Maintenance of genomic methylation patterns is mediated primarily by DNA methyltransferase-1 (DNMT1). We have solved structures of mouse and human DNMT1 composed of CXXC, tandem bromo-adjacent homology (BAH1/2), and methyltransferase domains bound to DNA-containing unmethylated CpG sites. The CXXC specifically binds to unmethylated CpG dinucleotide and positions the CXXC-BAH1 linker between the DNA and the active site of DNMT1, preventing de novo methylation. In addition, a loop projecting from BAH2 interacts with the target recognition domain (TRD) of the methyltransferase, stabilizing the TRD in a retracted position and preventing it from inserting into the DNA major groove. Our studies identify an autoinhibitory mechanism, in which unmethylated CpG dinucleotides are occluded from the active site to ensure that only hemimethylated CpG dinucleotides undergo methylation.  相似文献

Two greenhouse experiments were conducted to evaluate the performance of selected soybean- Bradyrhizobium combinations in a soil having low P-high aluminum stresses under controlled conditions. Growth parameters such as plant height, nodule numbers, dry weight, nitrogenase activity and seed yield were used as an index. Three selected acid (low P-high Al) tolerant ( Essex, Tracy and Bedford ) and three sensitive ( Peking, Hill and McNair 600 ) soybean ( Glycine max ) varieties were sown in each pot. Three of the most tolerant strains of Bradyrhizobium (USDA 6, 110, 122), a mixture of these strains, and a commercial inoculant (Nitragin) were used to inoculate the Hartselle sandy loam soil. The results indicated that soil acidity significantly affected the soybean Bradyrhizobium symbiosis. The combination of variety Tracy plus Nitragin (mixed-strain) produced the highest and variety Peking plus mix-strain Nitragin the lowest plant height at all pH levels. Plants inoculated with Nitragin had higher shoot dry weight and seed yields. Variety Essex in combination with Nitragin produced the highest seed yields at all pH levels. Plants inoculated with Nitragin developed significantly more nodules than the other strains except for variety Peking. There was no significant difference in total nodule numbers between tolerant and sensitive soybean varieties at all soil pH levels except for Nitragin. Shoot total N did not increase by increasing the pH level or by using different variety-strain combinations.  相似文献

Equine adenovirus 1 was recovered after four to six passages from two out of three cases of cauda equina neuritis (CEN) using kidney monolayers. Similar treatment of lumbo-sacral spinal cord from six normal horses did not yield adenovirus. All three cases of CEN had antibodies to the neuritogenic myelin protein P2 while immunofluorescence demonstrated that autologous IgG bound to the myelin of affected nerves. Adenovirus was not detected in neural tissue by immunofluorescence.  相似文献

The conformation and dynamics of the d(CGCGAATTCGCG) duplex, its analogs containing mismatched base pairs and helix interruptions, and its complexes with actinomycin and Netropsin, bound separately and simultaneously, have been investigated by nuclear magnetic resonance spectroscopy in aqueous solution. Structural information has been deduced from chemical shift and nuclear Overhauser effect parameters, while the kinetics have been probed from line width and saturation recovery experiments on proton and phosphorus markers at the individual base pair level. These studies lead to an improved understanding of the role of nucleic acid sequence on the structure, flexibility, and conformational interconversions in the duplex state. The nuclear magnetic resonance measurements readily identify helix modification and antibiotic binding sites on the nucleic acid and estimate the extent to which the observed conformational and dynamic perturbations are transmitted to adjacent base pair regions.  相似文献

Detection sensitivities of a few parts per billion for ten gaseous pollutants have been evaluated by measuring the strength of the absorption of infrared radiation from carbon monoxide and carbon dioxide lasers. Ethylene concentrations as small as 5 parts per billion have been detected in air. The measured absorption strengths indicate that in mixtures of pollutants, such as nitrogen dioxide and water vapor, the sensitivity is reduced by overlapping absorption bands. However, calculations indicate that it should be possible to detect nitrogen dioxide concentrations of 0.01 part per million in the presence of water vapor concentrations of 105 parts per million.  相似文献

A study was conducted to investigate the effect of dough formulation and hydration level on the rheological properties of pasta dough during pasta extrusion. Semolina 100%, whole wheat 100%, and the following mixtures semolina-whole wheat (49:51), semolina-flaxseed flour (90:10), whole wheat-flaxseed flour (90:10), and semolina-whole wheat-flaxseed flour (39:51:10) were the formulations used for the experiments. Dough was hydrated at 30, 32, and 34% moisture content. Pasta was extruded with a capillary and a semi-commercial pasta extruder to determine the apparent viscosity of the dough during extrusion conditions and its relationship to the behavior of the dough during pasta processing. Results showed that non-traditional pasta dough behaved like a shear thinning fluid that can be described by the Power Law model. Increased hydration levels and/or presence of flaxseed flour on the dough formulation decreased the apparent viscosity of the dough, which correlated with extrusion pressure, mechanical energy, and specific mechanical energy that were required to extrude the dough in the pasta extruder. The strong correlations found between the apparent viscosity of the dough and the pasta extrusion parameters indicates the possibility of using a capillary rheometer to determine the appropriate hydration level of ingredient formulations before extruding with a pasta press.  相似文献

Traditionally, chapattis are flatbreads made from atta (wholemeal Indian wheat flour). Non-atta chapattis have not been popular due to substandard product quality. To investigate what makes atta special for making chapattis, products were made using atta, Australian wholemeal wheat flour, gluten-free lupin flour, and a blend of lupin and wheat flours. Doughs were characterised by measuring strain-hardening and elastic recovery in compression and also bubble structures via 3-D X-ray micro-tomography. A method was developed to identify and separate bran, which appears as bubbles, in scans of doughs.  相似文献

A series of monoazo disperse dyes based on 2-amino-4-phenylthiazole was prepared using variousN,N-dialkylaniline derivatives as the coupling component. The dyes were characterized by IR spectral studies, visible absorption spectroscopy and elemental analysis. The dyeing performance of these dyes was assessed on cellulose triacetate and nylon fibers. These dyes were found to give a wide range of colour shades varying from bright red to royal blue with very good depth, brightness and levelness on fibers. The dyed fibers showed good to very good light fastness and very good to excellent fastness to washing, perspiration, rubbing and sublimation. The dyebath exhaustion and fixation on the fibers were found to be very good.  相似文献

Homo and copolymers of monomers 2-(N-phthalimido) ethylmethacrylate (NPEMA) and 4-Chloro-3-methyl phenyl methacrylate (CMPMA) were prepared in N,N-dimethyl formamide (DMF) solution at 70 °C using 2,2-azobisisobutyronitrile (AIBN) as initiator. The solution of poly(NPEMA-co.-CMPMA) in 20 % DMF was used to fabrication electrospun nanofiber by electrospinning technique. IR data were primarily employed to characterize polymers. The formation of nanofibers was identified by SEM study. The metal ion uptake capacity of copolymers and nanofibers were obtain by batch equilibrium method using different metal ion solution. The antimicrobial activity of the copolymers, Polymer nanocomposites and their nanofibers were tested against different microbial organisms by using quantitative method. The main objective of this investigation was to find whether nanofiber are better remover of metal ions compared to copolymers. It was also aimed to study the efficacy of nanofibers of copolymers and copolymer composite with nano Ag as water sanitizer.  相似文献
