory, regionalized hair loss affecting the same areas of the body in males and females, although an initial cyclical pattern associated with the oestrus cycle was identified. Hormonal investigations showed features suggestive of an abnormality of steroidogenesis. Histopathology revealed features similar to canine recurrent flank alopecia (CRFA) and follicular dysplasia associated with abnormal melanization, as in colour dilution alopecia, although the clinical features did not correlate with those conditions. Dietary changes improved coat and skin quality in most of the cases in this series but the role of the diet was not investigated further. This study suggests that hair loss in IWS is influenced by dietary factors and sexual hormones. Abnormalities of the steroidogenic pathways may contribute to the severity of the condition.  相似文献

This work consisted of two complementary sets of experiments in which breads differing in their recipe and/or process were characterised according to their odorant perception, volatile compound composition and physical properties. The results revealed that commercial partially baked and/or partially baked frozen breads were characterised by a different odour compared to commercial conventional, fully baked frozen and frozen dough breads, which were perceived similarly. These differences were explained by their variable aromatic composition. By analysing breads based on the same recipe but from different processes, it was demonstrated that adding a freezing stage before dough proofing or at the end of the conventional process, as well as after partial baking, did not influence bread aroma. Likewise, partial baking had no effect on bread odour and aromatic profile. Thus, the aromatic differences between commercial conventional, fully baked frozen and frozen dough breads on the one hand, and commercial partially baked breads on the other hand, were due to their different formulations. Concerning bread physical properties, the recipe also influenced bread crust/crumb ratio and density. Moreover, adding a partial baking stage to the process led to breads with a more compact crumb.  相似文献

Salmonella enterica serovar Typhimurium is an animal and zoonotic pathogen of worldwide importance. In pigs, transport and social stress are associated with reactivation and spread of Salmonella Typhimurium infection. The stress-related catecholamine norepinephrine (NE) has been reported to activate growth and virulence factor expression in Salmonella; however the extent to which NE contributes to stress-associated salmonellosis is unclear. We studied the impact of releasing NE from endogenous stores during Salmonella Typhimurium infection of pigs by administration of 6-hydroxydopamine (6-OHDA), which selectively destroys noradrenergic nerve terminals. Treatment of pigs with 6-OHDA 7 or 16 days post-oral inoculation with Salmonella Typhimurium produced elevated plasma NE levels and transiently, but significantly, increased faecal excretion of the challenge strain. Oral administration of NE to Salmonella Typhimurium-infected pigs also transiently and significantly increased shedding; however pre-culture of the bacteria with NE did not alter the outcome of infection. Salmonella has been proposed to sense and respond to NE via a homologue of the adrenergic sensor kinase QseC. A ΔqseC mutant of Salmonella Typhimurium was consistently excreted in lower numbers than the parent strain post-oral inoculation of pigs, though not significantly so. 6-OHDA treatment of pigs infected with the ΔqseC mutant also increased faecal excretion of the mutant strain, albeit to a lesser extent than observed upon 6-OHDA treatment of pigs infected with the parent strain. Our data support the notion that stress-related catecholamines modulate the interaction of enteric bacterial pathogens with their hosts.  相似文献
