Patterns of insertion sequence (IS)629, norV genotype, and Shiga toxin (Stx) genotype distribution were investigated amongst 203 enterohemorrhagic Escherichia coli O157 isolates collected in Yamaguchi Prefecture, Japan, between 2004 and 2013. A total of 114 IS629 patterns were identified; these were divided into eight IS groups (A–H). Ninety isolates carried an intact norV gene, whereas 113 isolates carried a norV with a 204-bp deletion. Other than one isolate from IS group G, all isolates with an intact norV belonged to groups A–F, whereas isolates with a mutant norV belonged to IS groups G and H. Seven stx genotypes were identified, and of those, stx1a/stx2a was predominant (n=105), followed by stx2c (n=32) and stx2a (n=27). The stx1a/stx2a genotype was associated with the mutant norV isolates, whereas isolates with an intact norV had the stx2c genotype. Therefore, certain combinations of IS type and stx genotype appear to be more frequent among O157 clades which may be useful for detection of predominant subtypes in the interest of public health.  相似文献

L-Cysteine (L-Cys) is a non-essential and glycogenic amino acid. Previously, we reported that the intracerebroventricular (i.c.v.) injection of L-Cys induced sedative effects under isolation-induced stress in neonatal chicks. L-Cys has an optical isomer, D-Cys. The aim of this study was to clarify the effect of L-Cys and D-Cys during a stressful condition in chicks. The i.c.v. injection of L-Cys and D-Cys (0.84 µmol) decreased both distress vocalization and spontaneous activity induced by isolation. However, the two cysteine isomers induced different behaviors. L-Cys increased sleep-like behavior while D-Cys caused abnormal behavior including syncope as well as sleep-like behavior. In conclusion, while both L-Cys and D-Cys caused a sedative effect when injected i.c.v, D-Cys caused abnormal behavior and may be detrimental to neonatal chicks.  相似文献

Ex ovo culture of avian embryos can be applied not only to embryology but also to various fields of basic research such as embryo manipulation, toxicology, and regenerative medicine. The windowing method, which facilitates various manipulations and observations by opening a hole in one part of the eggshell, and culture systems using surrogate eggshells, are widely used. Despite this, biology lessons in high schools cover shell-less culture systems, which involve the development of avian embryos in artificial vessels, such as rice bowls, without using surrogate eggshells. However, as embryo development stops at its early stages in this method, it is not possible to continuously observe the development of the embryo. This led to attempts to develop an embryo culture method using a complete artificial culture vessel that does not use surrogate eggshells, and Kamihira et al. (1998) succeeded in hatching quail embryos in an artificial culture vessel using polytetrafluoroethylene membranes. In addition, Tahara succeeded in hatching chick embryos in artificial culture vessels that used cling film made of polymethylpentene and reported their detailed methodology (Tahara and Obara, 2014). These technologies are being applied not only to school education but also to various fields of research.  相似文献

Many large earthquakes are preceded by one or more foreshocks, but it is unclear how these foreshocks relate to the nucleation process of the mainshock. On the basis of an earthquake catalog created using a waveform correlation technique, we identified two distinct sequences of foreshocks migrating at rates of 2 to 10 kilometers per day along the trench axis toward the epicenter of the 2011 moment magnitude (M(w)) 9.0 Tohoku-Oki earthquake in Japan. The time history of quasi-static slip along the plate interface, based on small repeating earthquakes that were part of the migrating seismicity, suggests that two sequences involved slow-slip transients propagating toward the initial rupture point. The second sequence, which involved large slip rates, may have caused substantial stress loading, prompting the unstable dynamic rupture of the mainshock.  相似文献

This study was carried out to investigate the presence of Torque teno sus virus types 1 (TTSuV1) and 2 (TTSuV2) in a longitudinally (14 to 150 days of age) collected paired pooled sera (pSE) and peripheral blood mononuclear cells (pPBMCs) using nested polymerase chain reaction. The detection rate of TTSuV1 in pSE increased from 14 to 90 days of age, but a progressive decline was observed from 120 to 150 days of age, while in pPBMC, a high value was maintained till the end of growing-finishing period. On the contrary, except in PBMCs at 30 days of age, high detection rates of TTSuV2 were found in both pSE and pPBMCs in all sampling ages. The detection rate of TTSuVs between pSE and pPBMCs was positively correlated at all sampling ages except for TTSuV1 at 150 days of age. This is the first study showing the presence of TTSuVs in PBMCs from pigs and describing the in vivo infection dynamics of TTSuV in paired sera and PBMCs during the entire growing and finishing periods of pigs reared in conventional farms.  相似文献

