Ethylidene-bridged Flavan-3-ols in red wine and correlation with wine age   总被引:2,自引:0,他引:2  
Condensed tannins are responsible for astringency and bitterness and participate in the color stability of red wines. During wine making and aging, they undergo chemical changes including, for example, acetaldehyde-induced polymerization. Following this study, the ethylidene-bridged flavan-3-ols were monitored in different vintage wines made from grapes collected in the same vineyard in three wineries in Bordeaux, Pauillac, and Saint Julien. Flavan-3-ol ethylidene bridges were quantified by wine 2,2'-ethylidenediphloroglucinol (EDP) phloroglucinolysis. This method was based upon the analysis of EDP, a product formed after acid-catalyzed cleavage of wine flavan-3-ols in the presence of excess phloroglucinol. The flavan-3-ol ethylidene bridges were then compared to flavan-3-ol contents (phloroglucinolysis), phenolic contents, and color measurements. Low amounts of flavan-3-ol ethylidene bridges (0.8-2.5 mg L(-1)) were quantified in wines. Flavan-3-ol ethylidene bridges represent less than 4% of flavan-3-ol bonds, but the proportion of these linkages relative to native interflavan bonds increased with wine age. This proportion correlated with pigmented polymers.  相似文献

Accumulation and compositional changes of flavonols, proanthocyanidins, and anthocyanins were measured in Vitis vinifera L. cv. Pinot noir in shaded and exposed treatments. In addition, extraction of these compounds into a model wine solution was measured. The study was conducted in a commercial vineyard within a uniform zone of relatively low vigor vines. Light exclusion boxes were installed on pairs of clusters on the same shoot (shaded treatment), and a second set of clusters on an adjacent shoot were labeled as the exposed treatment. Fruit samples were harvested at the onset of ripening (véraison) and at commercial harvest. Cluster shading resulted in a substantial decrease in mg/berry accumulation of flavonols and skin proanthocyanidins and minimal differences in anthocyanins. In analyzing seed proanthocyanidins by phloroglucinolysis, shaded and exposed treatments were similar at véraison; however, by harvest, the shaded treatment had higher extension and terminal subunits (nmol/seed) as compared to the exposed treatment. For skin proanthocyanidins, shaded fruit was lower for all subunits (nmol/berry) at both véraison and harvest. Shading caused an increase in the proportion of (-)-epicatechin and a decrease in (-)-epigallocatechin at harvest in skin extension subunits. Seed proanthocyanidins in shaded fruit contained a lower proportion of (+)-catechin and a higher proportion of (-)-epicatechin-3-O-gallate in extension subunits and a lower proportion of (+)-catechin and (-)-epicatechin-3-O-gallate and a higher proportion of (-)-epicatechin in terminal subunits. For anthocyanins, the shaded treatment had a proportional reduction in delphinidin, cyanidin, petunidin, and malvidin and a large increase in peonidin glucosides. The model extractions from the two treatments paralleled differences in the fruit with a lower concentration of flavonols, anthocyanins, and proanthocyanidins in the shaded treatment. The skin proanthocyanidin percent extraction was found to be approximately 17% higher in the exposed model extraction than the shaded treatment.  相似文献

为了明确草莓内源SA(Salicylic acid, 简称SA)含量与其抗病性的关系,并探索人们从草莓等植物性食物中摄取SA的总量,本研究用HPLC分析比较了露地与设施栽培条件下五个对白粉病抗性不同的草莓品种果实的SA含量。结果表明,所测五种草莓红熟期果实内源总SA含量差别明显,露地栽培条件下为1.904µg/g~5.023µg/g,设施栽培条件下为1.068µg/g~4.201µg/g,与各品种的抗白粉病能力无明显相关性。草莓果实中SA均以结合态为主,五个品种结合态SA含量是游离态SA的4~20倍。游离态SA和结合态SA含量都随着果实的成熟而逐渐下降。说明草莓果实中内源SA含量不仅受基因型的影响,并且受栽培环境和成熟度的影响。  相似文献

