Phytoene synthase (PSY) and phytoene desaturase (PDS), which catalyze the first and second steps of the carotenoid biosynthetic pathway, respectively, are key enzymes for the accumulation of carotenoids in many plants. We isolated 2 partial cDNAs encoding PSY (AsPSY-1 and AsPSY-2) and a partial cDNA encoding PDS (AsPDS) from Allium sativum. They shared high sequence identity and conserved motifs with other orthologous genes. Quantitative real-time PCR analysis was used to determine the expression levels of AsPSY1, AsPSY2, and AsPDS in the bulbils, scapes, leaves, stems, bulbs, and roots of garlic. High-performance liquid chromatography demonstrated that carotenoids were not biosynthesized in the underground organs (roots and bulbs), but were very abundant in the photosynthetic organs (leaves) of A. sativum. A significantly higher amount of β-carotene (73.44 μg·g(-1)) was detected in the leaves of A. sativum than in the other organs.  相似文献

Genetically matched pluripotent embryonic stem (ES) cells generated via nuclear transfer or parthenogenesis (pES cells) are a potential source of histocompatible cells and tissues for transplantation. After parthenogenetic activation of murine oocytes and interruption of meiosis I or II, we isolated and genotyped pES cells and characterized those that carried the full complement of major histocompatibility complex (MHC) antigens of the oocyte donor. Differentiated tissues from these pES cells engrafted in immunocompetent MHC-matched mouse recipients, demonstrating that selected pES cells can serve as a source of histocompatible tissues for transplantation.  相似文献

Rice seed was treated by high proton beam irradiation up to 1,200 Gy in order to elucidate physicochemical properties of irradiated rice starch. The decrease of amylose content in two rice cultivars ranged 3.5 to 3.6% compared to the non-irradiated rice under the highest proton beam irradiation (1,200 Gy). Gel consistency in japonica rice cultivar Ilpum was significantly increased with increasing proton beam irradiation. Its values approached 9.7 in the Ilpum rice cultivar and 8.7 cm in the Hanmaum rice cultivar. The peak viscosity, hot peak viscosity, cool peak viscosity, and setback were significantly decreased with increasing proton beam dose. High proton beam irradiation caused a significant decrease in the onset temperature (T o ), peak temperature (T p ), conclusion temperature (T c ), and enthalpy change (ΔH). Gelatinization range (R) in two rice cultivars was more broaden in higher proton beam irradiation than in low proton beam irradiation. The crystallinity degree of two rice starches irradiated with high proton beam was significantly increased and it ranged from 3.25 and 4.39% compared to the non-irradiated rice starches. It might be deduced that proton beam irradiation causes changes of starch, especially at high irradiation of proton beam.  相似文献

Inhibition of acyl CoA:diacylglycerol acyltransferase (DGAT), which is a key enzyme in triglyceride synthesis in eukaryotic organisms, has been proposed as one of the drug targets for treating obesity, type II diabetes mellitus, and metabolic syndrome. Bioassay-guided fractionation of EtOH extract of the flower buds of Tussilago farfara , using an in vitro DGAT enzyme assay, resulted in the isolation of four known sesquiterpenoids, tussilagonone (1), tussilagone (2), 7beta-(3-ethyl-cis-crotonoyloxy)-1alpha-(2-methylbutyryloxy)-3,14-dehydro-Z-notonipetranone (3), and 8-angeloylxy-3,4-epoxy-bisabola-7(14),10-dien-2-one (4). DGAT1 inhibitory activity was studied by in vitro DGAT assay using rat liver microsomes and HepG2 cell microsomes. They showed DGAT1 inhibition with IC(50) values of 99.2 (1), 18.8 (2), 47.0 (3), and 211.1 (4) microM (for rat liver microsomes) and >1 mM (1), 49.1 (2), 160.7 (3), and 294.4 (4) microM (for HepG2 cell microsomes), respectively. Compound 2 showed the most potent inhibition against microsomal DGAT1 derived from rat liver and human hepatocellular carcinoma HepG2 cells and also significantly inhibited triglyceride synthesis by suppressing incorporation of [(14)C]acetate or [(14)C]glycerol into triglycerides in HepG2 cells. These findings suggest that tussilagone is a potential lead compound in the treatment of obesity and type 2 diabetes.  相似文献

