Helicobacter (H.) suis colonizes the stomach of pigs and is the most prevalent gastric non-H. pylori Helicobacter species in humans. Limited information is available on host immune responses after infection with this agent and it is unknown if variation in virulence exists between different H. suis strains. Therefore, BALB/c and C57BL/6 mice were used to compare colonization ability and gene expression of various inflammatory cytokines, as determined by real-time PCR, after experimental infection with 9 different H. suis strains. All strains were able to persist in the stomach of mice, but the number of colonizing bacteria at 59 days post inoculation was higher in stomachs of C57BL/6 mice compared to BALB/c mice. All H. suis strains caused an upregulation of interleukin (IL)-17, which was more pronounced in BALB/c mice. This upregulation was inversely correlated with the number of colonizing bacteria. Most strains also caused an upregulation of regulatory IL-10, positively correlating with colonization in BALB/c mice. Only in C57BL/6 mice, upregulation of IL-1β was observed. Increased levels of IFN-γ mRNA were never detected, whereas most H. suis strains caused an upregulation of the Th2 signature cytokine IL-4, mainly in BALB/c mice. In conclusion, the genetic background of the murine strain has a clear impact on the colonization ability of different H. suis strains and the immune response they evoke. A predominant Th17 response was observed, accompanied by a mild Th2 response, which is different from the Th17/Th1 response evoked by H. pylori infection.  相似文献

Bovine tuberculosis surveillance alternatives in Belgium   总被引:1,自引:0,他引:1  
Belgium obtained the bovine tuberculosis (bTB) officially free status in 2003 (EC Decision 2003/467/EC). This study was carried out to evaluate the components of the current bTB surveillance program in Belgium and to determine the sensitivity of this program. Secondly, alternatives to optimize the bTB surveillance in accordance with European legislation (Council Directive 64/432/EEC) were evaluated. Separate scenario trees were designed for each active surveillance component of the bTB surveillance program. Data from 2005 to 2009 regarding cattle population, movement and surveillance were collected to feed the stochastic scenario tree simulation model. A total of 7,403,826 cattle movement history records were obtained for the 2,678,020 cattle from 36,059 cattle herds still active in 2009. The current surveillance program sensitivity as well as the impact of alternative surveillance protocols was simulated in a stochastic model using 10,000 iterations per simulation. The median (50% percentile) of the component sensitivities across 10,000 iterations was 0.83, 0.85, 0.99, 0.99, respectively, for (i) testing the cattle only during the winter screening, (ii) testing only imported cattle, (iii) testing only purchased cattle and (iv) testing only all slaughtered cattle. The sensitivity analysis showed that the most influential input parameter explaining the variability around the output came from the uncertainty distribution around the sensitivity of the diagnostic tests used within the bTB surveillance. Providing all animals are inspected and post mortem inspection is highly sensitive, slaughterhouse surveillance was the most effective surveillance component. If these conditions were not met, the uncertainty around the mean sensitivity of this component was important. Using an antibody ELISA at purchase and an interferon gamma test during winter screening and at import would increase greatly the sensitivity and the confidence level of Belgium's freedom from bTB infection status.  相似文献

The aim of conservation agriculture (CA) is to improve soil quality and crop yield whilst reducing runoff and topsoil erosion. An experiment was carried out in a rainfed field using a permanent raised bed planting system for 3 yr (2005–2007) in Adigudem, northern Ethiopia in order to evaluate the effect of CA on runoff, soil loss and crop yield. CA practices were introduced in fields with Vertisols in a randomized complete block design on permanent 5 × 19 m plots. Three treatments were evaluated: (1) conventional tillage (CT) with a minimum of three tillage operations and removal of crop residues, (2) terwah (TER) that was similar to CT except that contour furrows were included at 1.5 m intervals, and (3) derdero+ (DER+), which consists of permanent raised beds with a furrow and bed system, retention of 30% of standing crop residues and zero tillage on the top of the bed. All ploughing as well as the maintenance of the furrows of the permanent raised beds was done using a local ard plough called maresha. Results from monitoring over 3 yr showed that soil loss and runoff were significantly higher (P < 0.05) in CT followed by TER and DER+. Average soil losses of 5.2, 20.1 and 24.2 t/ha were recorded from DER+, TER and CT, respectively. Runoff was 46.3, 76.3 and 98.1 mm from DER+, TER and CT, respectively. Grain yield was significantly lower (P < 0.05) in DER+ under teff in 2006, probably due to the high sensitivity of teff to weeds. The yield of wheat in 2007 was significantly higher in DER+ followed by TER. The terwah system is recommended as a first measure for wider adoption to reduce runoff and soil loss and to increase crop yield. The long‐term goal is to achieve a derdero+ system, i.e. a permanent raised bed planting system along with the application of crop residues.  相似文献

