In nature, key molecular processes such as communication, replication, and enzyme catalysis all rely on a delicate balance between molecular and supramolecular chirality. Here we report the design, synthesis, and operation of a reversible, photoresponsive, self-assembling molecular system in which molecular and supramolecular chirality communicate. It shows exceptional stereoselectivity upon aggregation of the molecules during gel formation with the solvent. This chirality is locked by photochemical switching, a process that is subsequently used to induce an inverted chiral supramolecular assembly as revealed by circular dichroism spectroscopy. The optical switching between different chiral aggregated states and the interplay of molecular and supramolecular chirality offer attractive new prospects for the development of molecular memory systems and smart functional materials.  相似文献

Biotic soil parameters have so far seldom played a role in practical soil assessment and management of grasslands. However, the ongoing reduction of external inputs in agriculture would imply an increasing reliance on ecosystem self-regulating processes. Since soil biota play an important role in these processes and in the provision of ecosystem services, biological soil parameters should be an integral part of soil assessment. The general objective of the current study is to investigate to what extent biotic soil parameters provide additional value in soil quality assessment of grassland on sandy soils. We measured abiotic and biotic soil parameters together with process parameters underlying ecosystem services in 20 permanent production grasslands. Cross-validated stepwise regression was used to identify abiotic and biotic soil parameters that explained the soil ecosystem services soil structure maintenance, water regulation, supply of nutrients, and grassland production, respectively.Process parameters underlying the ecosystem service soil structure maintenance such as bulk density and the percentage of sub-angular blocky elements were mainly influenced by SOM and its qualities. The correlations between penetration resistance at 0-10 cm and the percentage of soil crumbs with earthworms suggested a relationship to earthworm activity. Parameters underlying the service of water regulation showed no clear relationship to biotic soil parameters. Water infiltration rate in the field was explained by the penetration resistance at 10-20 cm. Process parameters underlying the service of nutrients’ supply such as the potentially mineralizable C and N were mainly determined by soil total N. The potential C and N mineralization were more related to biotic soil parameters, whereby each parameter was the other’s antithesis. The grassland production without N fertilization viz. the nitrogen supply capacity of the soil measured as N yield, was mainly explained by soil organic matter (SOM) and soil moisture, and to a lesser extent by soil total N. One gram of SOM per kg of dry soil corresponded to 3.21 kg N yield ha−1, on top of a constant of 15.4 kg N ha−1. The currently applied calculations in the Dutch grassland fertilization recommendation, underestimated in 85% of the production grasslands, the measured nitrogen supply capacity of the soil by on average 42 kg N ha−1 (31%). This legitimizes additional research to improve the currently applied recommendations for sandy soils. The response of N yield to N fertilization ranged from 35 to 102%. This wide range emphasizes the importance of a better recommendation base to target N fertilizer. The response of N yield to N fertilization was predicted by the total number of enchytraeids, the underlying mechanism of which needs further investigation on different soil types. This knowledge can be important for the optimal use of fertilizer and its consequences for environmental quality.  相似文献

To investigate the incidence of co-colonization of different strains of Campylobacter species present in canine and feline stool samples, isolates were recovered by culture from 40 samples from dogs (n=34) and cats (n=6). Animals were of different ages, with diarrhoea or without clinical signs. Three isolation procedures were used: two selective agars and a filtration method. In each stool sample, multiple colonies were identified to the species level by PCR, subsequently genotyped by Amplified Fragment Length Polymorphism (AFLP) and pattern similarities (451 isolates) were calculated to investigate their phylogenetic relationships. Genetic heterogeneity of strains in individual stool samples was detected within the species Campylobacter jejuni, C. upsaliensis and C. helveticus, though to a different degree in dogs and cats. In 3 of the 34 (9%) canine samples, more than one genotype of the same Campylobacter species was present, while strain variation was detected in four of the six feline samples. The results show that preferably, multiple colonies should be analyzed in molecular epidemiological and aetiological studies.  相似文献

Potato cyst nematodes (PCNs) are quarantine organisms, and they belong to the economically most relevant pathogens of potato worldwide. Methodologies to assess the viability of their cysts, which can contain 200 to 500 eggs protected by the hardened cuticle of a dead female, are either time and labor intensive or lack robustness. We present a robust and cost-efficient viability assay based on loss of membrane integrity upon death. This assay uses trehalose, a disaccharide present at a high concentration in the perivitelline fluid of PCN eggs, as a viability marker. Although this assay can detect a single viable egg, the limit of detection for regular field samples was higher, ≈10 viable eggs, due to background signals produced by other soil components. On the basis of 30 nonviable PCN samples from The Netherlands, a threshold level was defined (ΔA(trehalose) = 0.0094) below which the presence of >10 viable eggs is highly unlikely (true for ≈99.7% of the observations). This assay can easily be combined with a subsequent DNA-based species determination. The presence of trehalose is a general phenomenon among cyst nematodes; therefore, this method can probably be used for (for example) soybean, sugar beet, and cereal cyst nematodes as well.  相似文献

Bacterial soft rot caused by Erwiniacarotovora subsp. carotovora is amajor disease in Zantedeschia spp.,particularly in cultivars from the sectionAestivae. The disease can partiallybe controlled by cultivation measures, so acombination with resistant cultivars couldeffectively protect the crop. However,resistant commercial Aestivaecultivars are not available yet. By meansof a recently developed non-destructiveresistance test, variation inaggressiveness was observed among fiveisolates of Erwinia carotovora subsp.carotovora without interactionsbetween the isolates and three Zantedeschia accessions. Within elevenaccessions of Z. aethiopica,variation was observed from almost completeto moderate resistance, while theZ. odorata accession was susceptible.All 21 Aestivae cultivars weresusceptible. Within the Aestivaespecies, Z. elliotiana and Z.pentlandii were also susceptible, butwithin twelve accessions of Z.albomaculata, as well as in six accessionsof Z. rehmannii, variation was foundfrom susceptible to moderately resistant.Hence, new sources of resistance wereidentified that show good potentials forresistance breeding.  相似文献

