The present investigation describes the synthesis and characterization of novel biodegradable nanoparticles based on chitosan for biomedical applications. The presence of primary amine groups in repeating units of chitosan grants it several properties like antibacterial activity, antitumor activity and so on. Chitosan forms nanoparticles spontaneously on the addition of polyanion tripolyphosphate which has greater antimicrobial activity than parent chitosan. In the present study, chitosan nanoparticles (ChNP) were prepared by the ionic gelation method. The physiochemical characteristics of nanoparticles were analyzed using XRD, SEM, FTIR. The antibacterial activity of chitosan nanoparticles against medical pathogens Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa was evaluated by calculation of minimum inhibitory concentration (MIC) and compared with chitosan and chitin activity. The mode of action and factors affecting antibacterial activity were also analyzed. ChNP compounds exhibited superior antimicrobial activity against all microorganisms in comparison with chitosan and chitin. The antibiofilm activity was studied using crystal violet assay and growth on congo red agar. The study is thus a good demonstration of the applicability of chitosan nanoparticles as an effective antimicrobial agent with antibiofilm activity as well.  相似文献

A detailed study was undertaken to measure mineral nitrogen status, mineralization rate of carbon and nitrogen, nitrification rate and fixation of added ammonium on 90 samples of coal mine spoils collected throughout Central Scotland. The inorganic nitrogen status of the spoils was very low (mean 3.4 mg N kg−1). The rate of carbon dioxide evolution was high compared to the low rate of nitrogen mineralized, resulting in a mean mineralized C:N ratio of 178. Nitrification of added ammonium was measurable on only half of the samples studied. Spoil with pH < 4.8 showed no nitrification of added ammonium. However, on some sites above this pH no net nitrification was measured, which suggested the absence of nitrifying bacteria. A considerable amount of applied ammonium nitrogen was lost either as a result of ammonium fixation by clay minerals, or assimilation by the heterotrophic population due to the wide mineralizable C:N ratio.  相似文献

Acute (24 hours) and chronic (90 days) oral toxicity studies on the ethanolic extracts of common spices Cinnamomum zeylanicum Nees bark and Piper longum L. fruits were carried out in mice. Acute dosages were 0.5, 1.0 and 3 g/kg while the chronic dosage was 100 mg/kg/day. All external morphological, hematological and spermatogenic changes, in addition to body weight and vital organ weights, were recorded. The extracts of both the plants caused no significant acute or chronic mortality compared to the control during this study. During chronic treatment there was no significant change in the pre- and post treatment body weight of the test animals while the weight gain in the control group was significant. C. zeylanicum treatment caused reduction in liver weight while P. longum caused a significant increase in the weight of the lungs and spleen of the treated animals compared to the control. Hematological studies revealed a significant fall in hemoglobin level of C. zeylanicum treated animals. Both of the extracts induced a significant increase in reproductive organ weights, sperm motility, sperm count and failed to illicit any spermatotoxic effect.  相似文献

A feeding trial was conducted in a recirculating system to determine the dietary protein requirement for juvenile black sea bass. Six isocaloric diets were formulated to contain varying levels of crude protein (CP) ranging from 36 to 56% (36, 40, 44, 48, 52, and 56%) by substituting a mixture of carbohydrates and lipid for fish meal. The feeding experiment was carried out in 18‐75 L aquaria stocked at a density of 15 juveniles (initial average weight 6.7 g) per tank. Fish were fed test diets in triplicate tanks to apparent satiation twice a day for 8 wk. Whole‐body proximate composition was analyzed after the feeding trial. After the feeding trial, weight gain and specific growth rate of fish fed the 44% CP diet were not significantly different from those fed the 48, 52, and 56% CP diets, but were significantly higher (P < 0.05) than those fed the 36 and 40% CP diets. Feed conversion efficiency and protein efficiency ratio were significantly affected by dietary protein level. The dietary requirement of protein for maximum growth of black sea bass juveniles, estimated using broken‐line regression analysis on weight gain, was 45.3% and maximum weight gain occurred at 52.6% based on polynomial regression analysis.  相似文献

