The prevalence of capsular and somatic serotypes were studied among 123 Pasteurella multocida strains isolated from chickens (n = 94), ducks (22), quails (4), turkeys (2) and geese (1) from different geographical regions of India. All strains exhibited similar cultural and morphological characteristics. Ninety-two of the isolates belonged to serotype A:1, the most prevalent serotype, with serotypes A:3, A:1,3, D:3 and F:3 having two isolates each. Only one isolate was positive for serotypes A:4 and D:1. Twenty isolates were untyped. A multiplex capsular PCR assay generated amplicons of sizes 460, 1044, 657 and 854 bp in 106 isolates identified as capsular serotype-A, 15 in serotype D and two in serotype F. Capsular types B and E were not detected in any of the avian isolates studied. The present findings suggest that a multiplex capsular PCR assay may be suitable for the rapid initial identification serotypes P. multocida during epidemiological studies of fowl cholera. 相似文献

Cadmium Enhances Generation of Hydrogen Peroxide and Amplifies Activities of Catalase, Peroxidases and Superoxide Dismutase in Maize 总被引:2,自引:0,他引:2
Maize (Zea mays L. cv. 777) plants grown in hydroponic culture were treated with 50 μm CdSO4. Growth and metabolic parameters indicative of oxidative stress and antioxidant responses were studied in leaves of plants treated with Cd. Apart from increasing lipid peroxidation and H2O2 accumulation, supply of Cd suppressed growth, fresh and dry mass of plants and decreased the concentrations of chloroplastic pigments. The activities of catalase (CAT; EC 1.11.1.6), peroxidase (POD; EC 1.11.1.7), ascorbate peroxidase (APX; EC 1.11.1.11) and superoxide dismutase (SOD; EC 1.15.1.1) were increased in plants supplied 50 μm Cd. Localization of activities of isoforms of these enzymes (POD, APX and SOD) on native gels also revealed increase in the intensities of pre‐existing bands. Stimulated activities of CAT, POD, APX and SOD in maize plants supplied excess Cd do not appear to have relieved plants from excessive generation of reactive oxygen species (ROS). It is, therefore, concluded that supply of 50 μm Cd induces oxidative stress by increasing production of ROS despite increased antioxidant protection in maize plants. 相似文献

The aim of this research was designed in order to optimize integrated plant nutrient supply (IPNS) through balanced fertilization of organic, inorganic and microbial inoculants in strawberry cv. Chandler. The potential efficiency of bio-organics used along with chemical fertilizers on cropping behaviour, physical-chemical and biological properties of rhizosphere soil, fruit yield, quality attributes and leaf nutrient content was investigated. The significant improvement in physico-chemical properties of the soil and nutrient uptake was recorded. The uninoculated control received farmyard manure (FYM) and inorganic nitrogen (N) recorded the highest cation exchange capacity (CEC) and soil organic carbon (OC) content. Highest available N and phosphorus (P) of soil were recorded in vermicompost and inorganic N applied in two and one split, respectively. The concentration of micronutrients cations viz., iron (Fe), zinc (Zn), manganese (Mn) in soil was higher in treatment received vermicompost and inorganic N in two splits. The integration of bio-organic nutrient supplements also significantly enriched the microbial status of the rhizosphere soil, leaf nutrient concentration and maintained soil health and productivity on long term basis for sustainable fruit production. 相似文献

of an effective subunit vaccine against brucellosis is an area of intense research. The outer membrane proteins (OMPs) of
Brucella species have been extensively studied for its immunogenicity and protective ability. We have investigated the potential of
CpG ODN to enhance the immunogenicity and protective efficacy of recombinant 28 kDa outer membrane protein (rOMP28) of Brucella melitensis. The study demonstrated vigorous immunoglobulin G (IgG) response of OMP28. The administration of rOMP28 with CpG caused increased
cell mediated immune response in terms of induced IgG2a, T-cell proliferation and up-regulation of type I cytokine expression.
In contrast, the free antigen suppressed the interferon gamma (type I cytokine) production on in-vitro stimulation of spleenocytes.
The result indicates the role of OMP28 in the down regulation of IFN-γ production. Moreover, the B. abortus S-19 vaccinated mice showed highest production of IL-4 and IFN-γ. The protective ability of the antigen was evaluated by
systemic bacterial clearance after challenging the mouse with B. abortus 544 pathogen. The level of protection was significant in rOMP28+CpG treated mice but was lower than the required level. The
results of the present study indicate that rOMP28 could be an immunogen capable of inducing both humoral and cellular immune

