Lingual antimicrobial peptide (LAP), one of the β-defensins in bovines, and lactoferrin (LF) are synthesized in mammary epithelium and have bactericidal and bacteriostatic functions. However, it is not known whether they have similar expression patterns. Therefore, the present study was undertaken to compare (1) immunolocalization of LAP and LF in the mammary gland and (2) changes in concentration of these two components in milk after lipopolysaccharide (LPS) challenge. Bovine mammary tissues without LPS challenge were collected and their sections were immunostained with antibodies to LAP or LF. Milk from our previous study was collected every hour up to 12h and twice daily from d 1 to 7 after LPS challenge (the day of infusion was considered as d 0). These milk samples were measured for LAP but not LF in our previous report. Therefore, concentration of LF was measured by enzyme immunoassay in the present study. Epithelial cells of some alveoli showed immunopositive reaction for LF, but negative for LAP. Conversely, some alveoli were LAP positive in their epithelial cells but LF negative. Many alveoli had immunoreactions for neither LAP nor LF. The concentration of LAP in milk was elevated significantly at 3h after LPS infusion compared with pre-infusion values and remained at a high level until 12h. However, LF concentration in milk remained low at d 0 and increased at d 2. These results suggest that LAP and LF were mostly differentially localized in the alveolar epithelium in mammary glands. The different spatial expressions between them may be associated with their different temporal expression mechanisms.  相似文献

Spatial genetic structure and diversity of Cryptomeria japonica trees in old growth forest but selectively logged in approximately 300 years ago were analyzed using seven nuclear microsatellite markers and one chloroplast microsatellite marker. The individuals were sampled from a 4-ha natural forest plot (200 m × 200 m), which are consisted of three size classes including newly regenerated individuals such as regenerated individuals (REG class; stem diameter at breast height (DBH), 5–110 cm), remaining individuals (REM class; DBH ≥ 110 cm), and old stumps. Genetic diversity was similar between the three groups. These results suggest that genes from the old growth forest may have been passed on to the next generation (i.e., the current forest). DNA extraction and analysis from old stumps was partly successful and we could compare the genetic diversity between old stumps and living trees using four microsatellite loci. Patterns of spatial genetic structure detected by analyses of the nuclear and chloroplast markers differed substantially; all nuclear markers showed significant positive autocorrelation (P < 0.05) over short distances, but the chloroplast marker showed no significant autocorrelation over any distance. We concluded that Moran's I spatial autocorrelation pattern for the chloroplast microsatellite may have been influenced by the extensive pollen flow characteristic of this species, because chloroplast DNA of C. japonica is paternally inherited through pollen and is a wind-pollinated species that produces abundant pollen every few years. In contrast, Moran's I spatial autocorrelation pattern for nuclear microsatellites may reflect limited seed dispersal in gaps where the forest has regenerated and low levels of seed shadow overlap due to past logging. The average seed dispersal distance was 86.0 m at the study site and about 34.0% of the seeds were from the outside of the site, which revealed by parentage analysis. The individuals remaining after logging, and the extensive pollen flow from outside our study plot, may play important roles in maintaining the forests on this island. Gaps created by the selective logging activities appear to have been important for the regeneration of this species in the past but in the present time, natural disturbances such as typhoon and landslide are also important for gap creation.  相似文献

A risk assessment of ozone (O3) impact on the annual carbon absorption (ACA) of Japanese representative conifers was conducted based on the results of an experimental study, monitoring data of oxidant concentrations and vegetation surveys. The areas with high O3-induced reduction in ACA did not necessarily correspond to the areas with relatively high O3-exposure. Widespread distribution of O3-sensitive tree species such as Pinus densiflora and Larix kaempferi, and high ACA were important factors that induced a high risk of O3 impact on the ACA. Therefore, we concluded that not only the accumulated O3-exposure but also the variety of tree habitat, the tree sensitivity to O3 and the ACA among the tree species must be taken into account to assess the risk of O3 impact on the ACA of Japanese conifers. The O3-induced reduction in the total ACA of the three tree species in Japan was estimated to be 0.8%.  相似文献

Population density-dependent dispersal is a well-characterized strategy of animal behavior in which dispersal rate increases when population density is higher. Caenorhabditis elegans shows positive chemotaxis to a set of odorants, but the chemotaxis switches from attraction to dispersal after prolonged exposure to the odorants. We show here that this plasticity of olfactory behavior is dependent on population density and that this regulation is mediated by pheromonal signaling. We show that a peptide, suppressor of NEP-2 (SNET-1), negatively regulates olfactory plasticity and that its expression is down-regulated by the pheromone. NEP-2, a homolog of the extracellular peptidase neprilysin, antagonizes SNET-1, and this function is essential for olfactory plasticity. These results suggest that population density information is transmitted through the external pheromone and endogenous peptide signaling to modulate chemotactic behavior.  相似文献

