Chromosome preparations from salmon (Salmo salar) were obtained by means of leukocyte culture. The chromosome number varied from 55 to 60 with a clear maximum of 2n = 58. Rhizopus rot, caused by Rhizopus stolonifer, is a major postharvest disease of stone fruits. The disease is related to the occurrence of mechanical and physical damage; however, observations at a Brazilian wholesale market suggest that direct penetration can occur. Therefore, the penetration mechanisms of R. stolonifer in stone fruits were evaluated. To identify the production of enzymes that help with direct penetration by the pathogen, esterase activity, both in mycelial discs and in spore suspensions of the fungus in water and in modified Van Etten nutrient solution, was measured. Assays were also conducted to evaluate the growth of R. stolonifer on glucose or cutin as a sole carbon source. The pathogen grew on both media, and higher esterase activity was observed in the cutin medium. Wounded and unwounded peaches and nectarines were inoculated with R. stolonifer spore suspensions in water or in modified Van Etten nutrient solution. Wounded fruit inoculated with either of the R. stolonifer spore suspensions developed rhizopus rot, whereas unwounded fruit developed the rot only in the presence of spores in the modified Van Etten nutrient solution. Scanning electron and light microscopic examination showed the fungus can directly penetrate the nectarine cuticle. Diisopropyl fluorophosphate, a serine hydrolase inhibitor, prevented rot development in peaches. The results provide valuable evidence for the ability of R. stolonifer to directly penetrate unwounded stone fruits, probably due to the production of esterase enzymes.  相似文献

Climate change is expected to increase the frequency and magnitude of extreme thermal events in rivers. The Little Southwest Miramichi River (LSWM) and the Ouelle River (OR) are two Atlantic salmon (Salmo salar) rivers located in eastern Canada, where in recent years, water temperatures have exceeded known thermal limits (~23°C). Once temperature surpasses this threshold, juvenile salmon exploit thermal heterogeneity to behaviourally thermoregulate, forming aggregations in coolwater refuges. This study aimed to determine whether the behavioural thermoregulation response is universal across rivers, arising from common thermal cues. We detailed the temperature and discharge patterns of two geographically distinct rivers from 2010 to 2012 and compared these with aggregation onset temperature. PIT telemetry and snorkelling were used to confirm the presence of aggregations. Mean daily maximum temperature in 2010 was significantly greater in the OR versus the LSWM (p = 0.005), but not in other years (p = 0.090–0.353). Aggregations occurred on 14 and 9 occasions in the OR and LSWM respectively. Temperature at onset of aggregation was significantly greater in the OR (Tonset = 28.3°C) than in the LSWM (Tonset = 27.3°C; p = 0.049). Logistic regression models varied by river and were able to predict the probability of aggregation based on the preceding number of hours >23°C (R2 = 0.61 & 0.65; P50 = 27.4°C & 28.9°C; in the OR and LSWM respectively). These results imply the preceding local thermal regime may influence behaviour and indicate a degree of phenotypic plasticity, illustrating a need for localised management strategies.  相似文献

An epidemiological survey for Theileria annulata infection was conducted in 12 selected villages around Ankara in Central Anatolia, Turkey, during the period April 1990 to January 1993. During the survey, 198 cattle of 30 local breeds, 84 Holstein-Friesian×local breeds and 84 Holstein-Friesian breed were examined for antibodies to T. annulata and the presence of the vector ticks. Four species of Hyalomma ticks were identified: Hyalomma anatolicum anatolicum, Hyalomma anatolicum excavtum, Hyalomma detritum and Hyalomma marginatum marginatum. Salivary gland staining indicated that infected adult ticks of all four species were present and, therefore, were implicated in the transmission of tropical theileriosis in the field. Generally, the Hyalomma infestation rate was low, with the heaviest infestations occurring on the older animals. Young adults and calves had very low infestation rates. Most ticks seen on cattle were adults, very few nymphs were found. The blood smear and serological examination of the 198 cattle conducted in March, before the start of the first disease season, showed that the prevalence of piroplasmosis was 11.1% (22 out of 198) and the seroprevalence of T. annulata was 10.6% (21 out of 198). Forty-three animals were then excluded from the study because they were seropositive and/or harboured piroplasms. Ninety-two seronegative animals showed piroplasmosis (92 out of 155) and 34 seronegative animals became seropositive for T. annulata (34 out of 155) during the three disease seasons. One animal became clinically ill with tropical theileriosis and required treatment. The incidence of cattle showing piroplasmosis and disease in the total study sample was 50.7% and 0.5% per disease season, respectively. The seroconversion rate of new infection with T. annulata in the total study was 14.3% per animal season. The number of cattle showing piroplasmosis was much greater than the number of seropositive cattle, which may indicate the presence of another species of Theileria. The two different management systems encountered in the study were considered to have influenced the tick infestation levels.  相似文献

