With an overarching objective of improving the hatchery production of Atlantic salmon (Salmo salar) all‐female progeny, this study comparatively evaluated the reproductive parameters between normal (genotype XY) and neo‐males (genotype XX). Four normal (XY) and seven neo‐ (XX) males, from the same brood stock, distinguished by their ability to or lack of expressing semen, respectively, were comparatively evaluated. The left testicular lobe was used for histomorphometric analyses, while the right for semen collection and sperm quality analyses. Histomorphometric observations revealed that neo‐male testes are irregularly shaped, and have poorly formed seminiferous ducts, higher proportions of interstitial tissue and lower gonadosomatic index (p < .05). In addition, hypertrophied and cyst forming Sertoli cells were found in these individuals which collectively appear to form a physical barrier, precluding the semen collection by standard stripping techniques and reducing sperm quality. Particularly, semen motility (80.69 ± 2.4% and 57.2 ± 36.5% for XY and XX respectively) and duration of motility (99.31 ± 28.03 s and 66.84 ± 23.83 s for XY and XX respectively) of neo‐males were most compromised (p < .05). Interestingly, the TUNEL assay indicated no signs of apoptotic tissue suggesting that the histological differences may relate to delayed physiological/sexual maturity of neo‐males.  相似文献

We developed a method for in vitro regeneration of Garcinia xanthochymus(yellow mangosteen) from matured seed segments. Multiple shoots were induced on woody plant(WP) medium supplemented with cytokinins.An average of 11 shoots per explant were regenerated from mature seed segments on WP medium containing 20 μM 6-benzylaminopurine. Histological analysis revealed that hypodermal cells of seed segments were initially involved in active division, which later developed into meristemoids, subsequently leading to the formation of shoot buds.Shoot elongation was achieved by repeated subculturing of seed explants in shoot regeneration medium. Rooting of shoots was achieved on WP medium supplemented with indole-3-butyric acid or a-naphthalene acetic acid. Plantlets were transplanted to pots containing soil: compost(1:1) and survival rate was 90%.  相似文献

BACKGROUND: Tetraconazole and difenoconazole are triazole fungicides with proven bioefficacy against grapevine powdery mildew disease. In the present work, the authors explored the residue dynamics of these two compounds in grapes and determined their preharvest intervals (PHIs) corresponding to multiple field applications at recommended and double rates considering the most critical use pattern in Indian viticulture. A confirmatory residue analysis method was validated for trace-level determination of both the compounds. RESULTS: Dissipation of both the fungicides followed non-linear two-compartment first + first-order rate kinetics. Tetraconazole and difenoconazole dissipated with PHIs of 12.5 and 25.5 days at recommended rates and of 28.5 and 38.5 days at double application rates respectively. On all the sampling days, the residues were below the maximum permissible intake, indicating consumer safety. The residues in the grape samples drawn from the farms where these two fungicides were applied, maintaining the above PHIs, were below their respective MRLs. CONCLUSION: The rate of degradation of tetraconazole was faster than that of difenoconazole. Thus, the growers will have the choice of using these new chemicals for the management of powdery mildews in succession, difenoconazole at early growth stages, followed by tetraconazole during the last month before harvest. The recommendations of PHIs proved to be effective in minimizing residues in farm grape samples. Thus, this work is of high significance to the grape industry of India, and will support the registration of these new fungicides for effective management of powdery mildews with minimum residue problems.  相似文献

Six actinomycetes isolates, namely Streptomyces toxytricini vh6, Streptomyces flavotricini vh8, S. toxytricini vh22, Streptomyces avidinii vh32, Streptomyces tricolor vh85 and vh41, an isolate of an unknown species of Actinomycetales, were tested for their efficacy in protecting tomato (Solanum lycopersicum) against Rhizoctonia solani under green house conditions. Actinomycetes treated plants showed better growth in terms of high chlorophyll content, higher phenylalanine ammonia lyase (PAL) activity and high total phenolic content. Qualitative and quantitative estimation of phenolic compounds from tomato leaves showed significant accumulation of six phenolic acids, gallic (29.02 μg g−1 fresh leaf wt), ferulic (11.44 μg g−1 fresh wt), cinnamic (56.84 μg g−1 fresh wt), gentisic (24.19 μg g−1 fresh wt), chlorogenic acid (1.72 μg g−1 fresh wt) and salicylic (0.39 μg g−1 fresh wt) acid, in actinomycetes treated plants. Biochemical profiling, when correlated with plant mortality in actinomycetes treated and untreated plants, indicated that isolates vh6 and vh8 offered 44.55% and 40.14% disease reductions, respectively compared to the control. These results established that these organisms have the potential to act as biocontrol agents.  相似文献

