The combined and separate effects of Cd and wood ash on Archaea from coniferous forest humus were studied in a microcosm experiment. Nonmetric multidimensional scaling of the denaturing gradient gel analysis of polymerase chain reaction amplified 0.9 kb 16S ribosomal DNA fragments revealed changes in archaeal communities due to the ash treatments. Cd with or without ash did not further influence the result. Representatives of the ash and control communities were cloned, grouped by restriction fragment length polymorphism analysis and finally sequenced. All sequences belonged to non-thermophilic Crenarchaea.  相似文献

On acid sandy soils of Niger (West Africa) fertilizer N recovery by pearl millet (Pennisetum glaucum L.) is often more than 100 per cent in years with normal or above average rainfall. Biological nitrogen fixation (BNF) by N2-fixing bacteria may contribute to the N supply in pearl millet cropping systems. For a long-term field experiment comprising treatments with and without mineral fertilizer (F) and with and without crop residue application (CR) a N balance sheet was calculated over a period of six years (1983-1988). After six years of successive millet cropping total N uptake (36-77 kg N ha?1 yr?1) was distinctly higher than the amount of fertilizer N applied (30 kg N ha?1 yr?1). The atmospheric input of NH4-N and NO3-N in the rainwater was about 2 kg N ha?1 yr?1, 70 % in the form of NH4-N. Gaseous NH3 losses from urea (broadcast, incorporated) were estimated from other experiments to amount to 36 % of the fertilizer N applied. Nitrogen losses by leaching (15 to > 25 kg N ha?1 yr?1) were dependent on the treatment and on the quantity and distribution of single rainfall events (>50 mm). Decline in total soil N content (0-60 cm) ranged from 15 to 48 kg N ha?1 yr?1. The long-term N balance (1983-1988) indicated an annual net gain between 6 (+CR-F) and 13 (+CR+F) kg N ha?1 yr?1. For the control (-CR-F) the long-term N balance was negative (10 kg N ha?1 yr?1). In the treatment with crop residues only, the N balance was mainly determined by leaching losses, whereas in treatments with mineral fertilizer application the N balance depended primarily on N removal by the millet crop. The annual net gain in the N balance increased from 7 kg ha?1 with mineral fertilizer to 13 kg ha?1 in the combination mineral fertilizer plus crop residues. In both the rhizosphere and the bulk soil (0-15 cm), between 9 and 45% of the total bacterial population were N2-fixing (diazotrophic) bacteria. The increased N gain upon crop residue application was positively correlated with an increase in the number of diazotrophic and total bacteria. The data on bacterial numbers suggest that the gain of N in the longterm N balance is most likely due to an N input by biological nitrogen fixation. In addition, evidence exists from related studies that the proliferation of diazotrophs and total bacteria in the rhizosphere due to crop residue application stimulated root growth of pearl millet, and thus improved the phosphorus (P) acquisition in the P deficient soil.  相似文献

ABSTRACT

One would expect foliar applied phosphorus (P) to have higher use efficiencies than when applied to the soil, but limited information is available concerning this. Experiments were conducted in 2002, 2003, and 2004 to determine the effect of foliar applications of P on winter wheat grain yields, P uptake, and use efficiency. Twelve treatments containing varying foliar P rates (0, 1, 2, and 4 kg ha? 1 in 2002 and 2003 and additional 8, 12, 16, and 20 kg ha? 1 in 2004) with and without pre-plant rates of 30 kg ha? 1 were evaluated. Foliar applications of P at Feekes 7 generally increased grain yields and P uptake versus no foliar P. Use efficiency was higher when P was applied at Feekes 10.54. Results from this study suggested that low rates of foliar applied P might correct mid-season P deficiency in winter wheat, and that might result in higher P use efficiencies.  相似文献

Formation of flavor compounds from branched-chain alpha-keto acids in fermented foods such as cheese is believed to be mainly an enzymatic process, while the conversion of phenyl pyruvic acid, which is derived from phenylalanine, also proceeds chemically. In this research, the chemical conversion of alpha-keto acids to aldehydes with strong flavor characteristics was studied, with the main focus on the conversion of alpha-ketoisocaproic acid to the aldehyde 2-methylpropanal, and a manganese-catalyzed reaction mechanism is proposed for this conversion. The mechanism involves keto-enol tautomerism, enabling molecular oxygen to react with the beta-carbon atom of the alpha-keto acid, resulting in a peroxide. This peroxide can react in several ways, leading to unstable dioxylactone or noncyclic intermediates. These intermediates will break down into an aldehyde and oxalate or carbon oxides (CO and CO(2)). All the alpha-keto acids tested were converted at pH 5.5 and in the presence of manganese, although their conversion rates were rather diverse. This chemical reaction might provide new ways for controlling cheese flavor formation with the aim of acceleration of the ripening process or diversification of the flavor characteristics.  相似文献

Orange oil is composed largely of terpene hydrocarbons but is a source of flavor and fragrance compounds (oxygenated) that are present in low concentrations. To increase the ratio of oxygenated compounds to terpene hydrocarbons, orange oil was partially fractionated by adsorption of the oxygenated compounds onto porous silica gel, with full utilization of its adsorbent capacity, and then further purified by desorption into supercritical carbon dioxide. The desorption of 24 compounds was monitored by GC and GC-MS. Adsorption alone removed three-fourths of the terpene hydrocarbons, and fractional extraction by supercritical carbon dioxide (SC-CO(2)) improved the separation further. Response surface methodology was used in the experimental design, and regression analysis was used to determine the effects of process variables. Extraction at low temperatures and flow rates improved separation by SC-CO(2). Decanal was concentrated to 20 times that of the feed oil by using SC-CO(2) at 13.1 MPa, 35 degrees C, and 2 kg/h. The systems were operating at close to equilibrium conditions because of the fine dispersal of the oils and the excellent mass transfer properties of supercritical carbon dioxide.  相似文献

