Despite continuing research efforts, knowledge of the transmission of the highly pathogenic avian influenza (HPAI) virus still has considerable gaps, which complicates epidemic control. The goal of this research was to develop a model to back-calculate the day HPAI virus is introduced into a flock, based on within-flock mortality data. The back-calculation method was based on a stochastic SEIR (susceptible (S) - latently infected (E) - infectious (I) - removed (= dead; R)) epidemic model. The latent and infectious period were assumed to be gamma distributed. Parameter values were based on experimental H7N7 within-flock transmission data. The model was used to estimate the day of virus introduction based on a defined within-flock mortality threshold (detection rule for determining AI). Our results indicate that approximately two weeks can elapse before a noticeable increase in mortality is observed after a single introduction into a flock. For example, it takes twelve (minimum 11 - maximum 15) days before AI is detected if the detection rule is fifty dead chickens on two consecutive days in a 10 000 chicken flock (current Dutch monitoring rule for notification). The results were robust for flock size and detection rule, but sensitive to the length of the latent and infectious periods. Furthermore, assuming multiple introductions on one day will result in a shorter estimated period between infection and detection. The implications of the model outcomes for detecting and tracing outbreaks of H7N7 HPAI virus are discussed. 相似文献

ABSTRACT: Vaccination of chickens has become routine practice in Asian countries in which H5N1 highly pathogenic avian influenza (HPAI) is endemically present. This mainly applies to layer and breeder flocks, but broilers are usually left unvaccinated. Here we investigate whether vaccination is able to reduce HPAI H5N1 virus transmission among broiler chickens. Four sets of experiments were carried out, each consisting of 22 replicate trials containing a pair of birds. Experiments 1-3 were carried out with four-week-old birds that were unvaccinated, and vaccinated at day 1 or at day 10 of age. Experiment 4 was carried out with unvaccinated day-old broiler chicks. One chicken in each trial was inoculated with H5N1 HPAI virus. One chicken in each trial was inoculated with virus. The course of the infection chain was monitored by serological analysis, and by virus isolation performed on tracheal and cloacal swabs. The analyses were based on a stochastic SEIR model using a Bayesian inferential framework. When inoculation was carried out at the 28th day of life, transmission was efficient in unvaccinated birds, and in birds vaccinated at first or tenth day of life. In these experiments estimates of the latent period (~1.0 day), infectious period (~3.3 days), and transmission rate parameter (~1.4 per day) were similar, as were estimates of the reproduction number (~4) and generation interval (~1.4 day). Transmission was significantly less efficient in unvaccinated chickens when inoculation was carried out on the first day of life. These results show that vaccination of broiler chickens does not reduce transmission, and suggest that this may be due to the interference of maternal immunity. 相似文献

In recent years it has become clear that cell-mediated immunity is playing a role in the control of lentivirus infections. In particular, cytotoxic T lymphocyte responses have been associated with improved outcome of infection, especially those directed against the regulatory proteins like Rev and Tat, which are expressed early after infection. Therefore, there is considerable interest in lentiviral vaccine candidates that can induce these types of immune responses. In the present study, we describe the construction and characterisation of expression vectors based on recombinant Semliki Forest virus system and modified vaccinia virus Ankara for the expression of feline immunodeficiency virus (FIV) accessory proteins Rev and OrfA. These recombinant viral vectors were used to immunize cats using a prime-boost regimen and the protective efficacy of this vaccination strategy was assessed after challenge infection of immunized cats with FIV. 相似文献

Experience with recent large-scale epidemics of Classical Swine Fever and Avian Influenza--among others in the Netherlands--have teached us several lessons that should prepare us better for future outbreaks. Among others, improving early detection of outbreaks--by using syndrome surveillance systems--is a key factor, in which farmers and veterinary practitioners have an imminent role. A major step in this respect is facilitation of the use of exclusion diagnostics without closing down the farm in clinical situations with non-specific clinical signs observed in sick animals. The hesitance of farmers and veterinary practitioners to report a suspect clinical situation on a livestock farm and how to facilitate that process is another major issue. Furthermore, the importance of communication between the field and the laboratory with respect to post mortem examination will be highlighted, and the need for outbreak simulation exercises with neighbouring countries in order to be better prepared, will be discussed. 相似文献

Highly pathogenic avian influenza A/H5N1 virus can cause morbidity and mortality in humans but thus far has not acquired the ability to be transmitted by aerosol or respiratory droplet ("airborne transmission") between humans. To address the concern that the virus could acquire this ability under natural conditions, we genetically modified A/H5N1 virus by site-directed mutagenesis and subsequent serial passage in ferrets. The genetically modified A/H5N1 virus acquired mutations during passage in ferrets, ultimately becoming airborne transmissible in ferrets. None of the recipient ferrets died after airborne infection with the mutant A/H5N1 viruses. Four amino acid substitutions in the host receptor-binding protein hemagglutinin, and one in the polymerase complex protein basic polymerase 2, were consistently present in airborne-transmitted viruses. The transmissible viruses were sensitive to the antiviral drug oseltamivir and reacted well with antisera raised against H5 influenza vaccine strains. Thus, avian A/H5N1 influenza viruses can acquire the capacity for airborne transmission between mammals without recombination in an intermediate host and therefore constitute a risk for human pandemic influenza. 相似文献

The historical geography of the landscape of a lowland brook valley in the sandy Kempen area (eastern Brabant, Netherlands) shows the interaction of ecological processes and land use, and helps to understand processes in the present landscape. In this location the human influence, especially on the groundwater hydrology, played a major role in the development of the landscape. Levels and flow of different types of groundwater interacted with vegetation development and human interference, to produce landscape patterns. Four main stages have been identified. In the prehistoric period, a natural deciduous forest covered the higher grounds, ombrotrophic peat was formed in the valley, and groundwater was relatively deep. In the medieval stage man settled on the edge of the valley, cleared parts of the forest and dug part of the peat. Groundwater levels were raised, which increased the rate of groundwater discharge and increased the amount of associated lowland peat formation in the valley. This tendency continued in modern times, when the area was completely deforested. Groundwater levels increased further due to decreased evapotranspiration, which gave rise to the use of ponds for fish and for water mills. Finally, in the most recent period the groundwater level has been lowered by extensive artificial drainage, partly on a regional scale. It was concluded that evaluation of historical changes in the landscape help provide landscape planners with a sound idea of the nature of the landscape. 相似文献

ABSTRACT: In the last decade many studies have been performed on the virulence of Newcastle disease virus (NDV). This is mainly due to the development of reverse genetics systems which made it possible to genetically modify NDV and to investigate the contribution of individual genes and genome regions to its virulence. However, the available information is scattered and a comprehensive overview of the factors and conditions determining NDV virulence is lacking. This review summarises, compares and discusses the available literature and shows that virulence of NDV is a complex trait determined by multiple genetic factors. 相似文献
