Glutaminase (GLS) is the key enzyme of glutamine (Gln) metabolism and utilization. In this study, a cDNA encoding GLS protein was identified from common carp Cyprinus carpio intestine. The open reading frame of GLS cDNA encodes a polypeptide of 595 amino acids, which shows a high similarity with its zebrafish Danio rerio counterpart. Bioinformatic analysis showed the protein belongs to kidney‐type GLS. The putative protein has glutaminase domain and ankyrin repeats domain, which are highly conserved among vertebrate orthologues. Real‐time quantitative PCR analysis revealed that the abundance of GLS mRNA was the highest in the white muscle, followed by the brain, eyeball and pituitary. Glutaminase was ubiquitously expressed in all intestinal segments of common carp. The activity of GLS did not distribute uniformly along the entire length of the intestine. 以草地早熟禾(Poa pratensis)‘Nuglade'品种为试验材料,研究叶片喷施乙烯利对草地早熟禾在水分充足、干旱及复水恢复3种处理下叶片电解质渗透率、叶绿素、可溶性蛋白、丙二醛(MDA)等生理指标的影响。结果表明,乙烯利与对照组(CK)相比,乙烯利在干旱胁迫下保持更高的叶片相对含水量23.5%(对照为11.5%)和更低的电解质渗透率68.1%(对照为90.7%)及MDA含量53.90μmol·g~(-1)(对照检查131.62μmol·g~(-1)),且复水后的电解质渗透率恢复程度更好;此外乙烯利处理提高了复水恢复过程草地早熟禾的可溶性蛋白含量;说明,叶片喷施乙烯利能够缓解草地早熟禾的干旱胁迫损伤,并促进干旱后的复水恢复。 相似文献

2009年8月,山东泰安某海兰褐商品蛋鸡场,开产后产蛋率只有60%~70%,31周龄时,死亡率突然增加,最高达20%.患病鸡经大体剖检、病理组织学、PCR和免疫组织化学检测确诊为J亚群禽白血病.病理组织学检测发现送检鸡除了髓细胞瘤外,还存在明显的纤维肉瘤、组织肉瘤、腺癌和血管内皮瘤等肿瘤.纤维肉瘤单独存在,组织肉瘤、腺癌和血管内皮瘤分别和髓细胞瘤混合存在,本文详细描述了各肿瘤的组织学特点.ALV-J从肉种鸡传向商品蛋鸡后引起肿瘤谱扩展的原因和机制值得进一步研究. 相似文献

In this study, we irradiated Astragalus polysaccharides (APS) using 25 kGy 60Co γ ray to obtain γ‐irradiated Astragalus polysaccharides (IAPS) and then investigated the effects of IAPS on growth performance and immune function of cyclophosphamide (CPM)‐treated broilers. The physicochemical properties of APS and IAPS (molecular weight, water solubility, viscosity, morphological and structural properties) were evaluated. Then, 384 one‐day‐old Arbor Acres broiler chicks with similar initial weight were randomly assigned into 6 groups: the non‐treated group (control), and CPM‐treated groups were fed either a basal diet or the diets containing 900 mg/kg APS, or 900, 600, 300 mg/kg IAPS, respectively. On days 16, 18, and 20, all broilers except for the control group were intramuscularly injected with 0.5 ml CPM (40 mg/kg·BW). Broilers in the control group were intramuscularly injected with 0.5 ml sterilized saline (0.75%, wt/vol). This trial lasted for 21 days. The physicochemical treatment showed that γ irradiation could decrease the molecular weight and viscosity, and increase the water solubility of APS (p < 0.05), whereas the structural properties of APS was not affected. In the animal trial, 900 mg/kg APS or 900, 600 mg/kg IAPS relieved the decreased growth performance, thymus index, T lymphocytes proliferation, serum IgG concentration, NOS activity and the increased blood heterophil:lymphocyte ratio in CPM‐treated broilers (p < 0.05). CPM‐induced decreases in B lymphocytes proliferation and serum IgM concentration were only increased by IAPS at 900 mg/kg (p < 0.05). Overall, both APS and IAPS alleviated CPM‐induced immunosuppression. Especially, IAPS possessed better immunomodulatory effect than APS, indicating that γ irradiation could be used as an effective method to enhance the immunomodulatory activity of APS. 相似文献

