Large tracts of land in South American native forests have been converted to monoculture for livestock production which could negatively affect soils and environmental quality. A proposed management alternative is to use agroforestry systems, but little information is available on how they affect the soils. The objective was to assess the effect of a silvopastoral chronosequence in a tropical region of Colombia on soil microbiological and physico/chemical properties. The systems (three replications) were: monoculture grass conventional pasture (CP), native forest (F), and a silvopastoral system (SS) chronosequence with ages of 3 to 6 (SS3), 8 to 10 (SS8), or 12 to 15 (SS12) years. Soil responses to these land management were determined by measuring soil chemical (total C and pH), physical (penetrometer resistance and bulk density), and microbiological properties (activities of ß-glucosidase, urease, and alkaline and acid phosphatase and microbial biomass). Because of differences in soil texture across management treatments, microbiological properties were normalized on organic C content basis. SS12 showed the highest microbial biomass and enzyme activities on a per unit C basis and was consistently and significantly different from CP. Additionally, microbiological to C ratios were significantly affected by SS establishment age (P?0.05). The low microbiological responses were consistent with high penetration resistance and bulk density of CP which indicates that the SS are improving soil quality. This study presented quantitative data that SS stimulated soil microbial biomass and enzyme activities, which indicates greater potential to carry out biogeochemical process, and that SS provides a more favorable microbial habitat. 相似文献

A high-throughput mass spectrometric method is presented for the simultaneous detection of Sudan I, II, III, IV, and Para-Red azodyes in foodstuff. The method is based on the use of deuterium-labeled internal standards and atmospheric pressure chemical ionization (APCI) in a triple-quadrupole instrument. The gas-phase breakdown pattern of each labeled and unlabeled analogue displays the naphthoic moiety as a common fragment. The search for the parents of this common species (parent ion scans) allows, by flow injection and in a single run, the evaluation of the presence of each polluting species spiked in typical foodstuffs. A detailed assay of each azodye was performed by LC-APCI and isotope dilution method, through the multiple reaction monitoring approach, using deuterium-labeled internal standards. Sudan dyes can be quantified above the threshold of 10 ppb except for Sudan Para-Red, for which the limit of quantification was 20 ppb, likely due to the different ionization efficiency. 相似文献

Many countries have introduced mandatory labeling requirements on foods derived from genetically modified organisms (GMOs). Real-time quantitative polymerase chain reaction (PCR) based upon the TaqMan probe chemistry has become the method mostly used to support these regulations; moreover, event-specific PCR is the preferred method in GMO detection because of its high specificity based on the flanking sequence of the exogenous integrant. The aim of this study was to evaluate the use of very short (eight-nucleotide long), locked nucleic acid (LNA) TaqMan probes in 5'-nuclease PCR assays for the detection and quantification of GMOs. Classic TaqMan and LNA TaqMan probes were compared for the analysis of the maize MON810 transgene. The performance of the two types of probes was tested on the maize endogenous reference gene hmga, the CaMV 35S promoter, and the hsp70/cryIA(b) construct as well as for the event-specific 5'-integration junction of MON810, using plasmids as standard reference molecules. The results of our study demonstrate that the LNA 5'-nuclease PCR assays represent a valid and reliable analytical system for the detection and quantification of transgenes. Application of very short LNA TaqMan probes to GMO quantification can simplify the design of 5'-nuclease assays. 相似文献

Fluorescence spectroscopy of a green fluorescent protein mutant at single-molecule resolution has revealed a remarkable oscillatory behavior that can also be driven by applied fields. We show that immediately before unfolding, several periodic oscillations among the chemical substates of the protein chromophore occur. The Valdostana is a local dual purpose cattle breed developed in Italy. Three populations are recognized within this breed, based on coat colour, production level, morphology and temperament: Valdostana Red Pied (VPR), Valdostana Black Pied (VPN) and Valdostana Chestnut (VCA). Here, we investigated putative genomic regions under selection among these three populations using the Bovine 50K SNP array by combining three different statistical methods based either on allele frequencies (FST) or extended haplotype homozygosity (iHS and Rsb). In total, 8, 5 and 8 chromosomes harbouring 13, 13 and 16 genomic regions potentially under selection were identified by at least two approaches in VPR, VPN and VCA, respectively. Most of these candidate regions were population-specific but we found one common genomic region spanning 2.38 Mb on BTA06 which either overlaps or is located close to runs of homozygosity islands detected in the three populations. This region included inter alia two well-known genes: KDR, a well-established coat colour gene, and CLOCK, which plays a central role in positive regulation of inflammatory response and in the regulation of the mammalian circadian rhythm. The other candidate regions identified harboured genes associated mainly with milk and meat traits as well as genes involved in immune response/inflammation or associated with behavioural traits. This last category of genes was mainly identified in VCA, which is selected for fighting ability. Overall, our results provide, for the first time, a glimpse into regions of the genome targeted by selection in Valdostana cattle. Finally, this study illustrates the relevance of using multiple complementary approaches to identify genomic regions putatively under selection in livestock. 相似文献