Differences in hardness of wheat cultivars have been related to differences in interactions between the starch granule surface and the gluten protein matrix that are mediated by the proteins puroindoline (PIN) A and B. We examined whether or not PINs and (polar) lipids are associated at the starch granule surface, and, if so, how they interact with the starch granule surface itself. Starch was isolated from a soft wheat cultivar containing both wild‐type PINs and incubated with peptidases or lipases, or in extraction media (typically used for defatting). Protein, PIN, and lipid levels revealed that PINs and lipids are tightly associated together at the starch granule surface. Our results imply that PINs need lipids for binding to the granule surface but not vice versa.  相似文献

Pyranose oxidase (P2O) improves wheat flour dough stability and bread quality. We related its effect on dough spread behavior to that on dough and bread crumb structure. Increasing P2O addition levels gradually reduced dough flow. High P2O addition levels further increased dough strength, significantly increased dough cell wall thickness, and decreased bread loaf volume. Taken together, affecting dough spread behavior impacts dough and bread (crumb) structure, and dough structure largely determines bread crumb structure.  相似文献

In 1922, the colonial government introducedAcacia mearnsii in the tobacco-growing region of Wonosobo. Soon this species was accepted by the local people who developed an agroforestry system based on a rotation ofA. mearnsii and agricultural crops.Now, the growing of the black wattle is becoming less popular. To study the prospects for this agroforestry system 143 farmers from 5 villages were interviewed. The functions of this agroforestry system are the production of fuelwood and bark (which contains tannin for the leather industry), soil conservation and soil improvement. The following factors affectA. mearnsii growing: population density (which affects farm size and demand for domestic fuelwood), tobacco-processing (which requires fuel), topography and commercialization. If current trends continue, the contribution of the functions of this agroforestry system to welfare will not be sufficient to compete successfully with subsistence and cash crops like potatoes and tobacco. ABSTRACT: Helicobacter (H.) suis has been associated with chronic gastritis and ulcers of the pars oesophagea in pigs, and with gastritis, peptic ulcer disease and gastric mucosa-associated lymphoid tissue lymphoma in humans. In order to obtain better insight into the genes involved in pathogenicity and in the specific adaptation to the gastric environment of H. suis, a genome analysis was performed of two H. suis strains isolated from the gastric mucosa of swine. Homologs of the vast majority of genes shown to be important for gastric colonization of the human pathogen H. pylori were detected in the H. suis genome. H. suis encodes several putative outer membrane proteins, of which two similar to the H. pylori adhesins HpaA and HorB. H. suis harbours an almost complete comB type IV secretion system and members of the type IV secretion system 3, but lacks most of the genes present in the cag pathogenicity island of H. pylori. Homologs of genes encoding the H. pylori neutrophil-activating protein and γ-glutamyl transpeptidase were identified in H. suis. H. suis also possesses several other presumptive virulence-associated genes, including homologs for mviN, the H. pylori flavodoxin gene, and a homolog of the H. pylori vacuolating cytotoxin A gene. It was concluded that although genes coding for some important virulence factors in H. pylori, such as the cytotoxin-associated protein (CagA), are not detected in the H. suis genome, homologs of other genes associated with colonization and virulence of H. pylori and other bacteria are present.  相似文献

A simulation model for the production of the Nile tilapia (Oreochromis niloticus niloticus L.) for mixed‐ or mono‐sex culture and for poly‐culture with the African catfish (Clarias gariepinus Burchell 1822) or African snakehead (Parachanna obscura Günther 1861) is presented. The model is based on an exponential decay model used in population dynamics and follows an individual‐based approach. The model consists of a tilapia sub‐model representing population dynamics, growth and recruitment and a predator sub‐model representing the predation process and population dynamics and growth of the used predator. The model was calibrated with data on mixed‐ or mono‐sex culture of Nile tilapia and for poly‐culture with the African catfish or African snakehead obtained in Congo Brazzaville and validated with similar data from the Philippines, Thailand and the Ivory Coast. The model visualized major underlying processes in tilapia farming and aspects for further improvement of the model; growth is one of the most sensitive input parameters and should be quantitatively related to feeding level and feed quality; length at first maturity and quantification of the recruitment of Nile tilapia has a relatively large influence and recruitment should be related to the length of the females; prey–predator relations are too coarse and should be more fine‐tuned with the relation between prey size and predator size. Incorporation of these features would provide the basis of a model that can serve as a predictive and decision‐making support tool.  相似文献
