Of all plant materials used to cover the roofs of traditional Japanese buildings, Japanese cypress (Chamaecyparis obtusa) bark, hiwada, has the longest service life and has been used from ancient times. However, wood and bark properties after hiwada harvest have not been evaluated in detail. We studied whether decortication for hiwada production in winter affected xylem and phloem formation. Decorticated trees still preserved all inner bark and part of the
outer bark, and both decorticated and control trees had similar annual ring structures at all stem heights in the xylem and
phloem. In both xylem and inner bark, no significant difference in ring width at any stem height was found between annual
rings before and after decortication. Thus, this study revealed that the decortication of bark for hiwada production does not affect the formation of xylem and the inner and outer bark if decortication is carried out by highly

The ability of the pine wood nematode,Bursaphelenchus xylophilus, a pathogen that causes pine wilt disease, to kill cortical cells of Japanese black pine,Pinus thunbergii, during early development of the disease was conjectured to be a function of nematode developmental stage. A tangential segment
of bark was removed from a 2-cm-long current-year stem. The cortex-exposed segments with cut cortical resin canals were designated
as + RC-segments and those without them as − RC-segments. When a nematode population containing many older juveniles and adults
(NL) was inoculated onto the cut surface, the − RC-segments were still alive 4 d after inoculation, as were non-inoculated
control segments. When cortex-exposed segments were inoculated with either a nematode population containing many younger nematodes
(NS) or with nematodes isolated from inoculated pine cuttings that also contained many younger juveniles, most tissue cells
in − RC-segments died 4 d after inoculation, suggesting that younger juveniles killed pine cells directly, in contrast with
older juveniles and adults. When nematodes were inoculated onto + RC-segments in which they could easily enter resin canals,
both NL and NS killed the segment tissues. This suggests that NL is pathogenic to pine cells while living in resin canals.
Such differences in the pathogenicity of NL and NS to pine parenchymatous cells were also demonstrated in a pathogenicity
assay system using bark peelings, which allowed an estimate of direct attack on the cambial cells by nematodes. Based on these
results, we hypothesize that younger juveniles are pathogenic to pine parenchymatous cells, while adults and older juveniles
are not pathogenic.

Bird species diversity and bird species richness were surveyed in a natural mixed forest (mature forest) and in a coniferous
plantation (30–40 years old) monthly from 1990 to 1994. These forests were 20 km apart and had similar weather conditions
and soil types. There were larch and cedar plantations, and hardwoods along streams in the coniferous plantation. In the natural
mixed forest, 55 bird species were found, whereas 40 species were recorded in the coniferous plantation. The species diversity
of bird communities was significantly higher in the natural mixed forest than in the coniferous plantation mixed with hardwood
trees. This result indicates that the coniferous plantation mixed with hardwood trees, presumed to be a more simplified environment,
is likely to be less inhabitable for many bird species than the natural mixed forest, suggesting a relationship between the
diversity of the forest environment and the diversity of the bird community. A higher number of bird species was recorded
in the small coniferous forest mixed with hardwood trees compared with those in the coniferous plantation. The mixing of broad-leaved
trees in a coniferous plantation was suggested to be effective in increasing number of bird species. These tendencies were
recorded all through the year during our study. In snow-covering periods, both the diversity and the bird species richness
fell in all study sites every year. Snow cover would have greater effect on bird species diversity in the coniferous plantation
than in other forest types probably due to diet shortages caused by the snow cover over the forest floor. 相似文献