The relationship between feeding behavior and performance of 274 feedlot cattle was evaluated using Charolais cross steers from 2 consecutive years averaging 293 ± 41 kg for yr 1 (n = 115) and 349 ± 41 for yr 2 (n = 159). Steers were blocked by BW and assigned to 3 (yr 1) or 4 (yr 2) feedlot pens equipped with a radio frequency identification system (GrowSafe Systems). Each pen contained 5 feeding stalls that allowed individual animal access to a feed tub suspended on load cells. The system recorded animal identification, duration, and frequency of feedings as well as the amount of feed consumed during each visit. Daily variation in DMI (DVI), calculated as the absolute difference in DMI from one day to the next, as well as eating rate were determined for each steer. Barley-based diets were delivered to meet steer ad libitum intake over the 213- and 181-d feeding periods for yr 1 and 2 of the study, respectively. The backgrounding periods included the first 85 and 56 d of yr 1 and 2, respectively, in which steers were fed a 14 to 30% concentrate diet, whereas the finishing periods included the last 116 and 101 d of feeding in yr 1 and 2, respectively, with the diet consisting of 77.9% concentrate. Steers were weighed individually every 14 d. To relate feeding behavior to performance, steers were grouped by ADG and G:F and categorized as high, average, or low (based on 1 SD greater than and less than the mean). In the backgrounding and finishing periods of both years of the study, steers classified as having high ADG exhibited greater (P < 0.001) DVI than steers classified as having average or low ADG. Total daily DMI was also greater (P < 0.001) for steers in the high ADG group than those in the low ADG group. Overall, those steers with the greatest G:F also tended (P = 0.15) to have greater DVI than average or low G:F steers. Compared with average or low G:F steers, DMI by high G:F steers in both years of the study was less during backgrounding, finishing, and overall (P = 0.02). Bunk visits and bunk attendance duration were less frequent and shorter (P ≤ 0.01) overall for high compared with low G:F steers. In this study, steers with more variable eating patterns exhibited greater ADG and tended to have greater G:F, a finding that is contrary to industry perception.  相似文献

Light emitting diode (LED) is more energy efficient than incandescent or fluorescent light. This study was to evaluate effects of different colored LEDs on milk production, milk composition, and physiology of Holstein cow. According to milk production and parity, cows (n = 186) were allotted to four treatments: control (natural daylight), white, yellow, and blue LED groups. Of these, 40 cows that had passed 57 day‐in‐milk were used. Yellow and blue LED groups demonstrated greater rates of decline in milk production than control and white LED groups. At the finish point, milk fat, protein, and lactose contents were the lowest in the blue LED group, whereas milk–urea–nitrogen levels were the highest in the yellow and blue LED groups. Extended exposure to blue LED light lowered antioxidant enzyme activity and insulin‐like growth factor‐1 levels. Prolactin concentrations were higher in the white and blue LED groups than in the control. Cortisol level was the highest in the blue LED group among the groups. Nonesterified fatty acid levels in the yellow and blue LED groups decreased to the greatest extent compared to the start point. These results suggest that blue LED light can decrease milk production and generate more stress than white and yellow LED lights.  相似文献

Bovine tuberculosis (BTB) is a widespread and endemic disease of cattle in Ethiopia posing a significant threat to public health. Regular surveillance by skin test, bacteriology, and molecular methods is not feasible due to lack of resources. Thus, routine abattoir (RA) inspection will continue to play a key role for national surveillance. A cross-sectional study was conducted at Woldiya municipal abattoir from April 1, 2009 to April 5, 2010 to estimate the prevalence of BTB in slaughtered cattle on the basis of detailed abattoir inspection and to compare efficacy of RA inspection with respect to detailed abattoir inspection and isolation and identification of Mycobacterium. Diagnostic accuracies (with corresponding measures of statistical uncertainty) were determined by computing test property statistics (sensitivity and specificity). Agreement between RA and detailed abattoir inspections was measured using kappa statistics. Out of 1,029 slaughtered heads of cattle examined during the study period, 63 (6.12 %) and 15 (1.45 %) were diagnosed with gross tuberculous lesions by detailed abattoir meat inspections and RA meat inspections, respectively, making a prevalence of 6.12 % (95 % CI: 5.2–7.1) on the basis of detailed abattoir inspection. About 59.45 % of tuberculous lesions were observed in the lungs and associated lymph nodes, whereas 35.13 % lesions were from the lymph nodes of the head. From 63 cattle suspected with tuberculosis (TB) based on detailed abattoir meat inspection, nine (19.05 %) were identified as Mycobacterium bovis, while three (4.8 %) as Mycobacterium tuberculosis. The sensitivity of RA meat inspection was 23.8 % in comparison to the detailed abattoir meat inspection and 25 % in comparison to culture, respectively. Poor agreement (k?=?0.37) was seen between RA meat examination and detailed abattoir meat examination methods. Similarly, poor agreement (k?=?0.013) was seen between RA meat examination and culture results. In conclusion, relatively higher prevalence (6.12 %) was recorded in Woldiya municipal abattoir on the basis of detailed Abattoir inspection and RA meat inspection protocols currently utilized in Ethiopia which are insufficient to detect the majority (76.19 %) of TB lesions at the gross level, which indicates the magnitude of meat borne zoonotic TB as an ongoing risk to public health. Detailed abattoir inspection protocols were demonstrated to improve the detection level by approximately fourfold. In conclusion, routine meat inspections have limitations in detecting BTB-suggestive lesions which indicate the magnitude of meat-borne zoonotic TB as an ongoing risk to public health.  相似文献