The isotopic investigation of vanillin has been extended to the new sources of natural precursors of vanillin recently introduced with a view of obtaining natural vanillin by biotechnological processes. To check the consistency of the isotopic composition of vanillin with that of the corresponding aromatic fragment, a selective degradation reaction into guaiacol was carried out. The reaction was shown to proceed without significant isotopic fractionation at the sites of interest, and an optimized procedure was defined from the results of an experimental design involving the quantity of reagent and the temperature and duration of the experiment. Guaiacol, which can be easily obtained in a reasonable time, is an interesting isotopic probe for carbon- and oxygen-isotope ratio mass spectrometry (IRMS) determinations. It provides (13)C information specific to the aromatic fragment and, combined with delta(13)C values measured on vanillin itself, it improves the authentication potential of carbon-IRMS. Thus, the natural status of ferulic acid may be characterized by significant (13)C depletion at the formyl site. Similarly, the oxygen-18 content of guaiacol is a better authentication tool than delta(18)O of vanillin because it does not suffer the drawback of being altered by chemical exchange of the sp(2) oxygen atom with water in industrial or laboratory procedures. Although collaborative studies are still necessary to improve the interlaboratory reproducibility of the delta(18)O parameters, consistent results can be obtained in an intralaboratory context. It is shown in particular that chemical oxidation of ferulic acid is characterized by a relative enrichment of the aromatic moiety of vanillin.  相似文献

Plant canopy optimization models predict that leaf nitrogen (N) distribution in the canopy will parallel the vertical light gradient, and numerous studies with many species have confirmed this prediction. Further, it is predicted that for a given canopy leaf area, a low vertical light extinction coefficient will promote rapid growth. Therefore, the ideal canopy of fast-growing plants should combine high leaf area index with a low light extinction coefficient; the latter being reflected in a flat vertical leaf N gradient throughout the canopy. Based on data from an experimental Salix stand (six varieties) grown on agricultural land in central Sweden, we tested the hypothesis that shoot growth is correlated with vertical leaf N gradient in canopies of hybrid willows bred for biomass production, which could have implications for Salix breeding. Tree improvement research requires screening of growth-related traits in large numbers of plants, but assessment of canopy leaf N gradients by chemical analysis is expensive, time-consuming and destructive. An alternative to analytical methods is to estimate leaf N gradients nondestructively with an optical chlorophyll meter (SPAD method). Here we provide a specific calibration for interpreting SPAD data measured in hybrid willows grown in biomass plantations on fertile agricultural land. Based on SPAD measurements, a significant and inverse relationship (r(2) = 0.88) was found between shoot biomass growth and vertical leaf N gradient across canopies of six Salix varieties.  相似文献

The effect of decortication on chemical composition and degradability characteristics of four Danish oat varieties was investigated. Effective degradability (ED) and post-ruminal disappearance (PRD) were measured by in situ and mobile bag techniques respectively. Decorticated oat showed higher (p = .01) concentrations of crude protein (CP; 134 vs. 108 g/kg DM) and crude fat (71.6 vs. 53.1 g/kg DM) and a higher (p = .001) organic matter digestibility (OMD; 888 vs. 703 g/kg OM) than oat. The content of total fatty acids (FA) in DM was higher in decorticated oat. The proportion of linoleic acid (C18:2 n6) increased (p < .05) due to decortication, while the linolenic acid (C18:3 n3) proportion of total FA decreased in decorticated oat. Decortication increased (p = .01) the concentration of amino acids (AA), but the proportion of lysine in total AA decreased (p < .002). Effective degradability (ED) of both DM and CP was (p < .001) higher in decorticated oat. Decortication increased the total tract disappearance (TTD) and PRD of CP (p < .001). In conclusion, decortication can be used as a practical approach to increase the nutritional value of oat.  相似文献

Six young tortoises Testudo marginata Schoepff, 1792 were experimentally infected with Hemolivia mauritanica (Sergent et Sergent, 1904). The prepatent period ranged from 6 to 8 weeks. Young, smaller, club-like forms (6-9 x 3-6 Am) of gametocytes appeared in the peripheral blood first, whereas mature, elongated, cylindrical forms (9-12 x 5-7 Am) were detected after 1-2 weeks and predominated during later patency. Three of the infected tortoises were euthanized and dissected to study the endogenous stages. Meronts occurred in the cells of the reticulo-endothelial system and in the erythrocytes; these were observed mostly in parenchymatous organs. Mature forms measured 14.2 x 9.3 microm and contained 7-12 merozoites. Cysts with two (exceptionally one) cystozoites were also found predominantly in parenchymatous organs and measured 14.8 x 7.9 microm. Pathological changes attributable to Hemolivia were mild and limited to liver and kidneys. The role of individual developmental stages of haemogregarines is discussed with respect to evolution of heteroxenous life cycle and long-term persistence of parasites in their intermediate hosts.  相似文献
