Antimetastatic properties on both murine and human osteosarcoma cell lines (POS-1 and KHOS) have been evidenced using exopolysaccharide (EPS) derivatives, produced by Alteromonas infernus bacterium. These derivatives had no significant effect on the cell cycle neither a pro-apoptotic effect on osteosarcoma cells. Based on this observation, these EPSs could be employed as new drug delivery systems for therapeutic uses. A theranostic approach, i.e., combination of a predictive biomarker with a therapeutic agent, has been developed notably by combining with true pair of theranostic radionuclides, such as scandium 47Sc/44Sc. However, it is crucial to ensure that, once complexation is done, the biological properties of the vector remain intact, allowing the molecular tropism of the ligand to recognize its molecular target. It is important to assess if the biological properties of EPS evidenced on osteosarcoma cell lines remain when scandium is complexed to the polymers and can be extended to other cancer cell types. Scandium-EPS complexes were thus tested in vitro on human cell lines: MNNG/HOS osteosarcoma, A375 melanoma, A549 lung adenocarcinoma, U251 glioma, MDA231 breast cancer, and Caco2 colon cancer cells. An xCELLigence Real Cell Time Analysis (RTCA) technology assay was used to monitor for 160 h, the proliferation kinetics of the different cell lines. The tested complexes exhibited an anti-proliferative effect, this effect was more effective compared to EPS alone. This increase of the antiproliferative properties was explained by a change in conformation of EPS complexes due to their polyelectrolyte nature that was induced by complexation. Alterations of both growth factor-receptor signaling, and transmembrane protein interactions could be the principal cause of the antiproliferative effect. These results are very promising and reveal that EPS can be coupled to scandium for improving its biological effects and also suggesting that no major structural modification occurs on the ligand.  相似文献

BACKGROUND

The Japanese beetle Popillia japonica Newman is an insect pest native to Japan that has spread into North America, the Azores and, recently, into continental Europe. Here, we present a study assessing the effectiveness of a long-lasting insecticide-treated net (LLIN), assembled in semiochemical-baited attract-and-kill devices (A&Ks) as a low environmental impact means to control P. japonica in the field. We compared the attractiveness of three different forms of A&Ks that were left outdoors throughout the summer, and the residence time of P. japonica landing on them. Moreover, we performed a preliminary study testing the effectiveness of new LLINs after storage. Collected data also allowed us to investigate the beetles' diel flight patterns in relation to meteorological conditions.

RESULTS

Killing effectiveness of the field-exposed A&Ks declined steadily over the flight season (from 100% to 37.5%) associated with a decrease in residues of α-cypermethrin, the active ingredient in the LLINs. The different A&K forms (pyramidal, octahedral and ellipsoidal) attracted similar numbers of beetles. Individual beetles' residence time ranged from 75 to 95 s and differed slightly between A&K forms. Effectiveness of LLINs decreased by ≈30% after 1 year storage. Based on numbers landing on A&Ks, the beetles' flight activity peaked about 14:30 h and was inversely correlated with relative humidity.

CONCLUSION

This study indicates that semiochemical-baited A&Ks are effective for controlling P. japonica in the field. Because of active ingredient decay, the LLINs should be replaced after 30–40 days of field exposure to ensure that the A&Ks remain fully functional. © 2023 The Authors. Effects of physiological variables (age, body weight and sex) on lipemia (total lipids, phospholipids, triglycerides and NEFA), fecal fat and chymotrypsin excretion were examined in dogs. On comparing various ages and body weights, they found statistically significant differences, while only total lipids showed a statistically significant difference between the two sexes. Moreover, there was a significant correlation between body weight and fecal fat and chymotrypsin excretion. The results obtained contribute to the evaluation of small intestine functionality, especially in relation to some diseases, as malabsorption, maldigestion and steatorrhea in the dog.  相似文献

To determine lignin content in triticale and wheat straws, calibration models were built using Fourier transform mid-infrared spectroscopy combined with partial least-squares regression. The best model for triticale and wheat straws was built using averaged spectra with raw spectrum in spectrum format and constant in path length as spectral pretreatments. The values of r(2), root-mean-square error of prediction (RMSEP), and residual predictive deviation (RPD) for the triticale straw model were 0.935, 0.305, and 3.89, respectively. The r(2), RMSEP, and RPD values for the wheat straw model were 0.985, 0.163, and 8.50, respectively. Both models showed good predictive ability. A model built using both triticale and wheat straws indicated that the values of r(2), RMSEP, and RPD were 0.952, 0.27, and 4.63, respectively. This model also showed good predictive ability and could predict lignin contents in triticale and wheat straws with the same high accuracy.  相似文献

