A putative link between Salmonella persistence in the agricultural sector and resistance to disinfectants has been sparsely investigated. Therefore, minimum inhibitory concentration (MIC) tests against five disinfectants commonly used in poultry premises (formaldehyde, glutaraldehyde/benzalkonium chloride compound, oxidising compound, tar oil phenol, iodophor) were performed on 286 Salmonella isolates, including 256 from Danish broiler houses, altogether representing nine serotypes. Six of these isolates were used for adaptation and de-adaptation studies involving the five disinfectants. Amongst 60 of these isolates selected for growth studies in cyclohexane (possibly associated with up-regulated efflux), only one isolate grew. From this isolate and the six isolates used in the adaptation and de-adaptation studies, mutants highly resistant to triclosan (a disinfectant linked with mar-type resistance) were selected. In addition, adaptation and de-adaptation studies with triclosan were performed. For the 286 isolates, the small variations in MICs could not be associated with Salmonella persistence in Danish broiler houses or previous use of relevant disinfectants. Adaptation and de-adaptation did not alter MICs to the five farm disinfectants. Compared to the parent isolates, MICs for the triclosan adapted and de-adapted isolates and the triclosan mutants were significantly increased to triclosan, but not to the five disinfectants. Moreover, most of the triclosan adapted and de-adapted isolates grew in cyclohexane. Thus, there was no correlation between triclosan and cyclohexane resistance on one hand and resistance to the five disinfectants on the other, suggesting that triclosan resistance is not linked with resistance to these disinfectants. 相似文献

The residence time of fine-root carbon in soil is one of the least understood aspects of the global carbon cycle, and fine-root dynamics are one of the least understood aspects of plant function. Most recent studies of these belowground dynamics have used one of two methodological strategies. In one approach, based on analysis of carbon isotopes, the persistence of carbon is inferred; in the other, based on direct observations of roots with cameras, the longevity of individual roots is measured. We show that the contribution of fine roots to the global carbon cycle has been overstated because observations of root lifetimes systematically overestimate the turnover of fine-root biomass. On the other hand, isotopic techniques systematically underestimate the turnover of individual roots. Time-resolved diffuse optical spectroscopy (TRS) was investigated as a nondestructive method to characterize the post-impregnation distribution of methacrylate monomers within spruce (Picea abies). TRS was also used to monitor the flow of methacrylate monomers in situ, within spruce, during impregnation with both spatial and temporal resolution. The data were compared to fluid flow models developed by Darcy and Bramhall demonstrating that neither of these models were able to accurately describe the experimental results, highlighting the need for development of new models. Nondestructive characterization by TRS did not require staining of the monomer treatment solution, multivariate analysis or complex sample pre-treatment, thus highlighting the facile applicability of this technique. 相似文献

Stomatal conductance was quantified with sap flux sensors and whole-tree chambers in mature Norway spruce (Picea abies (L.) Karst.) trees after 3 years of exposure to elevated CO(2) concentration ([CO(2)]) in a 13-year nutrient optimization experiment. The long-term nutrient optimization treatment increased tree height by 3.7 m (67%) and basal diameter by 8 cm (68%); the short-term elevated [CO(2)] exposure had no effect on tree size or allometry. Nighttime transpiration was estimated as approximately 7% of daily transpiration in unchambered trees; accounting for the effect of nighttime flux on the processing of sap flux signals increased estimated daily water uptake by approximately 30%. Crown averaged stomatal conductance (g(s)) was described by a Jarvis-type model. The addition of a stomatal response time constant (tau) and total capacitance of stored water (C(tot)) improved the fit of the model. Model estimates for C(tot) scaled with sapwood volume of the bole in fertilized trees. Hydraulic support-defined as a lumped variable of leaf-specific hydraulic conductivity and water potential gradient (K(l)DeltaPsi) -was estimated from height, sapwood-to-leaf area ratio (A(s):A(l)) and changes in tracheid dimensions. Hydraulic support explained 55% of the variation in g(s) at reference conditions for trees across nutrient and [CO(2)] treatments. Removal of approximately 50% of A(l) from three trees yielded results suggesting that stomatal compensation (i.e., an increase in g(s)) after pruning scales inversely with K(l)DeltaPsi, indicating that the higher the potential hydraulic support after pruning, the less complete the stomatal compensation for the increase in A(s):A(l). 相似文献

The spherical mean of the shoot silhouette-to-total leaf area ratio (STAR) and the shoot transmission coefficient (c) are two key structural parameters in radiative transfer models for calculating canopy photosynthesis and leaf area index. The standard optical method for estimating these parameters might introduce errors in the estimates for species with flexible shoots and needles by changing shoot inclination relative to its inclination in situ. We devised and tested two methods to address this problem. First, we modified the standard optical method by designing an apparatus that allows shoots to be photographed in their original orientation. Second, we developed a faster, model-based approach to replace photography and tested the results against the established approach. We used shoots of three pine species, Pinus echinata Mill. (needle length ~50 mm), P. taeda L. (~150 mm) and P. palustris Mill. (~300 mm). Values of the parameters simulated by the model were similar to those measured from the photographs. In our data, STAR varied about twofold among the pine species and was ~40% higher in shade shoots than in sun shoots of P. taeda. The transmission coefficient for P. taeda shade shoots was also ~40% higher than that of sun shoots of all three species. We tested the versatility of the model by employing it on shoots of two other pine species (P. strobus L. and P. thumbergiana Parl.) as well as on shoots of Tsuga canadensis L. Carr. and Picea pungens Engelm. Regardless of shoot characteristics, the model generated values of shoot structural parameters similar to those estimated with the optical method. Although species-specific and vertical gradients in parameter values are best for modeling radiative transfer in conifer canopies, our results suggest that, in the absence of adequate data, STAR can be approximated as 0.16 for a wide range of shoot structures. For applications requiring angle-dependent parameterization, our new model facilitates rapid generation of these radiative transfer parameters. 相似文献

Purpose
Assess the refractive states of donkeys and goats.Methods
Forty-two donkeys and 28 goats were enrolled. The mean ± SD ages were 7.68 ± 7.33 years for donkeys and 4.26 ± 2.33 years for goats. Seven donkeys and one goat were <6 months old. Retinoscopy was performed in alert animals, following cycloplegia in goats but not in donkeys. Normality was determined using the Kolmogorov–Smirnov test. The two primary meridians and two eyes were compared using Pearson's correlation and paired Student's t–tests. The association between refractive states and age was examined using one-way ANOVA in donkeys and a paired Student's t-test in goats. One-sample t-tests were conducted to assess if the refractive error distributions were significantly different from “0”.Results
The mean ± SD spherical equivalent (SE) refractive errors of the right and left donkey eyes were −0.80 ± 1.03 D and −0.35 ± 0.95 D, respectively. The majority (86%) of the donkeys had an astigmatic refraction and eight (19%) had anisometropia. The mean SE refractive errors of the right and left goat eyes were −0.15 ± 1.1 D and −0.18 ± 1.2 D, respectively. The majority (54%) of the goat eyes had an astigmatic refraction and five (18%) had anisometropia. The right and left eye SE refractive errors were positively correlated in both species (both p = .9). Age was not correlated with refractive error in both donkeys (p = .09) and goats (p = .6).Conclusions

Copper fungicides applied via low head sprinklers, reduced brown rot (caused byPhytophthora spp.) on oranges and grapefruit. The most effective fungicide was Bordeaux mixture, which reduced the disease rate to the same level as that obtained by standard spraying with this fungicide.