Genotyping of Mycobacterium avium subspecies paratuberculosis (MAP) is important for precise classification of bacterium and for understanding the molecular epidemiology. The present study reports detection and typing of the MAP from milk. On the basis of clinical signs of diarrhea and/or weakness, the dairy animals suspected for Johne’s disease were screened by Ziehl–Neelsen staining of fecal samples. The milk samples from 13 selected animals were processed for DNA extraction and direct IS900 polymerase chain reaction (PCR). MAP identified by IS900 PCR was genotyped using IS1311 PCR-restriction endonuclease analysis (REA). IS900 milk PCR revealed 30.8% animals positive for MAP, including 40% of the moderate and 50% of the heavy fecal shedders. All infected animals showed Bison type MAP in IS1311 PCR-REA. IS900 PCR can be used for screening of milk for MAP; however, the method needs to be evaluated for subclinical cases. IS1311 PCR-REA results indicated the predominance of Bison type MAP in the dairy animals of this region.  相似文献

The cDNAs of three cytokines, viz., IL-2, IL-4 and IFN-γ from Dromedary camels were amplified by PCR using Bactrian camel sequences and subsequently cloned for sequence analysis. Relationship based on amino acid sequences revealed that Dromedary camel IL-2 shared 99.5% and 99.3% identity at the nucleotide and amino acid levels with Bactrian camel IL-2. In the case of IL-4, the identity of Dromedary camel was 99.7% and 99.2% at the nucleotide and amino acid levels, respectively with that of Bactrian camel. The Dromedary camel IFN-γ shared 100% identity both at nucleotide and amino acid levels with Bactrian camel IFN-γ. Phylogenetic analysis based on amino acid sequences indicated the close relationship in these cytokine genes between the Dromedary camel and other camelids.  相似文献

Wild species of the genus Oryza are a good source of beneficial alleles for enhancing rice yield under normal and adverse conditions. BC2F3 population was derived from a cross between Oryza sativa IR58025B and Oryza meridionalis Ng. (2n = 24, AA) a heat tolerant wild species to evaluate 12 yield traits under irrigated and aerobic conditions. Analysis of variance and genetic estimates indicated there is substantial genetic variation among progenies under both conditions. Grain yield had high heritability (61.9%) and genetic advance (36.4%) under irrigated conditions but moderate heritability (49.6%) and genetic advance (13.3%) under aerobic conditions indicating that selection for yield will be effective under both conditions. Panicle number, grain number, spikelet fertility, and test weight showed significant positive correlation with grain yield under both conditions. Families out-performing IR58025B for yield under both conditions were obtained providing evidence that phenotypically inferior O. meridionalis contributed to yield increase. This species can be a novel source of natural genetic variation for the improvement of rice under irrigated as well as under aerobic condition.  相似文献

In the present study, the effect of irradiation, storage, and freeze drying on grapefruit bioactive compounds was investigated. Grapefruits were exposed to one of two irradiation doses: 0 (control) or 300 Gy (137Cs, a proposed treatment against fruit flies) and then stored for up to 6 days. At the last storage time point (6 days after harvest), grapefruit pulp from control and irradiated fruits was freeze-dried. Bioactive compounds were extracted from Rio Red grapefruit pulp and analyzed with reverse phase liquid chromatography while volatile compounds were analyzed using gas chromatography. Freeze-dried pulp from irradiated fruits had a higher (P < or = 0.05) flavonoid content (naringin and narirutin) as compared to the freeze-dried pulp from the control fruits. Freeze-drying treatment reduced (P < or = 0.05) the lycopene content, but the reduction (P < or = 0.05) in beta-carotene content occurred only in the control fruit. Reduction in d-limonene and myrcene was observed in the irradiated fruits at 6 days after harvest and in the freeze-dried samples. These results warrant testing of the effect of postharvest treatments and processing on bioactive compounds in functional systems as they have varied effects on different bioactive compounds of grapefruit.  相似文献
