This study describes a generic biological screening assay designed to detect anticoagulant rodenticides based on their inhibitory action on the vitamin K epoxide reductase protein complex, resulting in an accumulation of under-carboxylated prothrombin or proteins induced by vitamin K antagonism (PIVKA-II). A combined cell culture/ELISA assay was optimized to measure PIVKA-II production by the human hepatoma HepG2 cell line cultured in the presence of anticoagulant rodenticides. The specificity and sensitivity of the assay was validated using 41 grain extracts containing representative concentrations of rodenticide or appropriate nonrodenticide control compounds. In all cases, PIVKA-II produced by HepG2 cells in response to grain extracts spiked with rodenticides was detected by ELISA, while PIVKA-II was not detected in supernatants collected from cells exposed to nonrodenticide controls. This represents a novel, class-specific biological assay for the detection of anticoagulant rodenticides present in contaminated grain. 相似文献

A 13-million-year continuous record of Oligocene climate from the equatorial Pacific reveals a pronounced "heartbeat" in the global carbon cycle and periodicity of glaciations. This heartbeat consists of 405,000-, 127,000-, and 96,000-year eccentricity cycles and 1.2-million-year obliquity cycles in periodically recurring glacial and carbon cycle events. That climate system response to intricate orbital variations suggests a fundamental interaction of the carbon cycle, solar forcing, and glacial events. Box modeling shows that the interaction of the carbon cycle and solar forcing modulates deep ocean acidity as well as the production and burial of global biomass. The pronounced 405,000-year eccentricity cycle is amplified by the long residence time of carbon in the oceans. 相似文献

Increased cultivar tolerance to nitrogen (N) deficiency may increase productivity and reduce environmental impact of potato crops. In this study we screened 20 cultivars for the tolerance to N-deficient (0.05 mM nitrate) in comparison with N-abundant (7.5 mM nitrate) nitrate supply using plantlets grown for 15 days in a recirculating hydroponic system. Nitrogen deficiency increased the root-to-shoot ratio in 18 cultivars. Plant total dry weight (TDW) was reduced by an average of 61% under N-deficient nitrate supply. Tolerance to N deficiency was assessed as the TDW ratio (TDWR), calculated as the ratio of TDW under N-deficient to N-abundant nitrate supply. The cultivars Norland, Raritan, Nipigon and Langlade were significantly more tolerant to nitrogen deficiency (greater TDWR) than Eramosa, Carleton, and Epicure. The results indicate that the hydroponic system has capacity to rapidly screen a large number of cultivars for tolerance to N deficiency. 相似文献

Deserts exhibit extreme climatic conditions. Small desert‐dwelling vertebrates have physiological and behavioral adaptations to cope with these conditions, including the ability to seek shelter. We investigated the temperature (T) and relative humidity (RH) regulating properties of the soil cracks that characterize the extensive cracking‐clay landscapes of arid Australia, and the extent of their use by 2 small marsupial species: fat‐tailed and stripe‐faced dunnarts (Sminthopsis crassicaudata and Sminthopsis macroura). We measured hourly (over 24‐h periods) the T and RH of randomly‐selected soil cracks compared to outside conditions, during 2 summers and 2 winters. We tracked 17 dunnarts (8 Sminthopsis crassicaudata and 9 Sminthopsis macroura) to quantify their use of cracks. Cracks consistently moderated microclimate, providing more stable conditions than available from non‐crack points, which often displayed comparatively dramatic fluctuations in T and RH. Both dunnart species used crack shelters extensively. Cracks constitute important shelter for small animals during extreme conditions by providing a stable microclimate, which is typically cooler than outside conditions in summer and warmer in winter. Cracks likely play a fundamental sheltering role by sustaining the physiological needs of small mammal populations. Globally, cracking‐clay areas are dominated by agricultural land uses, including livestock grazing. Management of these systems should focus not only on vegetation condition, but also on soil integrity, to maintain shelter resources for ground‐dwelling fauna. 相似文献

Brook trout (Salvelinus fontinalis) are an iconic freshwater salmonid native to northeastern North America. They prefer cold, clean rivers and lakes, and have been culturally, socially and economically valued for decades. Overharvest and deforestation associated with European settlement in southern Ontario, Canada led to notable declines in brook trout abundance in rivers and streams by the late 1800s. Today, they are threatened by further habitat degradation, non-native species, and climate change. In this study, we document brook trout occupancy in rivers and streams of the Mixedwood Plains Ecozone in southern Ontario, between 1970–1980 and 2000–2010. We used generalised linear models to determine occupancy variation between the two time periods at five spatial scales spanning multiple watershed levels. We detected a decline in occupancy between time periods for the tertiary (from 23% to 17%) and quaternary (from 35% to 22%) watershed scales. In the Credit River-Sixteen Mile Creek watershed, mean occupancy decreased to a third of past estimate (from 31% to 10%). For a subset of 86 sites with overlapping sampling in both periods no temporal trend was detected, but a post-hoc analysis indicated low statistical power (0.07), which suggested that more intensive sampling would be needed to better quantify brook trout occupancy changes at this scale. The occupancies quantified for the past period in our study may serve as benchmarks for brook trout restoration efforts. 相似文献
