A 1-year-old Australian Shepherd (AS) was presented for a routine hereditary eye examination. During the examination multiple raised, brown to orange lesions were noted in the fundus, which could not be attributed to a known retinal disease in this breed. As they clinically most closely resembled canine multifocal retinopathy (cmr) and no indication of an acquired condition was found, genetic tests for BEST1 gene mutations were performed. These showed the dog to be homozygous for the cmr1 (C73T/R25X) gene defect. Furthermore, ultrasound (US), electroretinography (ERG), and optical coherence tomography were performed, confirming changes typical for cmr. Subsequently, the AS pedigree members were genetically and clinically tested, demonstrating autosomal recessive inheritance with no clinical symptoms in carrier animals, as was previously described for cmr. To our knowledge, this is the first reported case of canine multifocal retinopathy in the AS breed. Further investigations are under way. 相似文献
The presence of 12 genes associated with virulence in human attaching and effacing Escherichia coli (AEEC) was studied within a collection of 20 enterohemorrhagic E. coli (EHEC) and 206 atypical enteropathogenic E. coli (EPEC) isolated from ruminants. In addition, virulence genes and the clonal relationship of 49 atypical EPEC O26 strains isolated from humans and ruminants were compared to clarify whether ruminants serve as a reservoir of atypical EPEC for humans. A great diversity in the content of virulence gene was found. Thus, the espH, espG and map genes were detected in more than 85% of ruminant AEEC strains; the tccP2, espI, efa1/lifA, ehxA and paa genes were present in 50-70% of strains; and other genes such as tccP, espP, katP and toxB were detected in <25% of strains. EHEC strains contained more virulence genes than atypical EPEC strains. Our results suggest for the first time that the efa1/lifA gene is associated with diarrhea in newborn ruminants and that the AEEC strains with the H11 flagellar antigen are potentially more virulent than the non-H11 AEEC strains. Importantly, we identified a new intimin variant gene, eaeρ, in three ruminant atypical EPEC strains. The comparison of ruminant and human EPEC O26 strains showed that some ruminant strains possess virulence gene profiles and pulse-field gel electrophoresis pulsotypes similar to those of human strains. In conclusion, our data suggest that atypical EPEC is a heterogeneous group with different pathogenic potential and that ruminants could serve as a reservoir of atypical EPEC for humans. 相似文献
ABSTRACT: Leishmania major is the major cause of cutaneous leishmaniosis (CL) outside of the Americas. In the present study we have cloned six Leishmania genes (H2A, H2B, H3, H4, A2 and HSP70) into the eukaryotic expression vector pCMVbeta-m2a, resulting in pCMV-HISA70m2A, which encodes all six pathoantigenic proteins as a single polyprotein. This expression plasmid has been evaluated as a novel vaccine candidate in the BALB/c mouse model of CL. The DNA vaccine shifted the immune response normally induced by L. major infection away from a Th2-specific pathway to one of basal susceptibility. Immunization with pCMV-HISA70m2A dramatically reduced footpad lesions and lymph node parasite burdens relative to infected control mice. Complete absence of visceral parasite burden was observed in all 12 immunized animals but not in any of the 24 control mice. Moreover, vaccinated mice produced large amounts of IFN-gamma, IL-17 and NO at 7 weeks post-infection (pi), and they showed lower arginase activity at the site of infection, lower IL-4 production and a weaker humoral immune response than infected control mice. Taken together, these results demonstrate the ability of the HISA70 vaccine to shift the murine immune response to L. major infection away from an undesirable, Th2-specific pathway to a less susceptible-like pathway involving Th1 and Th17 cytokine profiles. 相似文献
Porcine circovirus type 2 (PCV2) is an economically important emerging pathogen associated with distinct syndromes and diseases in swine, collectively known as porcine circovirus associated diseases (PCVAD). The main purpose of this study was to investigate the presence of PCV2 in piglets affected with exudative epidermitis (EE) in Uruguay. In addition we aimed to analyze the phylogenetic relationships of the isolated strains. In June 2011 an outbreak of EE detected in a small herd was reported. Piglets presented skin lesions compatible with EE and symptoms associated with postweaning multisystemic wasting syndrome (PMWS) were also observed. Sera from affected and healthy animals were tested for the presence of viral DNA. Exclusively, diseased piglets were infected with PCV2. Phylogenetic analysis showed that PCV2 isolates belonged to PCV2b genotype. We report the detection and molecular characterization of PCV2 strains for the first time in Uruguay. 相似文献