Allyl isothiocyanate (AITC) and phenethyl isothiocyanate (PEITC) were isolated from Sinapis alba L. seeds and their effects against Dermatophagoides farinae and D. pteronyssinus were evaluated using the impregnated fabric disk method. The LD 50 values of their compounds and derivatives were then compared with those of a commercial acaricide, benzyl benzoate. On the basis of the LD 50 values against D. farinae, PEITC (0.21 microg/cm(2)) was the most toxic, followed by benzyl isothiocyanate (0.55 microg/cm(2)), phenyl isothiocyanate (1.09 microg/cm(2)), butyl isothiocyanate (1.24 microg/cm(2)), and AITC (1.36 microg/cm(2)); acetyl isothiocyanate (195.01 microg/cm(2)) was the least toxic. In addition, the acaricidal effects of AITC and PEITC against D. farinae were 7.4- and 47.8-fold greater than those of benzyl benzoate, respectively. Against D. pteronyssinus, PEITC was the most toxic (0.19 microg/cm(2)), followed by benzyl isothiocyanate (0.77 microg/cm(2)), phenyl isothiocyanate (1.37 microg/cm(2)), butyl isothiocyanate (1.50 microg/cm(2)), and AITC (2.88 microg/cm(2)); acetyl isothiocyanate (168.82 microg/cm(2)) was the least toxic. AITC and PEITC were 3.3- and 50.4-fold more active than benzyl benzoate against D. pteronyssinus, respectively. Taken together, these findings indicate that AITC, PEITC, and partial derivatives may be useful as preventive agents against dust mites. In addition, these results indicate that structure-activity is related to the aromatic structure, the number of carbon atoms, and the compounds hydrophobicity.  相似文献

Knowledge about the ecology of urban water systems is very scarce. We assessed the conservation value of urban drainage systems in lowland areas and compared these with similar watercourses in rural areas. A total of 36 water bodies in urban areas were selected to investigate the macroinvertebrate biodiversity in relation to environmental variables. Multivariate analysis of aquatic macroinvertebrate assemblages was used to distinguish urban water types and to link these types to key environmental variables. Several biodiversity indices for urban water systems were compared with those for other drainage systems in The Netherlands. Four types of macroinvertebrate assemblages were distinguished in the urban water systems, differing in environmental conditions and values of ecological indicators. The variation in macroinvertebrate assemblages was significantly explained by nitrate, pH, grain size (sediment composition), transparency, nymphaeid and submerged vegetation. Urban drainage systems can sustain a macroinvertebrate biodiversity comparable to that of drainage systems in rural areas (ditches and canals) and (semi)natural watercourses (lotic waters such as small streams and rivulets) and can even be a habitat for red list species. To optimize biodiversity values, urban water management should aim at lowering nutrient levels, stimulating vegetation (diversity of habitat structure) and increasing transparency, which are key factors for macroinvertebrate diversity. We show the potential conservation benefits of water systems in urban areas, but further studies are needed to investigate the optimal design of cities to include biodiversity as an integrated part of the urban environment, thereby sustaining a higher biodiversity in an increasingly urbanizing world.  相似文献

Piceid is widely used in food, cosmetics, and pharmaceuticals because of its therapeutic benefits. However, the use of piceid as a drug is limited because of its low solubility. To increase solubility, we synthesized piceid glucosides using maltosyltransferase from Caldicellulosiruptor bescii . The MTase gene was cloned and expressed in Escherichia coli . The enzyme had a unique transfer specificity to the transfer of maltosyl units. Four piceid transglycosylation products were present and identified by thin-layer chromatography and recycling preparative high-performance liquid chromatography. The major product was purified by C(18) and gel filtration chromatography, and its molecular structure was determined using nuclear magnetic resonance spectroscopy to be α-d-maltosyl-(1→4)-piceid. The solubility of maltosyl piceid was 8.54 × 10(3) and 1.86 × 10(3) times those of natural resveratrol and piceid, respectively, suggesting that the transglycosylation greatly increased the water solubility. This suggests that dietary intake of this compound can enhance the bioavailability of resveratrol in the human body.  相似文献
