Although reduced tillage (RT) may preserve soil biota and improve the productivity and sustainability of arable lands in temperate regions, the extension of RT is limited by difficulties in controlling weeds. We studied the effect of RT without herbicide application on weed communities and soil biota in a 1-year 2-crop rotation system with winter wheat (Triticum aestivum L.) and maize (Zea mays L.) on Andosols in Japan. RT of the surface 3 cm and conventional moldboard plowing (CT) were conducted before seeding twice per year. For the first 3 years, from autumn 1997 to spring 2000, one field was managed with RT and another with CT. For the second 3 years, from autumn 2000 to spring 2003, RT and CT were conducted in two replicated plots in each field. Weed communities and soil biota were studied in the last 2 years. Dominant weed species in winter wheat cropping were Italian ryegrass (Lolium multiflorum Lam.) in 2002 and common vetch (Vicia angustifolia L.) in 2003, and their biomass was high where RT or CT was continuously conducted. Switching of tillage methods, from RT to CT or vice versa, reduced the biomass of winter weeds. In summer maize cropping, several annual and perennial weed species tended to increase under RT in the second 3 years. However, redroot pigweed (Amaranthus retroflexus L.), the most dominant weed in 2002 and 2003, responded to tillage inconsistently and its biomass was not always increased by RT. Species diversity of winter weeds was decreased by CT conducted in the first 3 years, and that of summer weeds was decreased by CT conducted in the second 3 years. The seedbank in the 0–10-cm soil layer under recent RT was large (7200–16 300 seeds m−2) compared with that under CT (2900–7300 seeds m−2). The microbial substrate-induced respiration (SIR) and the population densities of nematodes and mites were higher under RT in the second 3 years and were not affected by previous tillage practices. Both were highly correlated with soil total nitrogen. The positive effect of RT on these soil organisms was primarily attributable to the accumulation of organic matter in soil, but not to plant cover as a result of incomplete weed control by RT. Occasional adoption of RT in current CT systems may be effective at enriching soil organisms with little risk of weed infestation.  相似文献

Eleven, 11 and 2 of 11 precolostral sera from normal calves and 13, 14 and 9 of 14 sera from normal fetuses, 7 to 10 months of gestation, neutralized bovine parvovirus, coronavirus and rotavirus, respectively.When assayed by single radial immunodiffusion, all the sera contained IgG at 360 to 1400 mg/dl, and some of them had much smaller amounts of IgM or IgA. Most of the neutralizing activities against bovine coronavirus and rotavirus were readily inactivated by treatment with acetone or 2-mercaptoethanol. Some sera fractionated by Sephadex G-200 gel filtration or starch block electrophoresis had neutralizing activities against bovine parvovirus or coronavirus in fractions containing no detectable amounts of immunoglobulins. These observations seem to indicate the presence of substance(s), other than immunoglobulins, capable of inhibiting replication of bovine parvovirus, coronavirus or rotavirus. The chemical nature and the mode of action of the inhibitors await elucidation.  相似文献

Expression and purification of recombinant swine interleukin-4   总被引:4,自引:0,他引:4  
The swine interleukin-4 (SwIL-4) cDNA was cloned by RT-PCR. It was expressed using an expression vector pQE30 in E. coli, a baculovirus AcNPV vector pVL1392 in insect cells, and a pCAGGS vector in mammalian cells. The rSwIL-4 proteins expressed from bacteria and insect cells were purified using a chelating affinity column and a mAb-coupled immunoaffinity column. The amount of the products and their bioactivities were compared. All recombinant cytokines were efficiently reacted with the specific antibodies and the molecular weight of rSwIL-4 was approximately 16 kDa in E. coli, 15 and 18 kDa in insect cells, and 15 and 20 kDa in mammalian cells. Variations of molecular weight observed in insect and mammalian cells were probably due to different modification ways of glycosylation. All these recombinant proteins retained their antigenicity and were biologically active in inducing human TF-1 cell proliferation in vitro. The simple purification method will make it possible to evaluate the in vitro and in vivo effects of IL-4 in pigs.  相似文献

Measles virus (MV) infection primarily targets epithelial cells of the respiratory tract, which have the potential to synthesize a variety of cytokines. In this report, we studied the effect of MV infection on the production of interleukin (IL)-8 by the pulmonary epithelial cells. A549 cells, a lower airway epithelial cell line, produced IL-8 after MV inoculation in a dose- and time-dependent manner. The IL-8 production was little affected by UV-inactivation of MV and scarcely suppressed by cycloheximide treatment. These results indicated that MV particle binding to and/or incorporation into cells stimulated IL-8 expression in A549 cells.  相似文献

A coronavirus (Kakegawa isolate) isolated from a cow with epizootic diarrhea was grown in BEK-1 cells and examined for biophysical and biochemical properties. The Kakegawa isolate was able to replicate in the presence or absence of 5-iodo-2′-deoxy-uridine, indicating that its viral nucleic acid was RNA. It was highly sensitive to ether and chloroform, and moderately sensitive to trypsin and heat. It was, however, readily stabilized by treatment with cation at 50°C for 1 h. Its infectivity was slightly reduced at pH 3.0. The virus passed through a membrane filter of 200 nm pore size, but not through one of 100 nm pore size. The buoyant density of the virus was determined in a sucrose density gradient. The peak of infectivity and hemagglutinin activity was found at a density of 1.182. Neutralization and hemagglutination inhibition tests showed a close serological relationship between the Kakegawa isolate and the American strain of calf diarrhea coronavirus.  相似文献

The prevalence of dental disorders in dogs was studied by applying index systems for human with some modifications. A total of 251 mongrel dogs including 143 stray dogs kept in the Animal Protection Offices in Tokyo and Hokkaido and 108 pet dogs visiting veterinary clinicians in Chiba Prefecture and Hokkaido were used. Periodontitis was prevalent among these dogs regardless of their sources and its incidence was increased with age. The lesion was more severe and more frequent in the premolar and molar regions than in the maxillary and mandibular incisor regions. Missing of teeth was observed at a high and increasing incidence with age. The tooth most commonly lost was the first premolar, followed by the other premolars and molars, where severe periodontitis was frequently found. Calculus was seen on many teeth, and aging agravated its prevalence and severity. Dental caries was observed in stray dogs, but neither to a serious degree nor at a significant level. These findings emphasize the necessity of dental hygiene, proper dental care and continuous periodical survey for dogs.  相似文献

The incidence of DIC in 208 dogs with a malignant tumor was evaluated. The incidence of DIC was 9.6% in dogs with a malignant tumor which was a solid tumor in all. In 164 dogs with a malignant solid tumor, the incidence of DIC was 12.2%. The incidence of DIC in dogs with hemangiosarcoma, mammary gland carcinoma and adenocarcinoma of the lung was significantly higher than that in dogs with other malignant tumors. These results suggested that special care in looking for DIC should be taken in dogs with a malignant solid tumor.  相似文献
