A novel staining procedure has been developed to visualize the gluten– starch matrix in wheat flour dough. Dough samples mixed to the final stage were stained with 26 fluorescent reagents, and each stained sample was observed with three sets of fluorescence filters (blue, green, and red). Of all the combinations of reagents and filters, the combination of acid magenta and the blue fluorescent filter set was the most effective in distinguishing starch granules from gluten network structure. Its effectiveness was further demonstrated with gluten and starch granule samples, in which the contrast was clearer when observed with the blue fluorescent filter set than without any fluorescent filter. Visualizing the gluten–starch matrix in dough samples at four mixing stages with the same procedure resulted in clear identification of the changes in gluten network structure because of the differences in mixing stages. The same procedure also enabled us to distinguish starch from gluten in white salted noodles, baked cookies, and flour particles. The proposed procedure is quicker, simpler, and has a lower risk of altering the sample than other conventional ones, and it is expected to become a useful tool in cereal studies.  相似文献

A gas detection apparatus equipped with a semiconductor gas sensor was employed for qualitative and quantitative measurement of hydrogen and methane emitted by termites. A gas sample of 2.5 ml was injected into the semiconductor gas sensor through the gas detection apparatus, and the maximum voltage of the sensor was converted into gas concentration. The gas samples were collected from three distinct experiments: (1) five combinations of workers and soldiers of Coptotermes formosanus with and without a wood specimen; (2) C. formosanus under six temperature conditions; and (3) four different termite species, C. formosanus, Reticulitermes speratus, Incisitermes minor, and Zootermopsis nevadensis. The hydrogen and methane concentrations increased with an increase in the number of termites. Concentrations were higher in samples with a wood specimen than without it. Both hydrogen and methane concentrations were the highest for the samples at 35 °C and were lowest at 15 and 5 °C. The concentrations were very low at 45 °C because all the termites had died in a few hours. The concentrations of hydrogen and methane were highest for Z. nevadensis, the dampwood termite, among the four species, and no methane was detected for I. minor, the drywood termite, at 28 °C and 75 % RH.  相似文献

Summary

To control the bolting of Japanese bunching onion (Allium fistulosum L.) photoperiodically, the effect of photoperiods before, during and after vernalization on flower initiation and development and the varietal differences were investigated using the two mid-season flowering cvs Kincho and Asagi-kujo, and a late-season flowering cv. Cho-etsu. A long-day photoperiod (LD, 16 h) given before vernalization inhibited flower initiation. Especially, the bolting rate of ‘Asagi-kujo’ decreased by about a half, compared with the short-day photoperiod (SD, 8 h). The interaction between the effect of night temperature (3°C, 7°C, 11°C or 15°C) and the effect of the photoperiod (SD and LD) during vernalization was also investigated. In ‘Kincho’, LD did not affect flower initiation at 3°C, but inhibited flower initiation at 7°C, 11°C and 15°C. In ‘Asagi-kujo’, flower initiation was significantly inhibited by LD under all temperature conditions. This inhibitory effect was stronger at 11°C and 15°C than at 3°C and 7°C. In ‘Cho- etsu’, LD significantly inhibited flower initiation at 3°C and 7°C, and flower initiation rarely occurred at 11°C and 15°C. In this study, generally, LD during vernalization inhibited flower initiation in all cultivars. Thus Japanese bunching onion required a short-day photoperiod in flower initiation, which was stronger in ‘Asagi-kujo’ and ‘Cho-etsu’ than in ‘Kincho’. From these results, we conclude that low temperature and a short-day photoperiod complementarily induce flower initiation in Japanese bunching onion. Varietal differences exist in the requirement of low temperature and a short-day photoperiod: the primary requirement in ‘Kincho’ is low temperature and that in ‘Asagi-kujo’ is a short-day. After flower initiation, the early stage of flower development is day-neutral, and after the floret formation stage, a long-day photoperiod promotes flower development and elongation of the seedstalk.  相似文献

Mazus pumilus is an annual self‐pollinating weed that is commonly found in arable land, vegetable gardens and roadsides. This weed harbours insects and pathogens that attack vegetables. The mechanism of resistance to paraquat of M. pumilus found in Ohita, Japan, was studied. Whole plant bioassays revealed that the resistant (R) biotypes were four to six times less susceptible than controls. Chlorophyll destruction of leaf discs by paraquat treatment in R biotypes was 4–20 times lower than those of susceptible (S) biotypes. Ferric reducing antioxidant power (FRAP) values in R biotypes were higher than those of S biotypes before and after paraquat treatments. The activity of superoxide dismutase (SOD) was also higher in R biotypes than those of S biotypes before and after treatment with paraquat, but the activities of ascorbate peroxidase (APX) and catalase (CAT) were not different between R and S biotypes. Change of ascorbate (AsA) contents before and after paraquat treatment was equivalent in both biotypes. These results indicate that the increased SOD activity and antioxidant capacity in R biotypes contribute to the resistance to paraquat of M. pumilus.  相似文献