Pintoi peanut (Arachis pintoi Krapov. & W.C. Greg.) is a warm‐season perennial legume with potential for use in grass–legume mixtures in Florida; however, limited information exists about its establishment in mixtures with bahiagrass (Paspalum notatum Flügge). The objective of this experiment was to evaluate the establishment of bahiagrass cv. “Argentine” and pintoi peanut cv. “Amarillo” as monocultures or mixture. The experiment was conducted in Ona, FL, from June to October of 2014 and 2015. Treatments were a split‐plot design of seeding strategies (bahiagrass monoculture, pintoi peanut monoculture or bahiagrass‐pintoi peanut mixtures; main plots) and two N fertilization strategies (30 or 80 kg/ha N; 30N and 80N; subplots), with four replicates. Measurements of plant density and frequency were taken every 4 weeks after seeding. Ground cover and herbage mass (HM) measurements were taken 112 days after seeding. Pintoi peanut ground cover was affected by seeding strategy × N level interaction. Ground cover was greater with 80N than 30N when pintoi was seeded in monoculture (3.6% vs 1.5% respectively) but not when it was seeded with bahiagrass (2.1%). There was no effect of seeding or N strategy on pintoi peanut proportion in HM (1.4%). Bahiagrass ground cover was not affected by seeding or N strategy (15.9%); however, its proportion in the HM was greater in 80N than 30N (12.1% vs 9.4% respectively). Mixed seeding did not negatively affect the establishment of bahiagrass and pintoi peanut and greater N fertilization levels improved some establishment parameters, with no negative effect for pintoi peanut. 相似文献

The use of grains alternative to wheat or rye is a challenging task for cereal technologists, and currently new technologies are under investigation as tools to improve the performances of these alternative grains. In this work the effects of high Hydrostatic Pressure (HP) on oat batters were investigated. Oat batters were treated for 10 min at 200, 300, 350, 400 or 500 MPa. Scanning electron microscopy and bright field microscopy showed that high HP significantly affected oat batter microstructure, and both starch and proteins were affected. Treatment at high HP significantly improved batter viscosity and elasticity. At pressures ≤300 MPa the increase in the viscous component was higher than the increase in the elastic component. On the contrary, at pressures ≥350 MPa the elastic component was predominant. Differential scanning calorimetry revealed that high HP induced starch gelatinisation, which started at 300 MPa and was almost complete after treatment at 500 MPa. High HP also affected water- and salt-soluble as well as urea-soluble oat proteins. Analysis of proteins soluble in different buffers revealed that pressures ≥300 MPa induced the formation of urea-insoluble complexes and/or disulfide bonds. Overall, the extent of starch gelatinisation and protein modification was dependent on the applied pressure, but the results collected so far clearly show that high HP can be used to improve the functionality of oat batters. 相似文献

Gluten is a fundamental component for the overall quality and structure of breads. The replacement of the gluten network in the development of gluten-free cereal products is a challenging task for the cereal technologist. The functionality of proteins from gluten-free flours could be modified in order to improve their baking characteristics by promoting protein networks. Transglutaminase (TGase) has been successfully used in food systems to promote protein cross-linking. In this study, TGase was investigated for network forming potential on flours from six different gluten-free cereals (brown rice, buckwheat, corn, oat, sorghum and teff) used in breadmaking. TGase was added at 0, 1 or 10 U/g of proteins present in the recipe. The effect of TGase on batters and breads was evaluated by fundamental rheological tests, Texture Profile Analysis and standard baking tests. Three-dimensional elaborations of Confocal Laser Scanning Microscopy (CLSM) images were performed on both batters and breads to evaluate the influence of TGase on microstructure. Fundamental rheological tests showed a significant increase in the pseudoplastic behaviour of buckwheat and brown rice batters when 10 U of TGase were used. The resulting buckwheat and brown rice breads showed improved baking characteristics as well as overall macroscopic appearance. Three-dimensional CLSM image elaborations confirmed the formation of protein complexes by TGase action. On the other side, TGase showed negative effects on corn flour as its application was detrimental for the elastic properties of the batters. Nevertheless, the resulting breads showed significant improvements in terms of increased specific volume and decreased crumb hardness and chewiness. Under the conditions of this study, no effects of TGase could be observed on breads from oat, sorghum or teff. Overall, the results of this study show that TGase can be successfully applied to gluten-free flours to improve their breadmaking potentials by promoting network formation. However, the protein source is a key element determining the impact of the enzyme. 相似文献