In a field study, we compared anatomy and diurnal gas exchange and chlorophyll fluorescence in sunlit mature leaves of Macaranga gigantea (Reichb. f. and Zoll.) Muell. seedlings, saplings, an adult tree and suckers originating from stumps. We tested the hypothesis that the pattern of resource use shifts across various life stages with ontogenetic changes in leaf anatomy and physiology. Among leaves of different developmental stages, seedling leaves were the smallest and thinnest, whereas adult tree leaves were the largest and thickest, and the air space within the lamina was largest in seedling leaves and smallest in adult tree leaves. Photosynthetic nitrogen-use efficiency (PNUE) was higher in seedling and sapling leaves than in adult tree leaves. Mean PNUE in seedling leaves was 1.6 times that in adult tree leaves. Nevertheless, among the developmental stages, net photosynthetic rate (Pn) per unit leaf area was lowest in seedling leaves because they have the lowest nitrogen (N) content per unit leaf area. In situ water vapor stomatal conductance (g(s) at a given leaf-to-air vapor pressure deficit was highest in sapling leaves, suggesting that they have a high hydraulic efficiency per unit leaf area. Among developmental stages, intrinsic water-use efficiency (Pn/g(s)) and photochemical capacity of photosystem II were lowest in seedling leaves. Sapling leaves had the highest N concentration and Pn per unit dry mass and the highest g(s), indicating that the gradual transition from the seedling stage to the sapling stage is accompanied by an accumulation of N in plant bodies and the development of hydraulic systems to counteract unfavorable environmental stresses. The properties of adult tree leaves (low PNUE, high carbon:N ratio, small and dense cells and thick lamina) indicate that, during the transition from the sapling stage to the adult tree stage, the priority of resource use in leaves gradually shifts from enhancement of photosynthetic performance to defense against herbivory and mechanical damage. We tested the hypothesis that, in tropical pioneer tree species, vertical leaf angle contributes to high carbon gain because it minimizes damage caused by high irradiances. Diurnal changes in leaf gas exchange and chlorophyll fluorescence were measured in east-facing (EL), west-facing (WL) leaves, and in leaves artificially held horizontal (HL) in the uppermost canopy of Macaranga conifera (Zoll.) Muell. Arg. Maximum values of net photosynthetic rate (P(n)) for EL and HL reached 12 &mgr;mol m(-2) s(-1), whereas maximum P(n) for WL was only 6 &mgr;mol m(-2) s(-1). Midday depressions of P(n) and stomatal conductance occurred at high photosynthetic photon flux densities (PPFD), especially for HL. Photosystem II quantum yield (DeltaF/F(m)') of HL for a given PPFD at the leaf surface was lower in the afternoon than in the morning. Values of DeltaF/F(m)' for HL measured at dusk were lower than those measured just before dawn, suggesting that HL suffered from high light and heat load. Variations in the morphology and physiology of the canopy leaves were associated with different light environments, and there was circumstantial evidence of a transitional point at a PPFD of about 20-30% of full sunlight. Maximum P(n) and nitrogen (N) content were higher in upper canopy leaves than in lower canopy leaves, and the differences were mainly associated with differences in lamina thickness. We examined the vertical profiles of leaf characteristics within the crowns of two late-successional (Fagus crenata Blume and Fagus japonica Maxim.) and one early-successional tree species (Betula grossa Sieb. et Zucc.) in a Japanese forest. We also assessed the contributions of the leaves in each crown layer to whole-crown instantaneous carbon gain at midday. Carbon gain was estimated from the relationship between electron transport and photosynthetic rates. We hypothesized that more irradiance can penetrate into the middle of the crown if the upper crown layers have steep leaf inclination angles. We found that such a crown has a high whole-crown carbon gain, even if leaf traits do not change greatly with decreasing crown height. Leaf area indices (LAIs) of the two Fagus trees (5.26-5.52) were higher than the LAI of the B. grossa tree (4.50) and the leaves of the F. crenata tree were more concentrated in the top crown layers than were leaves of the other trees. Whole-crown carbon gain per unit ground area (micromol m(-2) ground s(-1)) at midday on fine days in summer was 16.3 for F. crenata, 11.0 for F. japonica, and 20.4 for B. grossa. In all study trees, leaf dry mass (LMA) and leaf nitrogen content (N) per unit area decreased with decreasing height in the crown, but leaf N per unit mass increased. Variations (plasticity) between the uppermost and lowermost crown layers in LMA, leaf N, the ratio of chlorophyll to N and the ratio of chlorophyll a to b were smaller for F. japonica and B. grossa than for F. crenata. The light extinction coefficients in the crowns were lower for the F. japonica and B. grossa trees than for the F. crenata tree. The leaf carbon isotope ratio (delta(13)C) was higher for F. japonica and B. grossa than for F. crenata, especially in the mid-crown. These results suggest that, in crowns with low leaf plasticity but steep leaf inclination angles, such as those of F. japonica and B. grossa trees, irradiance can penetrate into the middle of the crowns, thereby enhancing whole-crown carbon gain. 相似文献

