Effect of L-lysine Added in Diluent on the Fresh Semen Quality of Dorper Sheep

In order to explore the effect of different L-lysine levels added in semen dilution on the fresh Dorper sheep quality stored at liquid state,3 health Dorper ram were used to collect semen which were equal-packaged and added to solution containing different levels (0,0.1,0.2,0.3,0.4 and 0.5 g) L-lysine in 120 mL diluent and at 0,6,24,30,48,54,72 and 78 h,the sperm motility,membrane integrity and acrosome integrity were detected.The results showed that the sperm motility in groups added 0,0.1 and 0.2 g L-lysine were higher than other groups,and that in 0.4 and 0.5 g L-lysine groups decreased faster than other groups;The sperm motility in 0.5 g L-lysine group reduced to 0 at 30 h,and it reduced to 0 at 48 h in the group added 0.4 g L-lysine;The effective survival time and survival index in the group added 0.1 g L-lysine was higher than other groups (P< 0.05);The membrane integrity of 0.1 g group was the highest,that had no significant difference with control group from 0 to 54 h (P >0.05),while after 72 h the two groups had significant differences (P< 0.05).The membrane integrity in 0.4 and 0.5 g groups were the worst,and that was significant different with other groups (P< 0.05);The acrosome integrity of 0.1 g group was the highest which had no significant difference with control group (P >0.05),0.4 and 0.5 g groups were the worst,the difference between them was not significant (P >0.05),while was extremely significant difference with other groups except for 0 and 6 h (P< 0.01).The results suggested that dilution added a high concentration of L-lysine could inhibit sperm quality,when the count of L-lysine was more than 0.2 g,sperm quality was significantly decreased,adding 0.1 g L-lysine could improve Dorper sheep fresh sperm quality stored at liquid state.     

TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE2分泌的影响

[目的] 进一步探讨牛磺鹅去氧胆酸（taurochenodeoxycholic acid,TCDCA）在抗炎免疫方面的潜在调节作用。[方法] 以AA大鼠成纤维样滑膜细胞作为研究对象,采用ELISA方法检测TCDCA和IL-1β作用下AA大鼠成纤维样滑膜细胞上清液中PGE₂的含量,分析TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE₂分泌情况的影响。[结果] TCDCA能够对IL-1β刺激下AA大鼠纤维样滑膜细胞PGE₂的分泌产生下调作用（P<0.05）。[结论] TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE₂的分泌具有抑制作用,为TCDCA在兽医临床应用提供依据。     

Expression,Purification and Activity Evaluation of Mb1230 Gene of Mycobacterium bovis

To explore the value of Mb1230 protein of Mycobacterium bovis in the diagnosis of bovine tuberculosis,we obtained the Mb1230 gene by PCR and constructed recombinant plasmid pET-22b-Mb1230.Recombinant Mb1230 protein was obtained by IPTG induction and purified by affinity chromatography.The activity of the recombinant protein was evaluated by TST test,IGRA test and indirect ELISA.The size of the recombinant protein matched with the theoretical value proved by SDS-PAGE;Western blotting result showed that the recombinant protein could react with mouse anti-His antibody,and had specific band;The results of TST test,IGRA test and indirect ELISA test also showed the recombinant protein had antigenic activity.The results indicated the recombinant protein Mb1230 had good B cell and T cell activity,so,it had the potential application in the diagnosis of bovine tuberculosis.