The aim of this study was to evaluate lipid peroxidation, protein oxidation and activity of enzymes that are indicators of oxidative stress in Rangelia vitalii infection in dogs. Animals were divided into two groups: negative control (n=5) and infected with R. vitalii (n=7). After inoculation, the parasitemia was estimated daily by microscopic examination of smears. Lipid peroxidation (TBARS) and advanced oxidation protein products (AOPP); and delta-aminolevulinate dehydratase (δ-ALA-D), superoxide dismutase (SOD) and catalase (CAT) activities in blood were evaluated. The samples were collected at days 10 and 20 post-inoculation (PI). TBARS and AOPP levels were higher in the infected group in both analyzed periods (P<0.01). The δ-ALA-D activity was reduced in blood of dogs infected with R. vitalii on days 10 and 20 PI. SOD activity was significantly increased (P<0.01) in the blood of dogs infected with R. vitalii at days 10 and 20 PI, while CAT activity was significantly increased (P<0.01) only at day 20 PI when compared to non-infected animals. A positive correlation was observed between the degree of parasitemia and TBARS and AOPP levels and activity of antioxidant enzymes. The δ-ALA-D activity was negatively correlated with the degree of parasitemia. Based on the increased levels of TBARS, AOPP, SOD and CAT activities, and inhibition δ-ALA-D activity, we concluded that dogs experimentally infected with R. vitalii develop a state of redox unbalance and that these changes might be involved in the pathophysiology of disease. 相似文献
Oil content and grain yield in maize are negatively correlated, and so far the development of high-oil high-yielding hybrids
has not been accomplished. Then a fully understand of the inheritance of the kernel oil content is necessary to implement
a breeding program to improve both traits simultaneously. Conventional and molecular marker analyses of the design III were
carried out from a reference population developed from two tropical inbred lines divergent for kernel oil content. The results
showed that additive variance was quite larger than the dominance variance, and the heritability coefficient was very high.
Sixteen QTL were mapped, they were not evenly distributed along the chromosomes, and accounted for 30.91% of the genetic variance.
The average level of dominance computed from both conventional and QTL analysis was partial dominance. The overall results
indicated that the additive effects were more important than the dominance effects, the latter were not unidirectional and
then heterosis could not be exploited in crosses. Most of the favorable alleles of the QTL were in the high-oil parental inbred,
which could be transferred to other inbreds via marker-assisted backcross selection. Our results coupled with reported information
indicated that the development of high-oil hybrids with acceptable yields could be accomplished by using marker-assisted selection
involving oil content, grain yield and its components. Finally, to exploit the xenia effect to increase even more the oil
content, these hybrids should be used in the Top Cross™ procedure. 相似文献
We isolated the rotifers Brachionus ibericus and Proales similis from the sediment of shrimp tanks and studied their individual demographic characters and competition between them at two food levels (0.25?×?106, 1.00?×?106 cells ml?1 of Nannochloropsis oculata at 25 °C) and salinities ranging from 10 to 30‰. Our hypothesis was that growth rates would be higher with increasing food levels and salinities. Observations were taken twice a day for life table studies and daily once for population growth experiments. Using survivorship and fecundity data, we derived various life history variables. Although the average life span (7.6?±?0.4 days) and gross reproductive rate (33.8?±?2.9 neonate female?1 day?1) of B. ibericus were higher than those of P. similis (average life span 5.4?±?0.6 days and gross reproductive rate 13.0?±?0.6 neonate female?1 day?1), the population growth experiments showed that P. similis had higher r values (0.32?±?0.005 day?1) than B. ibericus (0.23?±?0.002 day?1) at 1.0?×?106 cells ml?1 of N. oculata. The rotifer P. similis was more adversely affected due to the presence of B. ibericus than vice versa. The data are important for developing techniques for a large-scale culture of these rotifers as food in aquaculture. 相似文献
ABSTRACT Xylella fastidiosa is the causal agent of citrus variegated chlorosis (CVC), a destructive disease of sweet orange cultivars in Brazil. Polymerase chain reaction (PCR)-based assays constitute the principal diagnostic method for detection of these bacteria. In this work, we established a real-time quantitative PCR (QPCR) assay to quantify X. fastidiosa in naturally and artificially infected citrus. The X. fastidiosa cell number detected in the leaves increased according to the age of the leaf, and bacteria were not detected in the upper midrib section in young leaves, indicating temporal and spatial distribution patterns of bacteria, respectively. In addition, the X. fastidiosa cell number quantified in leaves of 'Pera' orange and 'Murcott' tangor reflected the susceptible and resistant status of these citrus cultivars. None of the 12 endophytic citrus bacteria or the four strains of X. fastidiosa nonpathogenic to citrus that were tested showed an increase in the fluorescence signal during QPCR. In contrast, all 10 CVC-causing strains exhibited an increase in fluorescence signal, thus indicating the specificity of this QPCR assay. Our QPCR provides a powerful tool for studies of different aspects of the Xylella-citrus interactions, and can be incorporated into breeding programs in order to select CVC-resistant plants more quickly. 相似文献