Knowledge of the effects of species diversity within taxonomic groups on nutrient cycling is important for understanding the role of soil biota in sustainable agriculture. We hypothesized that earthworm species specifically affect nitrogen mineralization, characteristically for their ecological group classifications, and that earthworm species interactions would affect mineralization through competition and facilitation effects. A mesocosm experiment was conducted to investigate the effect of three earthworm species, representative of different ecological groups (epigeic: Lumbricus rubellus; endogeic: Aporrectodea caliginosa tuberculata; and anecic: Lumbricus terrestris), and their interactions on the bacterial community, and on nitrogen mineralization from 15N-labelled crop residue and from soil organic matter.Our results indicate that L. rubellus and L. terrestris enhanced mineralization of the applied crop residue whereas A. caliginosa had no effect. On the other hand, L. rubellus and A. caliginosa enhanced mineralization of the soil organic matter, whereas L. terrestris had no effect. The interactions between different earthworm species affected the bacterial community and the net mineralization of soil organic matter. The two-species interactions between L. rubellus and A. caliginosa, and L. rubellus and L. terrestris, resulted in reduced mineral N concentrations derived from soil organic matter, probably through increased immobilization in the bacterial biomass. In contrast, the interaction between A. caliginosa and L. terrestris resulted in increased bacterial growth rate and reduced total soil C. When all three species were combined, the interaction between A. caliginosa and L. terrestris was dominant. We conclude that the effects of earthworms on nitrogen mineralization depend on the ecological traits of the earthworm species present, and can be modified by species interactions. Knowledge of these effects can be made useful in the prevention of nutrient losses and increased soil fertility in agricultural systems, that typically have a low earthworm diversity.  相似文献

Quantity and quality of soil organic matter (SOM) affect physical, chemical, and biological soil properties, and are pivotal to productive and healthy grasslands. Thus, we analyzed the distribution of soil aggregates and assessed quality, quantity, and distribution of SOM in two unimproved and improved (two organic and two conventional) grasslands in subarctic Iceland, in Haplic and Histic Andosols. We also evaluated principal physicochemical and biological soil properties, which influence soil aggregation and SOM dynamics. Macroaggregates (>250 µm) in topsoils were most prominent in unimproved (62–77%) and organically (58–69%) managed sites, whereas 20–250 µm aggregates were the most prominent in conventionally managed sites (51–53%). Macroaggregate stability in topsoils, measured as mean weight diameter, was approximately twice as high in organically managed (12–20 mm) compared with the conventionally managed (5–8 mm) sites, possibly due to higher organic inputs (e.g., manure, compost, and cattle urine). In unimproved grasslands and one organic site, macroaggregates contributed between 40% and 70% of soil organic carbon (SOC) and nitrogen to bulk soil, whereas in high SOM concentration sites free particulate organic matter contributed up to 70% of the SOC and nitrogen to bulk soil. Aggregate hierarchy in Haplic Andosols was confirmed by different stabilizing mechanisms of micro- and macroaggregates, however, somewhat diminished by oxides (pyrophosphate-, oxalate-, and dithionite-extractable Fe, Al, and Mn) acting as binding agents for macroaggregates. In Histic Andosols, no aggregate hierarchy was observed. The higher macroaggregate stability in organic farming practice compared with conventional farming is of interest due to the importance of macroaggregates in protecting SOM and soils from erosion, which is a prerequisite for soil functions in grasslands that are envisaged for food production in the future.  相似文献

Results are presented of a zero‐discharge marine recirculating system used for the culture of gilthead seabream Sparus aurata. Operation of the system without any discharge of water and sludge was enabled by recirculation of effluent water through two separate treatment loops, an aerobic trickling filter and a predominantly anoxic sedimentation basin, followed by a fluidized bed reactor. The fish basin was stocked for the first 6 mo with red tilapia Oreochromis niloticus × O. aureus at an initial density of 16 kg/m3. During this period salinity was raised from 0 to 20 parts per thousand. Then, gilthead seabream, stocked at an initial density of 21 kg/m3, replaced tilapia at day 167 and were cultured for an additional 225 d. Non steady‐state inorganic nitrogen transformations occurred as a result of these salinity changes. After day 210, the system operated at all times with those water quality parameters considered critical for successful operation of mariculture systems, within acceptable limits. Thus ammonia, nitrite, and nitrate concentrations did not exceed 1.0‐mg total ammonia‐N/ L, 0.5‐mg NO2:‐N/L and 50‐mg NO3‐N/L, respectively. Sulfide levels in the fish basin were below detection limits and oxygen > 6 mg/L after the oxygen generator was added at day 315. Ammonia, produced in the fish basin and to a lesser extent in the sedimentation basin, was converted to nitrate in the aerobic trickling filter. Nitrate removal took place in the sedimentation basin and to a lesser extent in the fluidized bed reactor. Sludge, remaining in the sedimentation basin at the end of the experimental period, accounted for 9.2% of the total feed dry matter addition to the system. The system was disease‐free for the entire year and fish at harvest were of good quality. Water consumption for production of 1 kg of tilapia was 93 L and 214 L for production of 1 kg of gilthead seabream. Additional growth performance data of gilthead seabream cultured in a similar but larger system are presented. During 164 d of operation of the latter system, maximum stocking densities reached 50 kgl M3 and fish biomass production was 27.7 kg/m3. Relatively poor fish survival and growth resulted from occasional technical failures of this pilot system.  相似文献
