While the use of NMR and stable isotopes in metabolism studies is hardly new, it is only recently that isotope-edited NMR spectroscopy has been applied in kinetic studies of glyphosate metabolism of soil microbes. NMR can detect multiple species simultaneously and non-destructively, yielding valuable information on structural identification of metabolites. T riple R esonance I sotope ED ited spectroscopy (TRIED), [2H]NMR, and [2H–13C] INEPT (I nsensitive N ucleus E nhancement through P olarization T ransfer) are three isotope-edited techniques which have been used in combination to examine the microbial degradation of glyphosate (N-phosphonomethylglycine). Using 13C- and 15N-labeled glyphosate, TRIED can detect multiple metabolites in crude matrices at submicrogram levels, an improvement over earlier techniques where milligrams were needed. It can detect 500 nanograms of 13C–15N-labeled compound in a crude sample (1 : 1400 mass ratio), only a few hours work being required. [2H]NMR and [2H–13C]INEPT were also used as complementary techniques to further examine metabolites whose 13C–15N bond has been cleaved. The three-isotope-edited methods produced results consistent with both radioactivity and HPLC analyses. Accordingly, we are able to detect minute levels of metabolites in the presence of complex mixtures, minimizing the costs and time of sample purification. ©1999 Society of Chemical Industry  相似文献

ABSTRACT Wheat (Triticum aestivum) cultivar Pavon 76 carries slow-rusting resistance to leaf rust that has remained effective in Mexico since its release in 1976. 'Pavon 76' was crossed with two leaf rust-susceptible wheat cultivars, Jupateco 73S and Avocet S, and between 118 and 148 individual F(2) plant-derived F(3) and F(5) lines were evaluated for adult-plant leaf rust resistance at two field sites in Mexico during different seasons. Evaluation of F(1) plants and parents indicated that the slow-rusting resistance was partially dominant. Segregation in the F(3) and F(5) indicated that the resistance was based on two genes with additive effects. Monosomic analysis was carried out to determine the chromosomal locations of the resistance genes. For this purpose, two or three backcross-derived cytogenetic populations were developed by crossing 'Pavon 76' with a monosomic series of adult-plant leaf rust-susceptible cultivar Lal-bahadur. Evaluation of such BC(2)F(3) and BC(3)F(3) lines from 16 confirmed 'Lalbahadur' monosomics indicated that one slow-rusting gene was located in chromosome 1B of 'Pavon 76'. This gene, designated as Lr46, is the second named gene involved in slow-rusting resistance to leaf rust in wheat.  相似文献

An insect growth regulator (IGR), lufenuron (Match 5EC), was tested for its toxicity toHelicoverpa armigera on cotton. Potency of the IGR against the larval stage of the pest was demonstrated with respect to larval instars; the LC90 values of 1st, 2nd, 3rd, 4th and 5th instar larvae were 5.63, 7.89, 8.03, 11.39 and 14.76 mg a.i.l −1, respectively. However, different larval instars did not differ significantly with respect to LC50 and LC10. IGR-treated larvae had swollen heads and were significantly smaller (1.5–2.3 mm) than the untreated control (2.9 mm). Larval weight was significantly reduced from 190 mg in the control to 50–70 mg in the lufenuron treatment. IGR treatment in the larval stage significantly affected both pupal length and pupal weight. Pupal duration of the test insect was significantly extended by IGR treatment. Pupal deformities, including an inability to shed the last larval skin and formation of larval-pupal intermediates, occurred following treatment. A significant reduction in adult emergence was recorded. In addition, abnormalities in the form of development of cavities in the forewings of adult were evident. Tan spot of wheat, caused by the fungus Pyrenophora tritici-repentis, is a destructive disease worldwide that can lead to serious losses in quality and quantity of wheat grain production. Resistance to multiple races of P. tritici-repentis was identified in a wide range of genetically diverse genotypes, including three different species Triticum aestivum (AABBDD), T. spelta (AABBDD), and T. turgidum (AABB). The major objectives of this study were to determine the genetic control of resistance to P. tritici-repentis races 1 and 5 in 12 newly identified sources of resistance. The parents, F(1), F(2), and F(2:3) or F(2:5) families of each cross were analyzed for the allelism tests and/or inheritance studies. Plants were inoculated at the two-leaf stage under controlled environmental conditions and disease reaction was assessed based on lesion-type rating scale. A single recessive gene controlled resistance to necrosis caused by P. tritici-repentis race 1 in both tetraploid and hexaploid resistant genotypes. The lack of segregation in the inter- and intra-specific crosses between the resistant tetraploid and hexaploid genotypes indicated that they possess the same genes for resistance to tan necrosis and chlorosis induced by P. tritici-repentis race 1. A single dominant gene for chlorosis in hexaploid wheat and a single recessive gene for necrosis in tetraploid wheat, controlled resistance to P. tritici-repentis race 5.  相似文献