Background

To face climate change and subsequent rainfall instabilities, crop breeding strategies now include root traits phenotyping. Rapid estimation of root traits in controlled conditions can be achieved by using parallel electrical capacitance and its linear correlation with root dry mass. The aim of the present study was to improve robustness and efficiency of methods based on capacitance and other electrical variables, such as serial/parallel resistance, conductance, impedance or reactance. Using different electrode configurations and stem contact electrodes, we have measured the electrical impedance spectra of wheat plants grown in pots filled with three types of soil.

Results

For each configuration, parallel capacitance and other linearly independent electrical variables were computed and their quality as root dry mass estimator was evaluated by a ‘sensitivity score’ that we derived from Pearson’s correlation coefficient r and linear regression parameters. The highest sensitivity score was obtained by parallel capacitance at an alternating current frequency of 116 Hz in three-terminal configuration. Using a clamp, instead of a needle, as a stem electrode did not significantly affect the capacitance measurements. Finally, in handheld LCR meter equivalent conditions, capacitance had the highest sensitivity score and determination coefficient (r 2 = 0.52) at 10 kHz frequency.

Conclusion

Our benchmarking of linear correlations between different electrical variables and root dry mass enables to determine more coherent practices for ensuring a sensitive and robust root dry mass estimation, including in handheld LCR meter conditions. This would enhance the value of electrical capacitance as a tool for screening crops in relation with root systems in breeding programs.

Field efficacy of five insecticides to a greenhouse strain ofFrankliniella occidentalis (Pergande) (Thysanoptera: Thripidae) of known resistance status was studied. In a greenhouse trial, the mortality of aF. occidentalis strain (Uncali) which exhibited resistance only against cypermethrin in laboratory assays, reached 56%, 68%, 82%, 94% and 99%, 24 h after application of abamectin, cypermethrin, endosulfan, malathion and methomyl, respectively. In the laboratory, for larval mortality equal to that achieved in the field, much lower doses — half of the field dose of cypermethrin and endosulfan, and one-third and one-sixth of malathion and methomyl, respectively, were required in topical application. Because of differences between laboratory and field responses ofF. occidentalis to insecticides, a sound resistance evaluation program to determine field efficacy of insecticides is needed. Resistance increased 28.0- and 139.3-fold at LC50 in theF. occidentalis strain subjected to 27 and 44 cycles of cypermethrin selection, respectively. Veterinary inspection in slaughterhouses allows for the detection of macroscopic lesions reminiscent of bovine tuberculosis, but the presence of Mycobacterium bovis must be confirmed by laboratory methods. This study aimed at comparing the performances of the standard diagnostic tools used to identify M. bovis in tissue specimens sampled from suspicious animals. During a two years period, 1390 cattle were inspected at the Machachi abattoir in the Mejia canton - Ecuador. A total of 33 animals with granulomatous lesions were detected, representing 2.33% (16/687) and 2.42% (17/703) animals examined in 2007 and 2008, respectively. Ninety-four tissue specimens were sampled and screened for the presence of mycobacteria. Acid-fast bacilli were identified in one third of the suspicious cattle (11/33) and suggestive microscopic lesions in 27.3% (9/33) of the samples examined by direct microscopy and histopathology, respectively. Culturing on Stonebrink medium and 16S-rRNA-based polymerase chain reaction (PCR) yielded 36.4% (12/33) and 27.3% (9/33) of positives, respectively. Compared to culture, other diagnostic procedures displayed a lower sensitivity, with 56.5% for PCR, and 43.5% for direct microscopy and histopathology; however, the specificity was higher (94.4% for PCR and microscopy, and 97.2% for histopathology). We conclude that reliable post-mortem laboratory testing either requires the combination of a set of available diagnostic tools or necessitates the development of improved new-generation tools with better sensitivity and specificity characteristics.  相似文献