Mastitis may be caused by a wide range of microorganisms able to induce distinct lesions in mammary tissues. This study aims to characterize the gross and microscopic features of mastitis in dairy cows and to correlate them with the pathogens involved. The udders of slaughtered dairy cows were inspected and milk samples from each mammary quarter or fragments of the parenchyma were sent for microbiological analysis, and tissue collected for histopathological evaluation. A total of 148 cows and 592 mammary quarters were collected. From these, 432 quarters (73%) had mastitis and in 160 (27%), no changes were observed. Mastitis was classified into seven patterns based on the histopathological findings, of which mixed, lymphoplasmacytic, and suppurative mastitides were the most prevalent with 35.9% (155/432), 27.1% (117/432), and 14.3% (62/432) of the cases, respectively. These patterns were associated with the same set of pathogens: Streptococcus spp., coagulase-negative Staphylococcus (CNS), Staphylococcus aureus, Streptococcus agalactiae, Streptococcus uberis, and Corynebacterium bovis. The pyogranulomatous pattern represented 7.2% (31/432) of the cases with distinct distribution based on the agent involved, mostly S. aureus and Nocardia sp. Abscedative mastitis accounted for 6.0% (26/432) of the cases; it was characterized by multiple abscesses in the parenchyma and was mainly caused by Trueperella pyogenes. Necrosuppurative mastitis represented 5.8% (25/432) of the cases which were characterized by severe parenchyma necrosis and were caused by bacteria such as CNS and Escherichia coli. The granulomatous pattern represented 3.7% (16/432) of the cases and was occasionally associated with Mycobacterium sp.
Background: Pregnancy and early life are critical periods of plasticity during which the fetus and neonate may be influenced by environmental factors such as nutrition.Maternal methionine(Met) supply in non-ruminants during pregnancy can affect offspring development and growth.Thus,the objective of this study was to investigate if increasing Met supply during late-pregnancy affects developmental parameters of the calf at birth and if either maternal Met or colostrum from Met-fed cows alters calf growth.Calves born to Holstein cows individually-fed a basal control [CON; 1.47 Mcal/kg dry matter(DM) and 15.3% crude protein] diet with no added Met or CON plus ethylcellulose rumen-protected Met(MET; Mepron? at 0.09% of diet DM; Evonik Nutrition Care GmbH,Germany)during the last 28 ± 2 d of pregnancy were used.A total of 39 calves were in CON(n = 22 bulls,17 heifers) and 42 in MET(n = 20 bulls,22 heifers).At birth,calves were randomly allocated considering dam treatment and colostrum as fol ows: 1) calves from CON cows and colostrum from CON cows(n = 21); 2) calves from CON cows and colostrum from MET cows(n = 18); 3) calves from MET cows and colostrum from MET cows(n = 22); and 4) calves from MET cows and colostrum from CON cows(n = 20).Al calves were housed,managed,and fed individual y during the first 9 wk of life.Results: Despite greater daily DM intake pre-partum in cows fed MET(15.7 vs.14.4 ± 0.12 kg/d,P 0.05),colostrum quality and quantity were not affected by maternal diet.At birth,MET calves had greater(P ≤ 0.05) body weight(BW,44.1 vs.42.1 ± 0.70 kg),hip height(HH,81.3 vs.79.6 ± 0.53 cm) and wither height(WH,77.8 vs.75.9 ± 0.47 cm).In contrast,concentrations of His,Lys,and Asn in plasma were lower(P ≤ 0.05) in MET calves.Regardless of colostrum source,the greater BW,HH,and WH in MET calves at birth persisted through 9 wk of age resulting in average responses of + 3.1 kg BW,+ 1.9 cm HH,and + 1.8 cm WH compared with CON.Average daily gain during the 9 wk was(P 0.05) 0.72 ± 0.02 kg/d in MET compared with 0.67 ± 0.02 kg/d in CON calves.Respiratory scores were normal and did not differ(P 0.05) due to maternal Met supply or colostrum source.However,fecal scores tended to be lower(P ≤ 0.10) in MET calves regardless of colostrum source.Conclusions: Increasing the maternal supply of MET during late-pregnancy enhanced growth in utero as wel as during the pre-weaning and early post-weaning periods.Although the ~ 1 kg/d greater DM intake during the last 2–3 wk prior to parturition could explain a portion of the 2 kg extra body mass of MET calves at birth,other mechanisms potential y encompassing nutrient assimilation efficiency likely played a role.Assessing the exact mechanisms sensitive to supply of Met or total amino acid supply during the latter stages of growth in utero merit further research. 相似文献