This paper examines the potential influence of soil management and land use on soil carbon on cropping farms in New South Wales (NSW), Australia. Soil organic carbon (SOC) data from ten farms spatially distributed across NSW were examined on two occasions. Soil cores to a depth 0–30 cm were measured for SOC and, as expected, SOC in the A horizon (1.16%) was significantly (p < .001) greater than in the B horizon (0.74%) of all profiles. Analysis of the 2013 and 2015 SOC data indicated that in many ways, the results runs counter to other SOC studies in Australia. Importantly, the mean SOC concentration in these agricultural soils was significantly (p < .001) less under cropping (2013-1.05%, 2015-0.97%) than in native sites (2013-1.20%, 2015-1.16%). Out of the total of 35 sites sampled from 10 farms, SOC in 49% of sites did not change significantly over 2 years, in 17% it increased significantly, whereas in 34% it decreased. Further, a clear implication of drought on SOC was seen on sites that were uncropped based on a critical value for a 95% confidence interval (p < .05) and complemented by the significant correlation (p < .05) between average annual precipitation deficit (ANPD) and SOC across the state with R2 = 0.39. The mean SOC was found to be directly proportional to standard deviation and standard error. In terms of spatial variability, the C0 (nugget) value was greatest for farms with a large mean SOC and the average variogram in this study has a range of approximately 200 m which is potentially useful in determining sampling spacing for soil carbon auditing purpose. Similar empirical data over more years are required to better estimate SOC levels and to determine whether at a farm scale, factors such as land management, land use and climate can be related to soil carbon change and variability.  相似文献

Soybean breeders have not exploited the diversity of the 26 wild perennial species of the subgenus Glycine Willd. that are distantly related to soybean [Glycine max (L.) Merr.] and harbour useful genes. The objective of this study was to develop a methodology for introgressing cytoplasmic and genetic diversity from Glycine tomentella PI 441001 (2= 78) into cultivated soybean using ‘Dwight’ (2= 40) as the male parent. Immature seeds (19–21 days post‐pollination) were cultured in vitro to produce F1 plants (2= 59). Amphidiploid (2= 118) plants, induced by colchicine treatment, were vigorous and produced mature pods and seeds after backcrossing with ‘Dwight’. The BC1 plants (2= 79) produced mature seeds in crosses with ‘Dwight’. Chromosome numbers in BC2F1 plants ranged from 2= 41–50. From BC2F2 to BC3F1, the number of plants in parentheses with 2= 40 (275), 2= 41 (208), 2= 42 (80), 2= 43 (27), 2= 44 (12) and 2= 45 (3) were identified. Fertile lines were grown in the field during 2012 and 2013. This is the first report of the successful development of new alloplasmic soybean lines with cytoplasm from G. tomentella.  相似文献

Segregation for salinity tolerance and ISSR markers based molecular polymorphism were investigated in a F3 plant population raised via single-seed descent method from a cross between salt-tolerant indica rice variety CSR10 and salt-susceptible premium traditional Basmati rice variety Taraori Basmati HBC19. A total of 130 F3plants were evaluated individually for salinity tolerance on 1–9 scale on the basis of seedling growth parameters; the average score ranged between 1.7 to 8.3. Frequency distribution curve obtained using the salinity tolerance data of F3 population and a chi-square analysis, showed a good fit to a normal distribution. Eleven plants each in the category of salt-tolerant and salt-susceptible were selected from the segregating F3 population for ISSR marker analysis. A total of 149 bands (4–11 bands per primer) ranging from 200 to 3530 bp were scored for the two rice varieties and the selected CSR10 × HBC19 segregating F3 plants using 26 ISSR primers. Of these, 89 were monomorphic and 60 were polymorphic. Of the 60 polymorphic bands,36 and 20 bands were specific to CSR10 andHBC19 respectively. The remaining four bands were amplified using UBC primers 810,848, 853 and 886 and present in only some of the CSR10 × HBC19 F3 plants. Notably, ISSR primers with dinucleotide repeat motif and 5'-anchored end amplified more number of bands (7.0 bands/primer) compared to3'-anchored dinucleotide primers (5.4bands/primer), but 3'-anchored dinucleotide primers revealed higher level of polymorphism (2.6 polymorphic bands/primer) compared to 5'-anchoreddinucleotide primers (1.43 polymorphic bands/ primer). While distribution of majority of the polymorphic bands were more or less in the expected ratios in salt-tolerant and/or salt-sensitive F3segregating plants, but some of the bands amplified using UBC ISSR primers 823, 825,826, 849, 853, 864, 866 and 884 showed highly skewed distribution. Such polymorphic bands stand greater chances of having a linkage with the genes/ QTLs for salinity tolerance and shall be the target for further studies. Summary Potato spindle tuber viroid (PSTV) and potato virus Y (PVY) were isolated from plants of cultivar Kennebec with severe necrotic symptoms in the field. In the greenhouse, severe necrotic symptoms were reproduced only when potato plants were infected either simultaneously with PSTV+PVY, or with PSTV prior to PVY infection. Thirteen additional potato cultivars were tested in the greenhouse for this synergistic reaction and eight developed necrotic responses similar to cv. Kennebec. PVY concentration was significantly higher in doubly infected plants, compared with those infected with PVY alone.  相似文献

