Gynoecious is an important economic trait of cucumber for determinant of earliness and yield, yet genetic mechanism is not well understood for this trait. The experiment was conducted using F2 mapping population by crossing of PPC-2, a gynoecious and parthenocarpic line with Pusa Uday (monoecious and non-parthenocarpic cultivar). Out of 179 SSR markers screened, 39 markers differentiated the gynoecious and monoecious parents. However, only 17 markers were segregating with F2 mapping population, those were used for genotyping and linkage map analysis and these markers were placed along with F locus on chromosome 6 covering a total distance of 100.4cM. The SSR markers, SSR13251 and UW020605 were found to be closely linked to gynoecious (F) locus at 1.0 and 4.5 cM, respectively. The segregation of F2 population of PPC-2 × Pusa Uday and GPC-1 × Punjab Naveen and test crosses for sex type herein suggested that single dominant gene controlled the gynoecious sex expression in cucumber particularly in gynoecious genotypes PPC-2 and GPC-1. Therefore, the monogenic dominant nature of gynoecious sex identified in the present experiment and SSR markers closely linked to the F locus will be useful in marker-assisted backcross breeding for transfering gynoecious trait into horticulturally desirable varieties. 相似文献

Iron deficiency chlorosis is an important abiotic stress affecting groundnut production worldwide in calcareous and alkaline soils with a pH of 7.5–8.5. To identify genomic regions controlling iron deficiency chlorosis resistance in groundnut, the recombinant inbred line population from the cross TAG 24 × ICGV 86031 was evaluated for associated traits like visual chlorosis rating and SPAD chlorophyll meter reading across three crop growth stages for two consecutive years. Thirty-two QTLs were identified for visual chlorosis rating (3.9%–31.8% phenotypic variance explained [PVE]) and SPAD chlorophyll meter reading [3.8%–11% PVE] across three stages over 2 years. This is the first report of identification of QTLs for iron deficiency chlorosis resistance-associated traits in groundnut. Three major QTLs (>10% PVE) were identified at severe stage, while majority of other QTLs were having small effects. Interestingly, two major QTLs for visual chlorosis rating at 60 days (2013) and 90 days (2014) were located at same position on LG AhXIII. The aroma compositions of Fragaria ananassa varieties are assessed by purge-and-trap high-resolution gas chromatography. Gas chromatography/mass spectrometry and gas chromatography/Fourier transform infrared spectroscopy allow the identification of 93 components from which 21 are for the first time described as constituents of strawberry aroma. Despite the complexity of the aroma, sensory properties are assigned to 40 components as perceived by three independent testers by application of a sniffing technique to the chromatographic effluent. Heavy metals show a wide range of effect on fishes, out of which arsenic (As) and lead (Pb) are among the leading heavy metal toxicants. These heavy metals are known to alter different biochemical parameters, including glycogen level, in different tissues of fishes. Glycogen level in fish serves as the main source of energy; hence, in this study, the acute toxicity test of As and Pb and their effect on the glycogen content and the enzymes involved therein (glycogen phosphorylase, glycogen synthase, hexokinase, phosphofructokinase and pyruvate kinase) were studied in the liver and muscle tissues of Heteropneustes fossilis. The 96 h LC50 values of As2O3 and PbCl2 on H. fossilis were found to be 35.09 ppm and 66.20 ppm, respectively. On acute exposure to 96 h LC50 values of As2O3 and PbCl2, the glycogen concentration showed a gradual decrease in both liver and muscle tissues of H. fossilis. However, on chronic exposure (LC50/20th ppm), the glycogen content in liver and muscle of H. fossilis was depleted till 20 days; whereas after 30 days, the glycogen level was recovered in both the tissues. The activities of glycogen metabolic enzymes (glycogen phosphorylase and glycogen synthase) and few selected glycolytic enzymes (hexokinase, phosphofructokinase and pyruvate kinase) were also altered in H. fossilis when exposed to acute and chronic concentration of As2O3 and PbCl2. Our present results showed that As and Pb induced toxicity stress on the catfish, H. fossilis, which might have caused to alter the carbohydrate metabolism in the fish. 相似文献

Teleost fish have developed their own specific adaptive mechanism, both behavioral and physiological, to maintain homeostasis
in response to unfavorable temperatures. Therefore, this study was aimed at assessing the critical thermal maxima (CTMax), critical thermal minima (CTMin), and oxygen consumption rate of Anabas testudineus (17.03 ± 1.2 g) after acclimating to three preset temperatures (25, 30, and 35°C) for 30 days. The CTMax and CTMin were 40.15, 41.40, 41.88°C and 12.43, 13.06, 13.94°C, respectively, and were significantly different (P < 0.05). The thermal tolerance polygon for the specified temperatures was 278.30°C2. The oxygen consumption rate (117.03, 125.70, 198.48 mg O2 kg−1 h−1, respectively) increased significantly (P < 0.05) with increasing acclimation temperatures. The compatibility of olive barb, Puntius sarana (Hamilton) with major carps was studied in grow-out carp polyculture system for one year in a set of nine earthen ponds of 0.08 ha each. Three different species combinations evaluated were Control: catla (Catla catla Ham.), silver carp (Hypophthalmichthys molitrix Valenciennes), rohu (Labeo rohita Hamilton) and mrigal (Cirrhinus mrigala Hamilton) at 0.5:0.5:1:1; T1: catla, silver carp, rohu and olive barb at 0.5:0.5:1:1 and T2: catla, silver carp, mrigal and olive barb at 0.5:0.5:1:1 at combined density of 7500 fingerlings/ha. While survival levels of the carps did not differ significantly in treatments (P > 0.05), silver carp recorded highest survival levels (94–96%) followed by olive barb (87–90%), mrigal (72–74%), rohu (72–73%) and catla (67–69%). The specific growth rate (SGR) and average harvested body weight (ABW) of catla and silver carp did not differ significantly among the treatments revealing their competition with mrigal or olive barb to be minimum. In absence of rohu in T2, both mrigal and olive barb showed higher SGR and ABW revealing minimal competition between these two species, while their lower performance in presence of rohu in Control and T1 indicated inter-specific competition with the latter. Such olive barb–rohu inter-specific competition, however, failed to yield significant effect on growth of rohu as revealed from its non-significant SGR difference in presence and absence of olive barb. The lower FCR (2.54 ± 0.06) and higher treatment biomass production (3418.4 ± 95.0 kg ha− 1 year− 1) in T1 with rohu–olive barb combination compared to T2 with mrigal–olive barb (2.84 ± 0.11; 3155.1 ± 104.7 kg ha− 1 year− 1) indicated feasibility and advantage of culturing rohu with olive barb rather than mrigal in carp polyculture. RAPD and microsatellite markers were used to determine the genetic fidelity of micropropagated plants from the three varieties of tea plant derived from explants of field grown mother bush as well as in vitro germinated seedlings. Rate of shoot multiplication was better from nodal explants than from shoot tips. A maximum of 32–33 shoots was observed in cotyledonary node in 1/2 MS medium with BAP (6 mg/l), GA3 (1 mg/l) and IBA (0.5 mg/l). 90% of the in vitro derived microshoots were micrografted into rootstocks. The micropropagated plantlets showed both cytological and genetical stability. SSR primers showed complete stability among the regenerants. The results convinced that plants derived from axillary as well as adventitious mode of propagation can be genetically true to type.