The objective of this study was to test the efficacy of a compounded long-acting progesterone formulation (BioRelease P4 LA 150; BETPHARM, Lexington, KY) containing 150 mg progesterone/ml for pregnancy maintenance in mares after prostaglandin (PG) F-induced luteolysis. On day 18 of gestation, mares were randomly assigned to one of four groups (n = 7/group): (1) saline-treated control (Saline); (2) PGF-treated control (PGF); (3) PGF- and Regu-Mate-treated (Regu-Mate); and (4) PGF- and BioRelease P4 LA 150-treated (BioRelease). On day 18, Saline mares received 1 ml sterile saline IM, whereas PGF, Regu-Mate, and BioRelease mares received 250 μg cloprostenol IM. Beginning on day 18, Regu-Mate mares received 10 ml Regu-Mate orally once daily and BioRelease mares received 10 ml BioRelease P4 LA 150 containing 150 mg/ml progesterone IM once every 7 days; treatments were continued until day 45 or until pregnancy loss occurred. Pregnancy diagnosis was performed every 3 days between days 18 and 45 (or until pregnancy loss). Pregnancy loss was defined as complete absence of a discernible embryonic vesicle as determined with transrectal ultrasonography. Pregnancy loss rates between days 18 and 45 were: Saline, 1/7; PGF, 7/7; Regu-Mate, 1/7; and BioRelease, 0/7. The pregnancy loss rate was higher (P < .01) in PGF-treated control mares compared with the other groups. There were no differences (P > .1) in pregnancy loss rates among the saline-treated control, Regu-Mate-treated, and BioRelease P4 LA 150-treated mares. These results indicate that intramuscular administration of BioRelease P4 LA 150 containing a total of 1.5 g progesterone every 7 days provided a sufficient level of progesterone to maintain pregnancy between days 18 and 45 of gestation in mares that lacked an endogenous source of progesterone; therefore, this long-acting formulation of progesterone appears to be an efficacious and suitable alternative to currently available progesterone formulations that require daily administration.  相似文献

Intensity-modulated radiation therapy (IMRT), especially image-guided IMRT as represented by helical tomotherapy, is a novel approach to therapy and is rapidly evolving. Both of these forms of therapy aim to allow targeted radiation delivery to the tumor volume while minimizing dose to the surrounding normal tissues. Adaptive radiation therapy and conformal avoidance are possible with intensity-modulated therapy and helical tomotherapy, which offer opportunities for improved local tumor control, decreased normal tissue toxicity, and improved survival and quality of life. Human and veterinary patients are likely to benefit from the continued development of this radiation delivery technique, and data over the next several years should be crucial in determining its true benefit.  相似文献

Background: Disease‐specific biomarkers hold diagnostic promise in both human and veterinary medicine, but serum biomarkers in low concentrations may be masked by the presence of abundant proteins, mostly albumin and IgG. Methods to deplete albumin and IgG exist, but efficacy of these methods for depleting equine serum of these proteins has not been established. Objective: The aim of this study was to determine if albumin and IgG could be depleted from equine serum using several commercially available kits and procedures. Methods: One‐dimensional gel electrophoresis followed by densitometry was used to determine percent of albumin, IgG, and both in pooled serum from 3 horses before and after application of 7 depletion methods. Repeatability was determined by applying the 2 best methods to serum samples from 6 grade horses. Results: For pooled serum, depletion rates varied from 35–90% for albumin and 0–94% for IgG. In the repeatability study, the ProteoExtract method combined with protein G Sepharose beads to remove additional IgG provided the best overall performance with 66% albumin depletion and 100% IgG depletion. A protocol using protein G Sepharose beads to remove IgG followed by ethanol precipitation of nonalbumin proteins with albumin remaining in the supernatant was the second most effective, with 85% albumin depletion and 55% IgG depletion. Although a multiprotein immunodepletion column effectively removed 90% of the albumin, the method was ineffective at removing IgG. Conclusion: Albumin and IgG removal kits optimized for human use have variable efficacy for equine serum. Combined use of the ProteoExtract kit and manual incubation with protein G Sepharose beads provided the most effective depletion.  相似文献

Lyme borreliosis (LB) is a tick‐borne disease caused by Borrelia burgdorferi sensu lato complex. In Europe, it is predominately transmitted by the sheep tick, Ixodes ricinus. Compared with other European countries, the United Kingdom (UK) is considered to have a low incidence of LB, although this varies regionally. To determine whether an association exists between tick bite consultations and LB incidence in the UK, retrospective questionnaires were sent to general practitioners (GPs) in high (Wiltshire), medium (Cumbria) and low (Wales) incidence areas. During 2011, the greatest incidence of consultations for tick bites was reported by GPs in Cumbria (204 consultations per 100,000 inhabitants), followed by Wiltshire (160 per 100,000 population) and Wales (54 per 100,000 population). In Wiltshire and Cumbria, GPs predominantly provided advice on tick removal, whilst Welsh GPs mostly advised patients on tick bite prevention. Focusing on Cumbria during 2011–2013, 72.5% of GPs removed ticks from patients (incidence of 101 consultations per 100,000 population), and more GPs diagnosed LB based on clinical features than laboratory‐confirmed diagnoses. To date, this is the first study to investigate the incidence of tick bite consultations and LB in England and Wales.  相似文献

Bacteria have developed mechanisms to communicate and compete with each other for limited environmental resources. We found that certain Escherichia coli, including uropathogenic strains, contained a bacterial growth-inhibition system that uses direct cell-to-cell contact. Inhibition was conditional, dependent upon the growth state of the inhibitory cell and the pili expression state of the target cell. Both a large cell-surface protein designated Contact-dependent inhibitor A (CdiA) and two-partner secretion family member CdiB were required for growth inhibition. The CdiAB system may function to regulate the growth of specific cells within a differentiated bacterial population.  相似文献