Epidemiological studies suggest that alcohol consumption increases the risk of developing colorectal cancer. However, the data are confounded by numerous cosegregating variables. To cast further light on the relationships between alcohol intake and colon cancer development, 21-day-old male F344/DuCrj rats were fed 200 ppm 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) in their diet for 8 weeks and doses of 0, 0.1, 0.3, 1, 3, 10 and 20% of ethanol in their drinking water ad libitum for 16 weeks thereafter. The rats were sacrificed after 24 weeks of experiment, and aberrant crypt foci (ACF), surrogate lesions for colon cancer, were examined under a light microscope at low magnification. Ethanol was found not to affect the ACF formation at any dose compared with the initiated-controls. Furthermore, ethanol did not alter colon epithelial cell proliferation. These data, obtained by analysis of a colon cancer surrogate marker lesion, indicate that ethanol lacks promotion activity for MeIQx-initiated rat colon carcinogenesis. 相似文献

Koi herpesvirus (KHV), which is believed to be an emerging virus, causes fatal diseases in carps. Since the 1990s, the presence of KHV has been confirmed in several countries. In Japan, from 2003 to 2004, large outbreaks of KHV infection in farmed carps resulted in the death of numerous fishes. From April to May 2004, we collected 43 dead or dying carps exhibiting typical symptoms of KHV infection in Gunma prefecture. To conduct a molecular epidemiologic study of KHV in our prefecture, we amplified DNA polymerase and the major envelope protein genes of KHV derived from carp gills using newly designed primers. We also performed sequence analysis of both genes of KHV. Sensitivity of our PCR method for amplification of DNA polymerase and the major envelope protein genes of KHV was 3 x 10(2) (100 fg) and 3 x 10(3) (1000 fg) copies of KHV genome, respectively. We detected both DNA polymerase and major envelope protein genes in 37 of 43 carps (86%). No mutation was found in both the genes sequenced from 11 strains, which included two foreign strains and one domestic strain. The results suggested that KHV strains derived from carps in our prefecture were closely related genetically to the other KHV strains. 相似文献

20Alpha-hydroxysteroid dehydrogenase (20alpha-HSD) catalyzes the conversion of progesterone to its inactive form 20alpha-dihydroprogesterone (20alpha-OHP). 20Alpha-HSD is expressed in the murine placenta, suggesting a role, yet unidentified, played by this enzyme during the course of pregnancy. To elucidate the possible roles of 20alpha-HSD during pregnancy, 20alpha-HSD gene expression in the placenta was examined by Northern blot analysis, and progestin (progesterone and 20alpha-OHP) concentrations in the maternal and fetal sera and the amniotic fluid were measured by radioimmunoassay in pregnant Shiba goats. The expression of 20alpha-HSD mRNA was observed in the placenta and the intercaruncular part of the uterus during mid to late pregnancy. Analysis by in situ hybridization revealed that 20alpha-HSD mRNA was mainly localized in the endometrial epithelium of the caruncle side of the placenta. Considerable enzyme activity of 20alpha-HSD was also detected in the cytosolic fraction of the placenta and intercaruncular part of the uterus. Although concentrations of progesterone and 20alpha-OHP in the maternal serum showed similar profiles, progesterone levels in the fetal serum stayed extremely low throughout the pregnancy. The 20alpha-OHP concentration in the fetal serum was always higher than that in the maternal serum. In the amniotic fluid, the concentrations of both progesterone and 20alpha-OHP remained at very low levels throughout the pregnancy. These results support the notion that 20alpha-HSD protects the fetus from the cytotoxic effects of progesterone, and thereby maintains the normal development of the fetus. 相似文献