Tick-borne encephalitis virus (TBEV) is a zoonotic agent causing severe encephalitis in humans. Rodent species that are potential hosts for TBEV are widely distributed in various regions in Japan. In this study, we carried out large-scale epizootiological surveys in rodents from various areas of Japan. A total of 931 rodent and insectivore sera were collected from field surveys. Rodents seropositive for TBEV were found in Shimane Prefecture in Honshu and in several areas of Hokkaido Prefecture. These results emphasize the need for further epizootiological and epidemiological research of TBEV and preventive measures for emerging tick-borne encephalitis in Japan.  相似文献

For 6 years, 5 Japanese Black cows of the same herd showed anorexia, depression, and dehydration with no feces in the rectum. Biomedical examination of 3 animals showed severe hypokalemia and hypochloremia. Although the first 3 animals died or were slaughtered (causes unknown), necropsy results showed that the cow in case 4 had intestinal obstruction due to phytobezoar derived from napier grass, fed mainly to the cattle as roughage. Therefore, farmers were recommended to avoid the hard root-stem portion of napier grass as roughage. Consequently, less phytobezoar was recovered from the fifth cow, and no similar clinical case of intestinal obstruction was observed thereafter. This is the first report on intestinal obstruction caused by phytobezoars derived from napier grass.  相似文献

In the present study, we used morphological and genetic analyzes to distinguish bones of domestic boars from those of wild boars. We analyzed 65 Sus bones (cranium, mandible and teeth) stored in three research institutes in Vietnam and in a village in Vietnam. Based on comparison of bucco‐lingual measurements of mandibular parts, the 58 specimens were morphologically classified into two size groups: a large bone group and a small bone group. Analysis of 572‐bp mitochondrial DNA (mtDNA) sequences indicated that the large bones had genetic links to wild boar lineage including Ryukyu, Taiwan and Korean wild boars, and that the small bone group was closely related to East Asian domestic pigs. The phylogenetic analysis and parsimonious networks constructed among mtDNA haplotypes belonging to Ryukyu wild boar lineage showed that the Ryukyu wild boar is closely related to the Vietnamese wild boars, and uniquely miniaturized on their islands after the Ryukyu archipelago became isolated from the Asian continent.  相似文献

A 13-year-old spayed mixed-breed dog was diagnosed with a gastrointestinal stromal tumor (GIST) after histopathological examination of an abdominal mass. Five months after surgical resection of the tumor, we detected the recurrence of GIST with multiple disseminated abdominal lesions. A sequence analysis of cDNA obtained from a biopsy of the recurrent tumors revealed a mutation within exon 9 of the c-kit gene (1523A>T, Asn508Ile), which has been shown to cause ligand-independent phosphorylation of the KIT protein in GISTs and canine mast cell tumors (MCTs). Upon detection of the recurrent tumors, we initiated treatment with imatinib mesylate (10 mg/kg, q 24 hr). After 2 months, the dog achieved complete remission. Our findings indicate that canine GIST, and possibly MCT, may be responsive to molecular-targeted therapy.  相似文献

Regenerative mechanisms that regulate intramuscular motor innervation. including configuration of the neuromuscular connections are thought to reside in the spatiotemporal expression of axon‐guidance molecules. Our previous studies proposed a heretofore unexplored role of satellite cells as a key source of a secreted neural chemorepellent semaphorin 3A (Sema3A) expression. In order to verify this concept, there is still a critical need to provide direct evidence to show the up‐regulation of Sema3A protein in satellite cells in vivo upon muscle injury. The present study employed a Sema3A/MyoD double‐immunohistochemical staining for cryo‐sections prepared from cardiotoxin injected gastrocnemius muscle of adult mouse lower hind‐limb. Results clearly demonstrated that Sema3A expression was up‐regulated in myogenic differentiation‐positive satellite cells at 4–12 days post‐injury period, the time that corresponds to the cell differentiation phase characterized by increasing myogenin messenger RNA expression. This direct proof encourages a possible implication of satellite cells in the spatiotemporal regulation of extracellular Sema3A concentrations, which potentially ensures coordinating a delay in neurite sprouting and re‐attachment of motoneuron terminals onto damaged muscle fibers early in muscle regeneration in synchrony with recovery of muscle‐fiber integrity.  相似文献