2017年11月在广东省仁化县4个感染柑桔黄龙病的贡柑园中采叶片样品,进行常规PCR和实时荧光定量PCR检测,从不同病症叶片、典型症状叶片不同部位、初期感病树不同部位(正常和显症)叶片等3个方面分析了贡柑叶片中黄龙病菌"Candidatus Liberibacter asiaticus"(CLas)的分布情况。结果表明,叶片斑驳黄化症和均匀黄化症是与黄龙病最相关的症状,CLas含量很高;缺锌黄化症,具有黄龙病阳性与阴性两类样品;无明显黄化但呈革质化的叶片,CLas含量高。在斑驳黄化与均匀黄化叶片中,CLas的含量从叶尖到叶柄、从叶片到主叶脉呈现递增趋势,即以主叶脉与叶柄的含量更高。初期感病树未显症的部位也会检出黄龙病菌。 相似文献

Peripartal cows often experience negative energy balance, and are therefore prone to suffering from metabolic diseases such as hyperketonemia, which causes financial losses in dairy farms. This study aimed to investigate the effect of green tea polyphenol (GTP) supplementation during the periparturient period on production performance, oxidative stress and immunometabolism in dairy cows with hyperketonemia. One hundred Holstein cows were assigned to GTP (0.2 g/kg DM; n = 50) or control (without GTP; n = 50) group based on body weight, previous milk yield, and parity on d 15 before expected parturition. Subsequently, 10 cows with hyperketonemia were selected from each group, according to blood β-hydroxybutyric acid (BHBA) concentration between 1.2 and 2.9 mmol/L from 2 to 3 d postpartum. All cows were fed a close-up diet and a lactation diet with or without GTP supply from 15 d prepartum until 30 d postpartum. Milk and blood samples were obtained from 20 cows selected with hyperketonemia on 10, 20, and 30 d postpartum. Compared with control cows, greater milk yield and lower somatic cell count were observed in GTP cows. The GTP group had lower concentrations of BHBA, free fatty acids, cholesterol, triglyceride, reactive oxygen species, malondialdehyde, and hydrogen peroxide, greater concentrations of glucose, lower activities of aspartate aminotransferase, alanine aminotransferase, and glutamyl transpeptidase, alongside greater activities of superoxide dismutase, glutathione peroxidase, and total antioxidant capacity. Additionally, GTP supplementation up-regulated concentrations of interleukin-6 and interleukin-10, but down-regulated concentrations of tumor necrosis factor-α, interleukin-1β, interleukin-2, interleukin-8, and interferon-γ in plasma. Greater concentrations of plasma immunoglobulin G were also detected in the GTP group. Overall, the data suggested that GTP supplementation from 15 d prepartum to 30 d postpartum improved the milk yield and health status in cows with hyperketonemia during early lactation. 相似文献

In order to detect the clinical efficacy of oligosaccharide sulfate(DEAE) and dextran sulfate (DS-4000) against bovine mastitis,56 cows with bovine mastitis were divided into 5 groups (DEAE group,DS-4000 group,DEAE+penicillin group,DS-4000+penicillin group and penicillin group),and their breasts were perfusion with different drugs twice one day for 3 days.The effective rates of drugs were observed by clinic symptoms.Milk samples were collected before and after treatment of drugs,and the bacterial negative conversion rate of each group were detected by microbial examination.The results showed that total effective rates of 5 groups were 10.00%,20.00%,58.33%,75.00% and 50.00%,respectively,and the bacterial negative conversion rates were 0,12.50%,50.00%,54.55% and 44.44%,respectively.In conclusion,the curative efficiencies of the polysaccharides against bovine mastitis were weak,but it could be improved by combination with penicillin. 相似文献
