The skin has an important role in infection by Leishmania chagasi. Apoptosis modulates the inflammatory response acting distinctively either on the progression or regression of the lesions. The parasites interact with multiple regulatory systems inducing apoptosis in host cells, during cell invasion, stabilization and multiplication of pathogens. In this context, the aim of this study was to evaluate cell death within the inflammatory infiltrates, and to correlate these results with parasite load and clinical features of dogs naturally infected with L. chagasi. Fragments of skin pinnas (8 symptomatic+8 asymptomatic+6 negative controls) were used to characterize and measure the inflammatory response, parasite load and apoptosis. Diagnosis of canine leishmaniasis was confirmed by the detection of anti-Leishmania antibodies by IFA and ELISA in serum, direct visualization of the parasite and culture in spleen, liver, pinna, bone marrow and lymph nodes, and PCR (pinna). Histomorphometry was performed with images obtained from 20 representative histological fields in a light microscope. Ultra-thin sections were mounted over a 300 mesh grids, contrasted with 2% uranyl acetate and lead citrate and examined under a Transmission Electronic Microscopy. Amastigotes were only found in the skin of symptomatic animals (31.94±18.81). The number of foci and cellularity of the inflammatory infiltrates in symptomatic dogs were higher than in other groups and in asymptomatics were higher than in controls (p<0.05; Tukey). The average area, perimeter and extreme diameters of the inflammatory infiltrates obtained in symptomatic dogs were higher than in controls (p<0.05; Tukey). The apoptotic index was higher in symptomatic than in other groups and there was no difference between asymptomatics and controls (p<0.05; Tukey). Ultrastructurally, apoptotic cells were shrunken, with condensed nuclear chromatin and cytoplasm. Condensed nuclei were frequently fragmented. Internucleosomal DNA fragmentation occurred only in symptomatic cases. Amastigotes were observed within neutrophils and macrophages. Apoptosis is directly related to parasite load, intensity of inflammatory response and clinical manifestations in L. chagasi naturally infected dogs. 相似文献

The ability to handle animals safely, competently, and with confidence is an essential skill for veterinarians. Poor animal-handling skills are likely to compromise credibility, occupational health and safety, and animal welfare. In the five-year veterinary science degree at Murdoch University, animal handling is taught in a prerequisite unit in the second semester of the second year. From 2008, however, this unit will be taught in the first year of the five-year course. Students are taught to handle sheep, cattle, pigs, and horses safely and competently. Each student receives 30 hours of formal practical instruction. Animal-to-student ratios are 2:1, and staff-to-student ratios vary from 1:8 (sheep, cattle, horses) to 1:17 (pigs). Students must pass the practical exam to proceed into third year. Additional experience with animals is gained during third year (14 hours of practical instruction with sheep, goats, pigs, and cattle) and during the 5 weeks and 2 days of vacation farm experience during the second and third years. In the fourth and fifth years, students consolidate their handling experience with sheep (including rams), goats, pigs, cattle (including bulls), horses (including stallions), and alpacas. As a result, students are able to handle and restrain client animals with confidence. There is no formal course in small-animal handling at Murdoch University. Factors that have enhanced the success of the large-animal handling program include purpose-built on-campus facilities. Inadequate resources (time, facilities, and animals) remain the main impediment to effective learning, further compounded by the increasing tendency of university administrators to make decisions based on economic expediency rather than educational benefit. 相似文献

Context
We address the issue of adapting landscapes for improved insect biodiversity conservation in a changing climate by assessing the importance of additive (main) and synergistic (interaction) effects of land cover and land use with climate.Objectives
We test the hypotheses that ant richness (species and genus), abundance and diversity would vary according to land cover and land use intensity but that these effects would vary according to climate.Methods
We used a 1000 m elevation gradient in eastern Australia (as a proxy for a climate gradient) and sampled ant biodiversity along this gradient from sites with variable land cover and land use.Results
Main effects revealed: higher ant richness (species and genus) and diversity with greater native woody plant canopy cover; and lower species richness with higher cultivation and grazing intensity, bare ground and exotic plant groundcover. Interaction effects revealed: both the positive effects of native plant canopy cover on ant species richness and abundance, and the negative effects of exotic plant groundcover on species richness were greatest at sites with warmer and drier climates.Conclusions
Impacts of climate change on insect biodiversity may be mitigated to some degree through landscape adaptation by increasing woody native vegetation cover and by reducing land use intensity, the cover of exotic vegetation and of bare ground. Evidence of synergistic effects suggests that landscape adaptation may be most effective in areas which are currently warmer and drier, or are projected to become so as a result of climate change.448.

Although somatotropins are potent growth promoters in salmonids, there is little information on how these proteins are metabolized by poikilotherms. In the present study, the plasma uptake and clearance rates of recombinant chicken somatotropin (rcGH) were investigated in juvenile coho salmon (Oncorhynchus kisutch). Two doses of rcGH were administered by intraperitoneal (ip) or intramuscular (im) injection and blood samples were collected over a period of 32 days. A specific radioimmunoassay was validated and used to discriminate rcGH from endogenous somatotropin. Plasma rcGH concentration was proportional to the dose delivered, but uptake and clearance rates were found to be independent of dose (between 0.5 and 5.0 g/g). Absorption of rcGH into the plasma was faster from the im site, but the peak levels attained were similar after im or ip treatment (using the same dose) as was area under the curve. Plasma half-life was calculated from the declining phase of the uptake/clearance profile but the results were biased by the concurrent uptake of rcGH from the ip or im reservoir of material, resulting in an over-estimation of the true half-life value. Ground running invertebrates were sampled by pitfall trapping in woodlands with high and low aerial pollution by heavy metals. Principal component analysis calculated for the environmental variables showed degree of metal contamination to be the most important difference between the sites. Calculation of canonical correspondence analysis (CCA) for different groups of animals e.g. Diplopoda, Opiliones, Aranaea and Carabidae was carried out and produced plots which can be interpreted with respect to the pollution effect. The effect of different environmental factors on individual species or subgroups delimited by size, breeding season, diet etc. was determined. Species associating with or disassociating from high pollution levels can be identified. No clear patterns were shown when examining size or breeding season in Carabids in relation to pollution. The highly polluted woods near the main pollution source at Avonmouth are lacking in lycosid spiders, the vacant niche may be filled by a large species of ground running agelenid which may be more adaptable in habits than the lycosids. CCA plots of both Aranaea and all species pooled together show a triangular shape. This is interpreted as representing a small number of species able to adapt to high pollution levels and a larger potential number found at ’clean' sites. 相似文献

Children's voices have been little heard in the Pacific research. The United Nations Convention on the Rights of the Child 1989 asserts the child's right to have a say on matters that affect them and for their views to be considered. There has been massive growth in technologically assisted participative research; however, we argue the value of hand drawn maps should not be underestimated in the rush to engage with more advanced techniques. We present data from 267 neighbourhood maps drawn by children in Fiji, Kiribati, Samoa and New Zealand. To better understand the social construction of knowledge in children's everyday lives, we propose two models to conceptualise the complexity of their world, a social connection and a spatial connection model. These models reveal how Pacific Island children negotiate different levels of social connection from home, family, and community through to transnational kinship relations. People, specifically family, provide the geographic basis on which their spatial encounters are overlaid. Irrespective of country or rural/urban/atoll setting, it is social space that is the strongest connector for children as displayed in their maps. Application of our models can be used to reveal how knowledge is socially constructed in Pacific children's everyday lives. 相似文献
