Over the past few years, new technological and scientific advances have reinforced the field of natural product discovery. The spirotetronate class of natural products has recently grown with the discovery of phocoenamicins, natural actinomycete derived compounds that possess different antibiotic activities. Exploring the MEDINA’s strain collection, 27 actinomycete strains, including three marine-derived and 24 terrestrial strains, were identified as possible phocoenamicins producers and their taxonomic identification by 16S rDNA sequencing showed that they all belong to the Micromonospora genus. Using an OSMAC approach, all the strains were cultivated in 10 different media each, resulting in 270 fermentations, whose extracts were analyzed by LC-HRMS and subjected to High-throughput screening (HTS) against methicillin-resistant Staphylococcus aureus (MRSA), Mycobacterium tuberculosis H37Ra and Mycobacterium bovis. The combination of LC-UV-HRMS analyses, metabolomics analysis and molecular networking (GNPS) revealed that they produce several related spirotetronates not disclosed before. Variations in the culture media were identified as the most determining factor for phocoenamicin production and the best producer strains and media were established. Herein, we reported the chemically diverse production and metabolic profiling of Micromonospora sp. strains, including the known phocoenamicins and maklamicin, reported for the first time as being related to this family of compounds, as well as the bioactivity of their crude extracts. Although our findings do not confirm previous statements about phocoenamicins production only in unique marine environments, they have identified marine-derived Micromonospora species as the best producers of phocoenamicins in terms of both the abundance in their extracts of some major members of the structural class and the variety of molecular structures produced.  相似文献

  • 1. A comparison was made between a variety of alien and endemic plant species from 272 aquatic and riparian habitats in Portuguese Mediterranean‐type streams in reference condition — i.e. near‐natural river corridors — and non‐reference condition. The objective was to detect differences in relative proportion and cover between these species groups. The differences in endemic and alien flora from siliceous and calcareous river types were also analysed. Environmental and human disturbance factors were related with the richness and cover of both species groups.
  • 2. A total of 568 species were found, of which 44 were alien and 28 were endemic. Alien species were present at 91% of the surveyed locations, and were consistently more widespread at non‐reference sites than at reference ones for both river types, with calcareous sites having a higher invasibility. Endemic species occurred at 45% of the sampling sites and displayed a significantly lower richness and cover than their alien counterparts.

Alien richness and cover were positively related to direct human disturbance within the river systems, and with floodplain uses such as urban occupation, intensive agriculture, and nutrient inputs. Endemic species also respond to anthropogenic variables, rather than to climatic and geographical ones, with richness and cover increasing as human impacts on fluvial systems and related floodplains decrease.

Comprehensive control of alien invasive species and the protection of endemic plant populations will require attempts at monitoring ecological river integrity, and the achievement of ‘good ecological status’ — one of the goals of the European Union's Water Framework Directive. Portuguese riparian areas must be managed in such a way as to protect the relatively few preserved riparian habitats by lowering the direct and indirect pressures in fluvial corridors and thus preventing future alien plant invasions.

Addressing the spatial and temporal variability of crops for agricultural management requires intensive and periodical information gathering from the crop fields. Unmanned Aerial Vehicle (UAV) photogrammetry is a quick and affordable method for information collecting; it provides spectral and spatial information when required with the added value of Digital Surface Models (DSMs) that reconstruct the crop structure in 3D using “structure from motion” techniques. In the full process from UAV flights to image analysis, DSM generation is one bottle-neck due to its high processing time. Despite its importance, the optimization of the required forward overlap for saving time in DSM generation has not yet been studied. UAV images were acquired at 50 and 100 m flight altitudes over two olive orchards with the aim of generating DSMs representing the tree crowns. Several DSMs created with different forward laps (in intervals of 5–6% from 58 to 97%) were evaluated in order to determine the optimal generation time according to the accuracy of tree crown measurements computed from each DSM. Based on our results, flying at 100 m altitude and with a 95% forward lap reported the best configuration. From the analysis derived from this configuration, tree volume was estimated with 95% accuracy. In addition, computing time was 85% lower in comparison to the maximum overlap studied (97%). It allowed computing the 3D features of 600 trees in a 3-ha parcel in a highly accurate and quick (a few hours after the UAV flights) manner by using a standard computer.  相似文献

The effect conjugated linoleic acid (CLA) accumulation in fish fillet on sensory and texture attributes has been evaluated. Fish were fed diets with graded levels of CLA, which were administered for different durations prior to harvest. Fish were sampled for fillet proximate composition and fatty acid profile for sensory and texture parameters at the end of the trial. Growth performance, feed conversion ratio and muscle proximate composition were not affected by dietary treatments. Muscle fatty acid profile was not affected by dietary treatments despite the increase in saturated fatty acids in fish fed diet with 10 g kg−1 of CLA for 16 weeks before harvest. Besides, the deposition levels of CLA isomers increased gradually depending on the CLA treatment, reaching its maximum level in fish fed 10 g kg−1 CLA diet for 20 weeks before harvest. Fish fed 10 g kg−1 were juicier (P < 0.05) compared with control group, whereas fish fed 5 g kg−1 CLA diet were less (P < 0.05) chewy than fish fed control diet regarding sensory attributes. Texture parameters were not significantly affected by dietary CLA. These results suggest that market size European sea bass could successfully incorporate CLA isomers with some minor positive effects on sensory analyses.  相似文献

Pepper (Capsicum annuum L.) fruits are highly appreciated by producers and consumers for their economical and nutritional value. Four different cultivars of coloured peppers in immature and mature stages were harvested throughout the spring and examined for their content of phenolic compounds, ascorbic acid and total antioxidant capacity (TAA) as well as for lipid peroxidation and carbonyl proteins as index of oxidative stress. Ripening and harvest period influenced the antioxidants and the development of oxidative processes in the cultivars differently: lipid peroxidation increased in mature peppers except in one cultivar (Y1075), while no changes in protein oxidation or in TAA were produced, except in Y1075 in which both parameters increased. Each cultivar presented differences in antioxidant compounds depending on the harvest period, but we could recommend May as the optimal if all cultivars have to be harvested at the same time, when levels of ascorbate, phenols and TAA were not decreased, fresh weight and proteins were elevated, and levels of oxidation were not as high as in June (except for Y1075). A previous study of the response of each cultivar to different environmental conditions results essential to establish a good program of selection of cultivars with high quality and productivity.  相似文献