An investigation was made into a novel system aimed at reducing the impact of highly polluting wastewaters, and based on the combined action of catalytic oxidation and microbial biotechnology. The experimental part incorporated the following three schemes: chemical treatment using Fenton’s reaction for a single process (stage 1); biological treatment only (stage 2); and chemical oxidation followed by biological treatment (stage 3). Wastewaters with 2-mercaptobenzothiazole (MBT; 7,200–7,400 mg O2 l?1) were oxidized by stoichiometric amounts of dilute hydrogen peroxide (35%) in the presence of water soluble iron catalysts, either Fe (II) or Fe (III), at concentrations up to 1% w/w and above. As a result, transformation by chemical means of recalcitrant organics to more easily attackable end-products occurs, that can subsequently undergo conventional or advanced (microflora and biomass dispersed or adhered) biological treatments, with 90% of chemical oxygen demand abatement and 95% of MBT.  相似文献

Summary As triploids produce seedless fruits the best production is investigated. Comparative studies of embryogenesis, seed set, size, and germination were carried out following 4x×2x and 4x×4x crosses. On the average, both crosses yielded similar numbers and proportions of fully developed seeds per fruit. Seed development and germination took place precociously in 4x×2x seeds as compared to 4x×4x ones. The size of 4x×2x seeds was about 1/6 to 1/3 the size of 4x×4x seeds; thus, they were distinguishable from one another on the basis of size. Similarly, it was found that 3x and 4x seeds could be identified based on size following 4x×2x+4x crosses. It was concluded that 4x×2x crosses would be more successful than 2x×4x crosses for producing triploids.  相似文献

This study describes a model named HydroGEN that was conceived for simulating hydrographs of daily volumes and hourly flow rates during peak-demand periods in pressurized irrigation delivery networks with on-demand operation. The model is based on a methodology consisting of deterministic and stochastic components and is composed of a set of input parameters to reproduce the crop irrigation management practices followed by farmers and of computational procedures enabling to simulate the soil water balance and the irrigation events for all cropped fields supplied by each delivery hydrant in a distribution network. The input data include values of weather, crop, and soil parameters, as well as information on irrigation practices followed by local farmers. The resulting model outputs are generated flow hydrographs during the peak-demand period, which allow the subsequent analysis of performance achievable under different delivery scenarios. The model can be applied either for system design or re-design, as well as for analysis of operation and evaluation of performance achievements of on-demand pressurized irrigation delivery networks. Olive tree (Olea europaea L.) is commonly grown under environmental conditions characterised by water deficit, high temperatures and irradiance levels typical of Mediterranean semi-arid regions. Measurement of gas exchange, chlorophyll content, chlorophyll fluorescence and photoinhibition was carried out on two-year-old olive trees (cv. ‘Coratina’) subjected to a 21-day period of water deficit followed by 23 days of rewatering. At the beginning of the experiment, plants were divided in to two groups and subjected to different light regimes: exposed plants (EP) under a mean photosynthetically active radiation (PAR) at mid-day of 1800 μmol m−2 s−1 and shaded plants (SP) under a mean PAR of 1200 μmol m−2 s−1. The effect of drought and high irradiance levels caused a reduction of gas exchange and photosystem 2 (PSII) efficiency, in terms of quantum yield of PSII (ΦPSII) both in EP and SP. Shading conditions allowed plants to maintain a high photosynthetic activity at low values of stomatal conductance, whereas in EP the reductions in photosynthetic efficiency and intrinsic water efficiency were due to non-stomatal components of photosynthesis. The decrease in photosynthetic activity and the increase of photoinhibition under drought were more marked in EP than in SP. Full sunlight caused in EP a higher non-photochemical quenching, whereas SP showed a better photochemical efficiency. The information here obtained can be important to understand the mechanisms by which olive plants can minimize photoinhibition when subjected to simultaneous abiotic stresses.  相似文献

Summary Cannabis sativa L. is a dioecious species with sexual dimorphism occurring in a late stage of plant development. Sex is determined by heteromorphic chromosomes (X and Y): male is the heterogametic sex (XY) and female is the homogametic one (XX). The sexual phenotype of Cannabis often shows some flexibility leading to the differentiation of hermaphrodite flowers or bisexual inflorescences (monoecious phenotype). Sex is considered an important trait for hemp genetic improvement; therefore, the study of the mechanism of sexual differentiation is of paramount interest in hemp research. A morphological and molecular study of Cannabis sativa sexual differentiation has been carried out in the Italian dioecious cultivar Fibranova.Microscopic analysis of male and female apices revealed that their reproductive commitment may occur as soon as the leaves of the fourth node emerge; the genetic expression of male and female apices at this stage has been compared by cDNA-AFLP. A rapid method for the early sex discrimination has been developed, based on the PCR amplification of a male-specific SCAR marker directly from a tissue fragment.Five of the several cDNA-AFLP polymorphic fragments identified have been confirmed to be differentially expressed in male and female apices at the fourth node. Cloning and sequencing revealed that they belong to nine different mRNAs that were all induced in the female apices at this stage. Four out of them showed a high degree of similarity with known sequences: a putative permease, a SMT3-like protein, a putative kinesin and a RAC-GTP binding protein.  相似文献