In recent years, kisspeptin peptides, encoded by kiss genes have been used to manipulate reproductive processes in farmed animals, including fish. Our previous studies demonstrated that the chub mackerel brain expresses kiss1 and kiss2 and intramuscular injection of synthetic Kiss1 pentadecapeptide (Kiss1‐15) but not Kiss2 dodecapeptide (Kiss2‐12) accelerates spermatogenesis in prepubertal male chub mackerel (Scomber japonicus). In the present study, we evaluated their effects in prepubertal female chub mackerel. The gonadosomatic index (GSI) values of experimental fish did not show any significant changes. Condition factor (CF) values increased significantly in Kiss1‐15 treated fish, in comparison with control and GnRH analogue (GnRHa) injected fish. Histologically, only perinucleolar oocytes were found in all experimental fish. However, Kiss and GnRHa treated fish showed a significant increase in the perinucleolar oocyte diameter, in comparison with the control fish. Gene expression analyses revealed decreased expression of gnrh1 in the telencephalon‐preoptic region of the brain of Kiss2‐12 and GnRHa injected fish, in comparison with control fish. In contrast, GnRHa injected fish exhibited higher levels of fshβ in the pituitary, with no changes in the levels of lhβ among different treatments. Levels of circulating sex steroids, testosterone, and estradiol‐17β were significantly higher in Kiss1‐15 injected fish, in comparison with control fish. These results indicate that synthetic kisspeptin peptides and GnRHa can induce oocyte growth in prepubertal female chub mackerel.  相似文献

To understand how soil color is influenced by soil components at the farm scale, we evaluated spatial variation in soil color and related soil properties in Japanese paddy fields. After harvest of rice, 246 surface soil samples were collected in 10-m grids from five contiguous irrigated paddy fields, each with an area of about 0.5 ha. The samples were analyzed to determine color parameters (L*, a*, and b*), and contents of total C, total N, Fe oxides, sand, and loss-on-ignition. The results obtained were modeled and mapped geostatistically. All color parameters indicated strong spatial dependence with long ranges (>85 m). In contrast, total C and N showed short ranges (about 40 m). The contents of Fe oxides, sand, and loss-on-ignition showed intermediate ranges (50–85 m). The ranges of these properties and their distribution patterns suggested that the contents of total C and N were influenced by long-term application of manure and that sand content was influenced by topography and past land consolidation. Further soil color analysis after removal of organic matter or silt plus clay particles revealed that soil organic matter, texture, and Fe oxides affected soil color parameters in a complex manner. Prediction of total C from soil darkness was hindered by the presence of silt plus clay particles containing Fe oxides. On the other hand, citrate-dithionite extractable Fe was estimated accurately from the b* value (yellowness), which can be useful for predicting the occurrence of akiochi (autumn decline) disease of rice at the farm scale.  相似文献

Early life survival is critical to successful replenishment of fish populations, and hypotheses developed under the Growth-Survival Paradigm (GSP) have guided investigations of controlling processes. The GSP postulates that recruitment depends on growth and mortality rates during early life stages, as well as their duration, after which the mortality declines substantially. The GSP predicts a shift in the frequency distribution of growth histories with age towards faster growth rates relative to the initial population because slow-growing individuals are subject to high mortality (via starvation and predation). However, mortality data compiled from 387 cases published in 153 studies (1971–2022) showed that the GSP was only supported in 56% of cases. Selection against slow growth occurred in two-thirds of field studies, leaving a non-negligible fraction of cases showing either an absence of or inverse growth-selective survival, suggesting the growth-survival relationship is more complex than currently considered within the GSP framework. Stochastic simulations allowed us to assess the influence of key intrinsic and extrinsic factors on the characteristics of surviving larvae and identify knowledge gaps on the drivers of variability in growth-selective survival. We suggest caution when interpreting patterns of growth selection because changes in variance and autocorrelation of individual growth rates among cohorts can invalidate fundamental GSP assumptions. We argue that breakthroughs in recruitment research require a comprehensive, population-specific characterization of the role of predation and intrinsic factors in driving variability in the distribution and autocorrelation of larval growth rates, and of the life stage corresponding to the endpoint of pre-recruited life.  相似文献