A colorimetric assay used to quantify the non-ionic contrast medium iodixanol in sera was validated and compared with high-performance liquid chromatography. The application of this assay to estimate glomerular filtration rates (GFR) in cattle was examined. Serum iodixanol was de-iodinated by alkaline hydrolysis and the amount of released iodine was subsequently determined using a ceric arsenite method. There was a close correlation between the two methods using identical specimens. In clinically healthy cattle with different body weights, the reference value (166.3-178.8 mL/min/m(2)) based on body surface area was fairly stable as compared with that (2.13-3.63 mL/min/kg) based on body weight. Based on GFR data in healthy and renal-impaired cattle, when the GFR decreased to more than 60% of the reference value, serum urea nitrogen and creatinine concentrations increased. The colorimetric assay is a simple method for the estimation of GFR in cattle and requires no expensive equipment.  相似文献

Remote sensing offers the potential to spatially map forest cover quickly and reliably for inventory purposes. We developed a new image analysis approach using an integrated methodology of “object-based” image classification techniques and field-based measurements to quantify forest cover in a degraded dry forest ecosystem on the leeward side of the Island of Hawaii. This new approach explicitly recognized the transitional areas between tree crowns and tree shades (tree shadows) as a unique class and fully utilized them for the quantification of canopy cover. Object-oriented classification of Ikonos-2 satellite images allowed delineation of tree shades and crowns and the transitional areas between them from objects with similar reflectance and size that were surrounding the trees. These included patches of fountain (Pennisetum setaceum) and kikuyu (Pennisetum clandestinum) grass, lava outcrops and lava–grass mixtures. Crown-shade transitions were clearly differentiated in spite of their wide range of spectral values and reflectance similarities with areas of lava–grass mixture. Segments representing tree shades and dark lava outcrops were also classified into their respective classes even if they were contiguous. The image estimates of canopy cover using the tree shade plus transition classes were linearly related with field estimates of canopy cover (R2 = 0.86 and slope = 0.976). Based on this relationship, dry forest cover throughout the 2627-ha area was estimated at 7.7 ± 1.9%. An immediate application of this new approach is to select and delineate areas with higher canopy cover in order to concentrate ecological restoration and conservation efforts.  相似文献

In the region contaminated by the Fukushima nuclear accident, radioactive contamination of live cattle should be checked before slaughter. In this study, we establish a precise method for estimating radioactive cesium concentrations in cattle blood using urine samples. Blood and urine samples were collected from a total of 71 cattle on two farms in the ‘difficult‐to‐return zone’. Urine 137Cs, specific gravity, electrical conductivity, pH , sodium, potassium, calcium, and creatinine were measured and various estimation methods for blood 137Cs were tested. The average error rate of the estimation was 54.2% without correction. Correcting for urine creatinine, specific gravity, electrical conductivity, or potassium improved the precision of the estimation. Correcting for specific gravity using the following formula gave the most precise estimate (average error rate = 16.9%): [blood 137Cs] = [urinary 137Cs]/([specific gravity] ? 1)/329. Urine samples are faster to measure than blood samples because urine can be obtained in larger quantities and has a higher 137Cs concentration than blood. These advantages of urine and the estimation precision demonstrated in our study, indicate that estimation of blood 137Cs using urine samples is a practical means of monitoring radioactive contamination in live cattle.  相似文献

One aim of conservation tillage is to preserve soil biological properties. This study was conducted to examine the effects of two contrasting tillage treatments on soil biota at different depths. We investigated the population dynamics and vertical distributions of microbes and several soil faunal groups for 2 years in field Andosols in northeastern Japan. The experimental plots were under no tillage (NT) or conventional tillage (CT, rotary tilled to 20 cm) management. In the 0–10-cm soil layer, bacterial and fungal substrate-induced respiration (SIR) and the population density of enchytraeids were higher under NT than under CT, but the population densities of protozoa, mites, and collembolans did not differ significantly. In contrast, at 10–20 cm, both SIR values were higher under CT, where larger populations of mites and collembolans were recorded. At both depths, nematodes were more abundant under CT. Thus, the effects of tillage on these soil organisms differed according to soil depth, and negative impacts of tillage were smaller in the deeper layer. Larger amounts of earthworm casts at the soil surface in NT plots showed a greater biomass of earthworms than in CT. To evaluate the activities of soil biota, we buried litterbags with three different mesh sizes at the two depths and examined the rate of decomposition. The daily decay constant of litter in the surface soil layer (1.5–8.5 cm) was greater under NT. We suppose that the activities of soil biota in this layer were stimulated under NT, and that especially microbes and enchytraeids, which were abundant at 0–10 cm, contributed greatly to the decomposition.  相似文献

A homoeologous quantitative trait locus to that of eps5L on barley chromosome 5H was identified in a syntenic region of wheat chromosome 5A. Wheat single chromosome recombinant lines (SCRs) were developed from a cross between ‘Chinese Spring’(‘Cappelle-Desprez’ 5A) and ‘Chinese Spring’(Triticum spelta 5A), these were grown together with the parental controls under different vernalization and photoperiod regimes. The variation for ear emergence time accelerated heading induced by the T. spelta segment indicated an effect associated with the Xcdo412-Xbcd9 interval. Since no differences between the SCRs and controls in responses to vernalization and photoperiod treatments were detected, this effect was identified as an earliness per se gene, Q Eetocs-5 A.2, which may be homoeologous to the eps5L quantitative trait locus of barley. Xbcd926 has been found to be closely linked to the rice flowering time quantitative trait loci, QHd9a or FLTQ2, on chromosome 9, suggesting possible relationships among the quantitative trait loci across wheat, barley and rice genomes.  相似文献
