During the last few decades, the physiological and genetic background of dormancy, and correlated pre-harvest sprouting (PHS) have been intensively investigated. Special attention has often been paid to genetic factors that may explain and predict PHS susceptible behaviour. A major candidate is the Vp1 gene which is involved in embryo development and maturation as well as in dormancy establishment. In this study, Vp1 gene expression during kernel development was studied in wheat, triticale and rye as a potential biomarker for selecting PHS tolerant varieties in cereal breeding programs. Plants of known PHS tolerant and PHS susceptible varieties were grown under controlled conditions from flowering until harvest ripeness. During that period, kernels were regularly harvested for RNA extraction and cDNA synthesis. Calibrated and normalized relative Vp1 expression levels were obtained in an RT-qPCR assay. During kernel development, Vp1 expression levels generally showed a typical peak during the soft dough stage, after which they decreased and remained low until harvest maturity. Differences in Vp1 expression levels could be observed between the PHS susceptible and PHS tolerant varieties of wheat, with the PHS tolerant variety showing higher levels of relative Vp1 expression compared to the PHS susceptible variety. In triticale, however, this difference was only seen once and could not be confirmed in further experiments. It seems that the Vp1 gene in triticale behaves in a similar way as in rye, in which no specific trends could be observed.  相似文献

BACKGROUND: Saponins are a class of secondary plant metabolites consisting of a sugar moiety glycosidically linked to a hydrophobic aglycone (sapogenin) that often possess insecticidal activities. Four saponins were selected: two triterpene saponins, Q. saponaria saponins and aescin, and two steroidal saponins, digitonin and diosgenin. Their effects were investigated on an important pest species and a model piercing‐sucking insect, the pea aphid Acyrthosiphon pisum. The triterpene Q. saponaria saponins bark saponin received special attention because of its high activity. Aphids were challenged by oral and contact exposure to demonstrate aphicidal activities, and in choice experiments to support use as a natural deterrent. RESULTS: When aphids were exposed to supplemented artificial diet for 3 days, a strong aphicidal activity was recorded for three of the four saponins, with an LC50 of 0.55 mg mL?1 for Q. saponaria saponins, 0.62 mg mL?1 for aescin and 0.45 mg mL?1 for digitonin. The LT50 values ranged between 1 and 4 days, depending on the dose. For diosgenin, only low toxicity (14%) was scored for concentrations up to 5 mg mL?1. In choice experiments with treated diet, a deterrence index of 0.97 was scored for Q. saponaria saponins at 1 mg mL?1. In contrast, direct contact showed no repellent effect. Spraying of faba bean plants with Q. saponaria saponins resulted in an LC50 of 8.2 mg mL?1. Finally, histological analysis in aphids fed with Q. saponaria saponins demonstrated strong aberrations of the aphid gut epithelium, and exposure of midgut CF‐203 cell lines to Q. saponaria saponins in vitro confirmed the cytotoxic effect. CONCLUSIONS: The present insect experiments provide strong evidence that saponins, as tested here with triterpene Q. saponaria saponins, can be useful as natural aphicides and deterrents. The orientation and distribution of reinforcing particles in artificial composites are key to enable effective reinforcement of the material in mechanically loaded directions, but remain poor if compared to the distinctive architectures present in natural structural composites such as teeth, bone, and seashells. We show that micrometer-sized reinforcing particles coated with minimal concentrations of superparamagnetic nanoparticles (0.01 to 1 volume percent) can be controlled by using ultralow magnetic fields (1 to 10 milliteslas) to produce synthetic composites with tuned three-dimensional orientation and distribution of reinforcements. A variety of structures can be achieved with this simple method, leading to composites with tailored local reinforcement, wear resistance, and shape memory effects.  相似文献

A 4-month-old dog was presented with a progressive swelling of the submandibular area. The history, course, cytological, and sialographic findings were consistent with an aseptic pyogranulomatous sialadenitis with concurrent duct blockage. This rare entity, responsive to medical treatment, appears to be similar to the granulomatous giant cell sialadenitis of humans.  相似文献

Tryptophan (Trp) as a precursor of serotonin (5-hydroxytryptamine, 5-HT) has long been used to extenuate aggressive behavior and control stress of humans as well as several farm animals. This study was conducted to determine the effect of supplemental L-Tryptophan (L-Trp) on growth, cerebral 5-HT concentration, stress hormone concentration, oxidative stress status, and behavior response of pigs under social stress, and also to determine an optimal daily total Trp intake that would benefit nursery pigs under social stress. Seventy two individually housed barrows at 6 wk of age were randomly allotted to 6 treatments with supplementation of 0, 2, 4, 6, 8, or 10 g L-Trp/kg to corn and soybean meal-based feedstuffs. Pigs were fed assigned feedstuffs for 15 d. Body weight was measured on d 0, 5, 10, and 15. Saliva and blood were collected on d 5, 10, and 15. On d 5 and 10, all 12 pigs in each treatment were paired in 6 new pens to record behavior for a 2-d period and returned to original individual pens. On d 15, pigs were euthanized to obtain hypothalamus. During the first 5 d, ADG and G:F increased (linear, P < 0.01) with increasing supplemental L-Trp. During the entire 15 d, ADG and G:F increased (linear, P = 0.01 and P < 0.01, respectively) with increasing supplemental L-Trp. Estimates of the daily total Trp intake based on ADG on d 15 were 10.8 g/d (P < 0.01; R(2) = 0.16) using a 1-slope broken-line analysis. Hypothalamic 5-HT and 5-hydroxyindoleacetic acid increased (linear, P < 0.01 and P = 0.03, respectively) with increasing supplemental L-Trp. Malonedialdehyde in plasma and hypothalamus, as well as salivary cortisol, on d 15 decreased (linear, P = 0.01, P < 0.01, and P < 0.01, respectively) with increasing supplemental L-Trp. Plasma urea nitrogen decreased (linear, P < 0.01) with increasing supplemental L-Trp. Increasing supplemental L-Trp affected pig behaviors during the first 2-d observation period by decreasing (quadratic, P = 0.04) lying, decreasing (linear, P = 0.04) sitting, and increasing (linear, P = 0.02) drinking. Overall, supplementation of L-Trp improved growth performance of 6 wk-old nursery pigs under social stress in association with increasing hypothalamic 5-HT production, reducing stress hormone concentrations, decreasing lipid peroxidation, increasing drinking, and reducing sitting and lying. Increase in BW gain of nursery pigs under social stress was maximal when daily total Trp intake was 10.8 g.  相似文献
