AIM:To explore the effects of PMA(phorbol-12-myristate-13-acetate, a tumor promoter, mimicking the action of diacylglycerol on PKC)and laminin on the adhesion and the proliferation of human hepatocellular carcinoma cells, and provide a new clue to liver cancer treatment.METHODS:Human hepatocellular carcinoma cell line(BEL-7402)was used to identify the endogenous laminin and protein kinase C-α(PKC-α) expression, and the effects of laminin and PMA on the adhesion and the proliferation were also investigatedin vitro.RESULTS:By the effect of exogenous laminin, human hepatocellular carcinoma cell (BEL-7402) possessed endogenous laminin expression and increased the adhesion and the proliferation, which was showed the synergistic action by the effect of PMA in combination. By the action of PMA alone, the proliferation and the PKC-α expression increased by exogenous laminin were decreased, and the adhesion and the endogenous laminin expression were increased.CONCLUSIONS:The finding suggested that the adhesion and the proliferation of human hepatocellular carcinoma cell were closely related to the effects of endogenous or exogenous laminin, which were associated with cPKC-α activity. Therefore, the application of anti-laminin antibody in combination with PKC antagonist might be a new clue to find out the therapy for liver cancer. 相似文献

AIM: To investigate the application of atomic force microscope (AFM) in clinic diagnosis. METHODS: Topographic images and some parameters of large range field and microstructures of erythrocytes in the blood of normal subjects, lung cancer and myelodisplastic syndrome (MDS) patients were examined by atomic force microscope. RESULTS: Many clear topographic images of many erythrocytes, single erythrocyte, and microstructure of erythrocyte membrane surface were obtained. Many erythrocytes in lung cancer patients were found to change into echinocytes. One erythrocyte had 10-20 protuberances, most of which, with a mean width of 589. 0 nm and a length of 646. 7 nm, were on the edge of cells. The protuberances on the center of echinocytes are lodged and embedded. The erythrocytes of MDS patients were biconcave in shape. Many apertures with different diameters of tens to hundreds nanometer appeared on the surface of cell membrane. CONCLUSIONS: AFM can be widely applied in clinic pathological inspection, including quantification of cells, obtainment and comparison of many parameters (such as diameter, thickness, volume, surface, surface area/volume ratio), observation of topograph of single cell, and observation and comparison of membrane surface microstructure of cells, and so on. 相似文献

Articular cartilage repair is one of the most challenging issues which remains to be resolved in clinic work. Discovering of bone marrow stromal cells (BMSC) and its application in tissue engineering provide new methods for the treatment of cartilage defects. 甜樱桃作为一种早熟、优质、保健果品,而且种植效益较高,在国内国际都正处于发展时期。根据世界甜樱桃生产和贸易情况,指出了我国在甜樱桃科学技术与生产上与世界先进甜樱桃生产国的差距,分析了我国加入WTO之后,我国甜樱桃面临的机遇和挑战,并且机遇大于挑战。如果有适当的科技投入,我国甜樱桃产业将可克服现在科技滞后的状态,发挥我国劳动力低廉和国土辽阔的优势,迅速扩大其生产总量,提高品质,扩大出口,增加农民的收入,2015年可望形成32.4~43.2亿元的社会总产值。 相似文献

AIM: To investigate the effects of PD98059 on the differentiation from mesenchymal stem cells to osteoblasts.METHODS: hMSC were separated from human marrow and expanded in cuture medium. hMSC were induced with dexamethasone, β-glycerophosphate, vitamin C which acted as osteoblast differentiation inducer. PD98059 was added into the osteoblasts induction medium. The cells were assayed with cell morphology, alkaline phosphatase (AP) activity and calcium deposition. RESULTS: The isolated cultured MSC comprised a single phenotypic population and displayed a fibroblast-like morphology. After induced with osteoblasts induction medium, the cells showed changes in cell morphology from spindle-shape to cuboidal and polygonal. The AP activity increased gradually and reached the peak in 12 days, then decreased. Many scattered tangerice calcium nodes were observed. PD 98059 significantly inhibited AP activity and calcium deposition in a dose-dependent manner. A striking observation of the present study was that a few adipocytes appeared in cultures that were treated with PD 98059 and osteogenic differentiation medium. CONCLUSION: These results indicated that osteogenic diferentiation from the hMSCs was related to the activation of the ERK. 相似文献

引起糖甜菜细菌性叶斑病的萎蔫短小杆菌新致病变种 总被引:3,自引:0,他引:3
1995年在内蒙古临河市新发现了糖甜菜细菌性叶斑病,从病斑所分离的10个细菌菌株经柯赫氏法则验证,均确系该病的病原菌。采用形态观察、表型特征和生理生化特性测定、数值分析、血清学反应、细胞化学成分分析、DNA G+C mol%和DNA-DNA同源性测定进行了鉴定,并与植物病原棒形细菌15个标准菌株进行了比较。该病原菌为革兰氏阳性细菌,不规则短杆状,有一根鞭毛、亚极生或侧生,结合其生理生化特性、细胞化学成分和DNA G+C mol%和DNA-DNA同源性测定结果,认为应属于短小杆菌属(Curtobacterium)的萎蔫短小杆菌(Cur. flaccumfaciens),数值分析也支持这一结论。此外,据血清学反应结果及其对短小杆菌属的其它植物寄主的致病情况,认为该病原菌应是萎蔫短小杆菌种下的一个新的致病变种,定名为Curtobacterium flaccumfaciens pv. beticola pv. nov. 菜豆萎蔫病菌的血清检测鉴定技术研究 总被引:2,自引:0,他引:2
本研究对我国一类检疫性有害生物菜豆萎蔫病菌(Curtobacteriumflacumfa-cienspv.flacumfaciens,Cf)进行血清检测鉴定技术的研究。试验中制备了抗Cf的血清,应用抗0020,0237,0238菌株的抗血清及其FITC标记的抗体进行间接和直接免疫荧光染色试验(IF/IF)。通过对19个Cf菌株和其它6个属14个种的植物病原细菌20个菌株的测定,结果发现约50%的Cf菌株为强阳性反应,且与Cf种下致病变种也有强阳性交叉反应。由于单个血清检测易产生假阴性反应,故将三种血清1∶1∶1混合后进行IF试验,结果表明与所有的Cf菌株反应,且与Rfa和Cmt有弱阳性反应。对含104~107cfu/ml细菌及30g种子含1粒人工接种种子的种子抽提液均能检测到典型的阳性细胞,血清检测的灵敏度为4.5×104cfu/ml,可用于种子的检测。本研究还进行了初步的免疫荧光菌落染色(IFC)试验 相似文献