The effects of extender composition, cryoprotectant concentration and freezing and thawing on the fertilization efficiency of cryopreserved spermatozoa of Puntius gonionotus were evaluated. Computer‐aided motility analysis of semen was conducted to check the suitability of spermatozoa for cryopreservation after mixing with different extenders and cryoprotective agents (CPAs). Extender‐4 with an osmolality 260 mOsmol kg−1and pH 7.6 was used for the cryopreservation study. Among the CPAs, dimethyl sulphoxide (DMSO) was least toxic and more than 60% fertilization was achieved when used at 1.4 M at 0 °C for 10 and 30 min, whereas the toxicity of all CPAs to spermatozoa was evident when tested at 30 °C. Semen frozen at −16 °C min−1 with 1.4 M DMSO showed 70% fertilization, which was significantly higher (P<0.05) than other freezing rates. Samples thawed at 35 °C water showed a fertilization rate comparable with that of fresh semen. Computer‐assisted semen analysis of fresh and frozen semen after thawing showed variations in different types of motility in spermatozoa and in their class. There was no significant difference in motility before or after cryopreservation; however, significant differences could be observed in the average path velocity (VAP), straight line velocity (VSL) and curve linear velocity (VCL). Semen of silver barb could be cryopreserved with extender‐4 by addition of 1.4 M DMSO to a final cryopreservation medium (MED 2) cooled at a rate of −16 °C min−1, stored in liquid nitrogen (−196 °C) and utilized after thawing at 35±2 °C. 相似文献

The health of the pond environment in shrimp farming is important for sustainable and profitable aquaculture, in which sediment–water interface is the most important influencing area. With this objective, the key parameters of water and sediment at the interface was studied in shrimp ponds with varying salinities and compared with the surface water and soil to understand the variations and the underlying causes. Total ammonia nitrogen and total alkalinity were higher in water at the interface as compared to surface water, whereas pH, nitrite, oxide forms of nitrogen (NO3), phosphorus (PO4) and oxidizing bacteria were observed to be lower. There was no significant difference with respect to macrominerals such as sodium, potassium, calcium and magnesium between water at the interface and surface. These changes are profoundly influenced by the uppermost 1.0‐cm thick sediment layer, where the maximum transition of nutrients takes place with the water column. The outcome of the study highlights the effectiveness of water sampling for metabolites at the interface, which reflects at an early stage any potential deterioration of pond environment, which will help the farmer to initiate timely mitigation measures. 相似文献

Grain legumes, the important constituents of sustainability‐based cropping systems and energy‐limited vegetarian diets have long been the subject of scientific research. Tremendous technological strides were made in the so‐called orphan crops, in terms of both varietal improvement and generation of basic information. Despite recalcitrancy and high genotype dependency, in vitro culture techniques such as organogenesis, in vitro mutagenesis, embryo rescue and in vitro gene transfer have been deployed for improvement of several grain legumes and these played an important role in introgression of desirable genes from related and distant species and creation of additional genetic variability. Stable and reproducible regeneration protocols resulted in the development of genetically modified chickpea, pigeon pea, cowpea, mungbean, etc., while embryo rescue was deployed successfully for recovery of interspecific recombinants, a few of them exploited for the development of commercial cultivars. Nevertheless, doubled haploidy witnessed limited success and protoplast regeneration and in vitro mutagenesis remained of academic interest. Present research has delineated the biosorption potential of three different nonliving biomasses namely eucalyptus bark saw dust, mango bark saw dust, and pineapple fruit peel with respect to Zn (II) ion removal from liquid phase through batch experiments. The efficacy of Zn (II) ion biosorption onto surface of biosorbents was judged and correlated with biosorbent particle size, surface chemistry, and surface texture. Maximum metal ion uptake capacity, percentage removal, and minimum equilibrium concentration as 1.688 mg/g, 84.4%, and 1.56 mg/l, respectively, was obtained using eucalyptus bark saw dust mediated biosorption followed by mango bark saw dust as 1.028 mg/g, 51.4%, and 4.867 mg/l and pineapple fruit peel as 0.45 mg/g, 22.9%, and 7.71 mg/l, respectively, at a particle size of 0.5 mm. Additionally, present investigation also proved that biosorption efficiency and metal ion interaction with adsorbent surface also depends upon presence of functional groups involved in metal ion adsorption and surface porosity. 相似文献
