A cluster of Chlamydia psittaci (C. psittaci) cases was reported in Zhejiang Province, China, 2019. This study evaluates the extent of the outbreak and determines the source of infection. Real-time PCR and sequencing of the ompA gene of C. psittaci were performed to identify the cases, the domesticated poultry and close contacts. The index patient was a 76-year-old woman with chronic vertigo, and Case 2 was a 64-year-old female farmer with herpes zoster. Both women bought psittaci-infected chickens or ducks from the same mobile street vendor and raised them for 10 days and 23 days before fever onset. There were no direct contact between the two women. C. psittaci test was positive for the two patients, one sick chicken, three healthy ducks and the vendor's chicken cage. Phylogenetic analysis showed that all seven C. psittaci positive samples carried identical ompA genotype A of C. psittaci. Of all of the patients' 148 close contacts, none tested positive for C. psittaci, or developed acute respiratory symptoms. Both patients were discharged after a 4-week hospital stay. In conclusion, the source of this cluster was the poultry infected with C. psittaci, which occasionally cause infections in farmers, but inter-human transmission seems unlikely.  相似文献

Mannan-binding lectin (MBL), a pattern recognizing serum protein, participates in the innate immune system of mammals as an opsonin. In humans, single-nucleotide polymorphisms (SNPs) in MBL2 gene were found to cause various innate immune dysfunctions. In the present study, we discovered three single-nucleotide polymorphisms of the MBL1 gene in Chinese native cattle and analyzed their associations with milk traits. By screening the genetic variation of MBL1 in 1053 individuals of three Chinese native cattle breeds including China Holstein, Luxi Yellow and Bohai Black using created restriction site–polymerase chain reaction (CRS–PCR), PCR–restriction fragment length polymorphism (PCR–RFLP) and DNA sequencing techniques, three new SNPs, g.855G>A, g.2651G>A and g.2686T>C, were found to have allele frequencies of 0–12.65%, 24.07–42.39% and 56.95–73.68%, respectively. While SNP g.855G>A is located within intron ?, the other two SNPs reside in the exon II region with one mutation being non-synonymous (GTT (Val) > ATT (Ile)) and the other synonymous (GCT (Ala) > GCC (Ala)). Among the 596 Chinese Holstein cattle with at least 3 lactation Dairy Herd Improvement (DHI) records, eight different haplotypes and 19 genotype combinations were detected. Statistical analyses revealed no correlation between either g.855G>A or g.2686T>C and somatic cell score (SCS), however significant association was found between g.2651G>A and SCS, suggesting a possible role of this SNP in the host response against mastitis. Our data also suggested that the combined genotypes of GGC/AAC with the lowest SCS, AAT/AAT with the highest protein content and AGC/AGC with the highest 305-d milk yield were favorable combinations for mastitis resistance and milk production traits. Therefore, GGC/AAC, AAT/AAT and AGC/AGC can be used as possible candidates for marker-assisted selection in the dairy cattle breeding program.  相似文献

The objective of this study was to investigate the pharmacokinetics and bioavailability of valnemulin in broiler chickens after intravenous (i.v.), intramuscular (i.m.) and oral administrations of 10 mg/kg body weight (bw). Plasma samples were analyzed by high‐performance liquid chromatography–tandem mass spectrometry (HPLC‐MS/MS). Pharmacokinetic characterization was performed by non‐compartmental analysis using WinNonlin program. After intravenous administration, distribution was wide with the volume of distribution based on terminal phase(Vz) of 4.27 ± 0.99 L /kg. Mean valnemulin t1/2β(h), Clβ(L /h /kg), Vss (L /kg) and AUC(0–∞)(μg·h /mL) values were 2.85, 0.99, 2.72 and 10.34, respectively. After intramuscular administration, valnemulin was rapidly absorbed with a Cmax of 2.2 μg/mL achieved at 0.43 h (tmax), and the absolute bioavailability (F) was 88.81%; and for the oral route the same parameters were 0.66 ± 0.15 μg/mL, 1.54 ± 0.27 h and 74.42%. A multiple‐peak phenomenon was present after oral administration. The plasma profile of valnemulin exhibited a secondary peak during 2–6 h and a tertiary peak at 32 h. The favorable PK behavior, such as the wide distribution, slow elimination and acceptable bioavailability indicated that it is likely to be effective in chickens.  相似文献

鹦鹉热衣原体(Chlamydia psittaci)是一种人兽共患病原体,能引起自然疫源性衣原体病,目前广泛分布于世界各地。本研究应用环介导等温扩增技术(LAMP),针对鹦鹉热衣原体23S RNA基因序列设计引物,并进行筛选以及敏感性和特异性试验,建立了鹦鹉热衣原体恒温荧光扩增方法。该方法在63℃恒温条件下1 h内即可显示结果,操作简单、快速,特异性强,灵敏度可达100拷贝/μL,且与流产衣原体、动物布鲁氏菌、牛结核分枝杆菌、沙门氏菌、大肠杆菌、金黄色葡萄球菌无交叉反应;用国产仪器可直接通过检测荧光信号判读结果,既增强了准确性,也避免了开盖污染产生假阳性;应用建立的LAMP方法对实验室保存的临床DNA样品进行检测,发现检测结果与QPCR相同。该方法的建立弥补了传统检测技术的不足,为实现鹦鹉热衣原体的现场快速诊断提供了技术支持。  相似文献