Weeds caused serious problem on yield reduction of basmati rice worldwide. Losses caused by weeds varied from one country to another, depending on the presence of dominant weeds and the control methods practiced by farmers; therefore, suitable plant population and weed management practices should be adopted. Keeping these in mind, a field experiment was carried out during kharif seasons of 2009 and 2010 at crop Research Centre of SVPUA&T, Meerut, India comprising 4 planting geometries, viz. 20, 30, 40, and 50 hills m?2 as main plot factor, and 5 weed management practices (Butachlor @ 1.0 kg ha?1, Butachlor @ 1.0 kg ha?1 fb (followed by) one hand weeding, Butachlor @ 1.0 kg ha?1 fb Almix @ 4 g ha?1, two hand weedings and weedy check) in a split plot design with 3 replications. Experimental results revealed that plant population of 50 hills m?2 proved superior over that of 20 hills m?2 in respect of weed density, weed dry weight, number of tillers m?2, yield attributes, grain, straw, and biological yields. The maximum grain yield (29.00 and 31.00 q ha?1) and straw yield (51.30 and 52.50 q ha?1) were recorded in 50 hills m?2 followed by 40 hills m?2 during 2009 and 2010, respectively. In respect of nitrogen, phosphorus, and potassium removal, a reverse trend was observed: the highest in 20 hills m?2 followed by 30, 40, and 50 hills m?2. As far as the weed management practices are concerned, both chemical and mechanical methods of weed control were found superior over weedy check. The lowest weed density, dry weight, and highest weed control efficiency, maximum length of panicle?1, number of panicle (m2), and 1000-grain weight and grain yield of 30.40 and 32.60 q ha?1 were recorded with two hand weedings which was at par with Butachlor @ 1.0 kg ha?1 fb one hand weeding over rest of the weed management practices.  相似文献

In the 2014 and 2015 crop seasons, the efficacies of different types, rates and combinations of mineral oil and insecticide foliar sprays for reducing Potato virus Y (PVY) spread were tested in controlled field trials in New Brunswick (NB), Canada. Experimental plots were planted with certified PVY-free Goldrush, supplemented with known virus-infected seed to raise PVY inoculum to 2.3% and 3% at the beginning of the 2014 and 2015 seasons, respectively. Treatments consisted of mineral oil-only sprays at different application rates, insecticide-only sprays of differing numbers, and several combined mineral oil and insecticide spray regimes, all compared to a no-spray control treatment. PVY spread to 18% (2014) and 22% (2015) of initially virus-free plants in no-spray control plots, with significant reductions observed in PVY spread in several treatments. Greatest PVY reductions, as low as 4% (2014) and 12% (2015), were in combined mineral oil and insecticide spray treatments, followed by oil-only sprays; while insecticide-only sprays did not significantly reduce PVY spread. As well as measuring PVY spread to marked test plants and randomly collected post-harvest tuber sample from the plots, exhibited similar treatment pattern for PVY incidence. Multiple logistic regression modeling confirmed the relative efficacy of combined oil and insecticide sprays for reducing PVY spread, while accounting for variable inoculum and aphid factors. Modeling also highlighted the importance of planting low-PVY seed initially, and of early application of foliar sprays. Local best management practice recommendations for reduction of in-field PVY spread were discussed.  相似文献