棚室专用薄皮甜瓜新品种‘永甜4号’的选育

‘永甜4号’薄皮甜瓜是用地方薄皮甜瓜品种自交系‘白B10’为母本、‘郑引55’自交系‘A97-I-14-63’为父本配制的杂交一代新品种。该品种早熟,全生育期93d,果实发育期31d。在棚室种植坐瓜率高,产量稳定,果皮浅黄色,梨形,单果质量450⁴80g,极少裂瓜,畸形瓜少,中心可溶性固形物含量12%480g,极少裂瓜,畸形瓜少,中心可溶性固形物含量12%¹3%。货架期长,耐运输。2006年通过黑龙江农作物品种审定委员会审定。     

Effects of Addition of Three Types of Biochar on the Soil Nitrogen Balance and Cotton Yield

The objective of this study was to determine the effect of biochar addition on the soil N balance and cotton yield in a drip-irrigated cotton field. The experiment included four soil amendments: control, cotton straw biochar, corn cob biochar, and poultry manure biochar. The biochar was applied at a rate of 4.5 t·hm^－2 except control. The results showed that all three types of biochar significantly reduced both the apparent soil N loss and the N surplus. The effect of cotton straw biochar treatment was most significant, relative to the control. Plant nitrogen uptake differed significantly between the different treatments, decreasing in the order cotton straw biochar > poultry manure biochar > corn cob biochar. Cotton yield also differed significantly between the different treatments, decreasing in the order poultry manure biochar > corn cob biochar, and cotton straw biochar.     

断奶日龄对五指山仔猪生长性能和血清生化指标的影响

本试验旨在研究断奶日龄对五指山仔猪生长性能和血清生化指标的影响。选取日龄相近(±1 d)、体重[(0.56±0.07)kg]相近的五指山仔猪96头,随机分成4组,每组设4个重复,每个重复6头猪。4组仔猪分别在21、28、35和42日龄断奶。试验从8日龄开始,至56日龄结束,试验期间定期测定体重、采食量和血清生化指标。结果表明:各组仔猪断奶后平均日增重(ADG)出现不同程度的下降,且断奶日龄越早,ADG下降幅度越大;35日龄断奶组和42日龄断奶组21~28日龄、28~35日龄、35~42日龄、49~56日龄阶段的ADG显著高于21日龄断奶组(P0.05)。各组仔猪断奶后平均日采食量(ADFI)显著增加(P0.05),42日龄以后,各组ADFI无显著差异(P0.05)。21日龄断奶组、28日龄断奶组、35日龄断奶组血清总蛋白(TP)、白蛋白(ALB)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)含量以及谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)活性在断奶后7 d均发生显著变化(P0.05),并以21日龄断奶组波动最大;与35日龄断奶组和42日龄断奶组相比,21日龄断奶组28、35日龄时血清TP、ALB、IgG、IgM含量显著降低(P0.05),ALT、AST、LDH活性显著提高(P0.05),到56日龄时,血清TP、IgG、IgM含量仍显著低于35日龄断奶组和42日龄断奶组(P0.05)。21日龄断奶组血清甘油三酯(TG)、胆固醇(CHOL)含量在28~56日龄间存在显著变化(P0.05),35日龄断奶组和42日龄断奶组血清TG、CHOL含量在整个试验期内均无显著变化(P0.05)。35日龄断奶组血清TP、ALB、IgG、IgM含量以及ALT、AST、LDH活性除在42日龄时与42日龄断奶组存在显著差异(P0.05)外,其余时间点与42日龄断奶组均无显著差异(P0.05)。由此得出,35或42日龄断奶的五指山仔猪在生长性能、血清生化指标及免疫机能方面优于21日龄断奶的五指山仔猪。     

农村非正式组织对农户收入影响的实证分析——以浙江省慈溪市为例

农民作为“三农”改革中受影响最深的群体，有着强烈的利益表达愿望，目前农村生产现状及国家相关法制尚不完善，使得农户的利益受到了损害，农户需要寻找一种途径来使自己受损的这部分利益得到补偿。在这种情况下，非正式组织的出现很好地弥补了这一空缺。国家可以根据非正式组织的特点，了解其运行模式的优越性，制定并健全相关政策制度，进行正面引导；另外对一些具有正面借鉴意义的非正式组织进行保护和支持，派遣相应的技术人员进行实地指导和培训，同时国家可以给予适当的补助，使其健康有序持续发展。
