When wheat was grown under conditions of severe sulfate depletion, dramatic increases in the concentration of free asparagine were found in the grain of up to 30 times as compared to samples receiving the normal levels of sulfate fertilizer. The effect was observed both in plants grown in pots, where the levels of nutrients were carefully controlled, and in plants grown in field trials on soil with poor levels of natural nutrients where sulfate fertilizer was applied at levels from 0 to 40 kg sulfur/Ha. Many of the other free amino acids were present at higher levels in the sulfate-deprived wheat, but the levels of free glutamine showed increases similar to those observed for asparagine. In baked cereal products, asparagine is the precursor of the suspect carcinogen acrylamide, and when flours from the sulfate-deprived wheat were heated at 160 degrees C for 20 min, levels of acrylamide between 2600 and 5200 microg/kg were found as compared to 600-900 microg/kg in wheat grown with normal levels of sulfate fertilization. 相似文献

Poor seed germination is a significant problem for propagation of teak and it particularly hampers the deployment of genetically improved material into plantations. Seed dormancy is the putative cause for delayed and sporadic germination of teak seed but specific dormancy mechanisms have not been proven. In this paper we investigate whether physical or mechanical dormancy could affect teak germination. Physical dormancy was disproved when we observed water in every examined locule of fruit which had been immersed for 12–24 h (n = 1,700 fruits from six diverse sources). Mechanical dormancy was found to affect teak seed germination by means of valve structures which must open for the radicle to emerge. All 1,450 germinating seeds in 16 samples emerged from the endocarp via the detachment of a valve. Germination over 16 days was compared between extracted seeds (ex situ) and seeds within their endocarp (in situ) to determine the effect of mechanical dormancy on germination. Significantly greater germination of the ex situ seeds (62 ± 2% SE; n = 486) than of the in situ seeds (32 ± 2% SE; n = 564) indicated that mechanical dormancy is an important factor in the poor germination of teak. 相似文献

Growing research interest in crop wild relatives (CWR) has highlighted their value for crop improvement, particularly to mitigate the impact of climate change and contribute to global food security. As most conservation activities are implemented at national level there is a requirement for each country to develop and implement a national CWR conservation strategy. This will contribute to regional and global CWR conservation actions and so help achieve CBD Aichi Targets 13 of improved genetic conservation of socio-economically important taxa. Cyprus has historically lacked systematic CWR conservation but is rich in CWR diversity being located in the eastern Mediterranean Vavilov centre and abutting the Fertile Crescent. Therefore a more coherent approach to CWR conservation is desirable. From an initial checklist of 1,722 Cypriot CWR taxa, 178 CWR were prioritized and eco geographic data was collated and analysed using GIS techniques. The Troodos and Pafos mountains are CWR hotspots and ten priority sites containing the genetic diversity of 74.7 % of priority CWR taxa were identified. These will form the basis of a national network of in situ genetic reserves and 78 priority CWR taxa that have no accessions in the national Cyprus gene bank are highlighted for collection and ex situ storage. 相似文献

Wheat flour from plants deficient in sulfur has been shown to contain substantially higher levels of free amino acids, particularly asparagine and glutamine, than flour from wheat grown where sulfur nutrition was sufficient. Elevated levels of asparagine resulted in acrylamide levels up to 6 times higher in sulfur-deprived wheat flour, compared with sulfur-sufficient wheat flour, for three varieties of winter wheat. The volatile compounds from flour, heated at 180 degrees C for 20 min, have been compared for these three varieties of wheat grown with and without sulfur fertilizer. Approximately 50 compounds were quantified in the headspace extracts of the heated flour; over 30 compounds were affected by sulfur fertilization, and 15 compounds were affected by variety. Unsaturated aldehydes formed from aldol condensations, Strecker aldehydes, alkylpyrazines, and low molecular weight alkylfurans were found at higher concentrations in the sulfur-deficient flour, whereas low molecular weight pyrroles and thiophenes and sugar breakdown products were found at higher concentrations in the sulfur-sufficient flour. Hardsetting soil properties are undesirable in agricultural soils because they hamper crop production by limiting seedling emergence and root growth via increased mechanical soil resistance at low moisture contents. The objective of this study was to determine the effect of additions of organic matter on the penetration resistance of a hardsetting soil for the entire water tension range. Investigations were carried out on Saalian glacial till, which is used as a reclamation substrate in post-lignite-mining reclamation. Proportions of 0%, 1%, 2%, 3% and 4% by mass of organic matter (OM) were used. The remoulded samples were saturated under a constant load of 2.4 kPa to achieve bulk densities equivalent to a soil depth of 15–20 cm via water-induced consolidation. Subsequently, the mixtures were adjusted to water tensions between 100 and 107 hPa and penetrated using a small cone penetrometer. Compared to 0% OM, the addition of 1% OM led to a very small but significant (P < 0.01) increase in the bulk density, while between 1% and 4% OM bulk density was seen to decrease in a linear fashion. At moisture contents greater than field capacity, penetration resistance values were consistent with the observed changes in bulk density, leading to an increase in the samples containing 0–1% OM to critical values for root-growth and a decrease for samples containing 2% and more organic matter reaching to values non-critical for roots. At moisture contents smaller than field capacity, penetration resistance values were inversely related to the bulk density, supporting the concept that the type of organic matter added contributed to soil cohesion. A cross-sectional study was designed to investigate the prevalence and risk factors for Cryptosporidium infection in young calves. Forty-one farms in a discrete, densely farmed 100 km2 area of North West England were visited over a 3-week period and 215 faecal samples were collected from young calves. Farms were not selected on the basis of existing scour problems. At the time of sampling, several investigator-observed variables were recorded at the pen, animal and stool levels. Samples were screened and 60/215 were confirmed as positive by PCR of the 18S rRNA gene. Risk factors for infection were explored using multilevel multivariable logistic regression with farm as a random effect. Age was significant in the final model, with a higher risk of infection in calves aged 8-21 days, when compared to those aged 0-7 days. The depth of the bedding was also significant in the final model, with calves housed in bedding 11-15 cm deep being at lower risk of infection than those on beds 0-5 cm deep. Consistency of the faeces was highly correlated with age and colour of the faeces and was not significantly associated with infection when these variables, and clustering at farm-level, were accounted for. This is interesting as Cryptosporidium is considered to be a primary enteropathogen. The results suggest that intervention strategies should be targeted at calves under 21 days old. These animals represent a significant reservoir of infection on the farm and may also pose a risk to public health, assuming that the species and genotypes shed are zoonotic pathogens. 相似文献

A novel proximity ligation assay (PLA) using a pan-serotype reactive monoclonal antibody was developed and evaluated for the detection of foot-and-mouth disease virus (FMDV) in clinical samples collected from field cases of disease. The FMDV-specific PLA was found to be 100 times more sensitive for virus detection than the commonly used antigen capture-ELISA (AgELISA). As few as five TCID50 were detected in individual assays, which was comparable with the analytical sensitivity of real-time RT-PCR. Although this assay was capable of detecting diverse isolates from all seven FMDV serotypes, the diagnostic sensitivity of the PLA assay was lower than real-time RT-PCR mainly due to a failure to detect some SAT 1, SAT 2 and SAT 3 FMDV strains. In conclusion, this new PLA format has high analytical sensitivity for the detection of FMDV in clinical samples and may prove valuable as a rapid and simple tool for use in FMD diagnosis. 相似文献
