Clostridium difficile is an emerging enteric pathogen of humans and animals with a known main reservoir in the intestinal tract of various warm‐blooded animals. This study was carried out to evaluate the prevalence of C. difficile in 150 rectal swab samples collected from cattle and goats in Switzerland. The overall prevalence of C. difficile was 6.6%. The isolates belonged to the PCR ribotypes 033, 066, 070, 003, 001 and 137. In addition, the occurrence of C. difficile in faecal samples collected from farm ground was also evaluated and C. difficile was detected in 21% (7/30) of these samples. These isolates belonged to ribotypes 033, 066, 014 and 137. Several isolates pertained to ribotypes known to cause Clostridium difficile infections in humans.  相似文献

In order to apply state-of-the-art molecular breeding techniques in fibre crop it is necessary to have a good knowledge of major polymer biosynthesis gene sequences and their expression pattern. Polymerase chain reaction was employed to isolate sequences of the major genes for lignin and cellulose biosynthesis in a kenaf cultivar. CeSA, 4cl, c4h, cad, and ccr gene primers were designed according to their conservative regions; partial sequences of lignin and cellulose biosynthesis genes were obtained. One actin II gene sequence was also isolated from the kenaf genome as a housekeeping gene to be employed in qPCR analysis. Expression levels of genes c4h, cad and CeSA in bark and core from plants harvested at three different growth stages were evaluated. Using qPCR analyses it was found that the expression levels of the two biosynthesis lignin genes in bark tissues increased during plant growth, while a negative trend was recorded in core tissues. In both bark and core, the quantity of lignin was positively correlated to plant growth while cellulose content decreased.  相似文献

The specific activity of a whole cell acid urease preparation was tested in five wines manufactured in the Apulia region of Italy in the 2003 vintage at both short and long treatment times, thus confirming the validity of the pseudo-first-order kinetic model to describe urea removal in real wines. The ratio between the experimental pseudo-first-order kinetic rate constant (kIe) for any real wine tested and that (kI) referred to a model wine solution having the same composition and pH reduced from about 0.21 to 0.02 as the overall content of phenolic compounds (P) increased from 109 to 853 g m-3 of gallic acid equivalent (GAE). The specific inhibitory effect of such compounds was explained by accounting for the equilibrium constant (KP) of the reaction of polyphenols with acid urease, which was found to be about 21 g of GAE m-3 for the real wines tested, whereas it ranged from about 16 to 45 g of GAE m-3 when the model wine solution was enriched with tannins extracted from grape seeds or skins, respectively. A sequential experimental procedure consisting of accelerated acid urease tests at high doses of enzyme followed by accelerated ethyl carbamate tests on the resulting acid urease treated wine was recommended to assess preliminarily the technoeconomic feasibility of the acid urease hydrolytic process for the wine of concern. Trichinella sp. infection has been documented in both humans and animals in most Mediterranean countries, yet in the past 60 years no infections have been reported on the Mediterranean islands. We describe the first outbreak of Trichinella sp. infection to have been reported on the island of Sardinia. The outbreak occurred in two villages in 2005 and involved 11 persons who had eaten raw sausages made from the same free-ranging sow. All 11 persons developed signs and symptoms of trichinellosis and seroconverted within 48 days of consuming the infected meat. The etiological agent was Trichinella britovi. Sardinia, like all Mediterranean islands, had been considered to be Trichinella-free. The present report, together with a recent report of T. britovi infection in animals on the nearby island of Corsica (France), raises questions as to the validity of the concept of Trichinella-free areas or regions.  相似文献

A standardized extract of red orange juice (ROE) was shown to inhibit proliferation of fibroblast and epithelial prostate cells. Twenty-four dogs naturally infected by Leishmania spp. were treated with four different protocols using meglumine antimoniate (aNm) and allopurinol in combination or in monotherapy. Aiming to compare the efficacy of the different treatments the reverse to normal of clinico-pathological findings and the disease free interval time (DFIT) were evaluated. Treated dogs were monitored for 1 year and, in absence of relapses, the DFIT was postponed to the last available follow-up. Seven dogs treated with aNm alone showed relapses during the year of observation. In the group of dogs treated with the combination of aNm (50 mg/kg/SC 12 hourly up to clinico-pathological recovery) and allopurinol (15 mg/kg/PO 12 hourly administered for 6months) no relapses were registered in the year of monitoring and the DFIT reached up to 65 months. Our results showed that this combination represents the best choice to treat canine leishmaniosis compared to other protocols.  相似文献

Nitrate leaching is one of the many forms of environmental pollution resulting from irrigation and intensive agriculture. In this work, a method of combining an agronomic simulation model (EPIC) and a mathematical multi-objective programming model is used to analyse the effects of three agricultural policies on farmer’s revenue and nitrate leaching. An evaluation of the net social costs associated with the different policy measures is also given. The farmer’s behaviour in different policy scenarios was studied in terms of selected crops, irrigation technique and method, and adopted management practices with focus on farm management practices and water application efficiency. Irrigation water pricing, subsidies to adopt improved management levels, and taxation on the use of nitrogen fertilizer were examined. A trade-off emerges between the levels of nitrate leaching and net farmer’s revenue more pronounced for nitrogen tax policies than for water pricing. The results obtained indicate that nitrate leaching can be reduced by about 40% with an associated net social cost of 269 €/ha for the water pricing policy, 183 €/ha for the tax on fertilizer and 95 €/ha for subsidies to high efficiency management.  相似文献

Radiation use efficiency () is a crop parameter widely used in crop simulation models, derived here as the slope of the relationship linking crop carbon gain to cumulative intercepted solar radiation. Our objectives were to: (i) determine of field-grown sunflower, sorghum, wheat and chickpea, at three levels of aggregation—day-time net assimilation (A), daily net carbon gain as difference between A and night-time dark respiration (R) and biomass; (ii) assess the robustness of parameter in terms of ability to discriminate between C3 and C4 species, pre- and post-anthesis and impact of nitrogen status; (iii) evaluate the opportunity to normalize for climate. Field experiments were conducted in 1998 and 1999 in southern Italy. All crops were well watered. Sunflower and sorghum had two nitrogen application treatments, wheat only one and chickpea had no added nitrogen. Daily intercepted solar radiation was derived from measurements of daily incident solar radiation and the percentage of mid-day light interception. Closed-system canopy chambers monitored canopy gas-exchange rates. , expressed in terms of A, AR or above-ground biomass, was linear over the entire growth cycle of sorghum and wheat, and up to anthesis in sunflower and chickpea, independent of temperature, vapour pressure deficit and radiation regimes. In sunflower, deviation from linearity was observed after anthesis, due to higher carbon cost in yielding oil seeds. No conclusions could be drawn for post-anthesis chickpea due to the interruption of the experiment caused by a thunderstorm. Overall results showed a great variability in values, independently of classes of species (C4 and C3), crops and nitrogen treatments. This indicates that the robustness of to predict biomass productivity in crop simulation models is constrained. Attempts to normalize by vapour pressure deficit (D), for reducing its variability due to climate and overlaps between crops failed.  相似文献
