Dietary guar gum and cellulose were studied as purified soluble and insoluble nonstarch polysaccharide (NSP) sources, respectively. A control diet containing 14% cornstarch was formulated. A 7% guar gum, a 7% cellulose, and a 7% guar gum + 7% cellulose diet were formulated by adding the NSP to the control diet at the expense of cornstarch (wt/wt), forming a 2 x 2 factorial arrangement. The objectives were to determine whether guar gum and cellulose altered 1) the passage rate of digesta through the small intestine and total tract; 2) the digestibility of energy and CP, characteristics of the digesta, and microbial populations in the ileum; 3) plasma glucose and ghrelin concentrations; and 4) short-term voluntary feed intake and growth performance of grower pigs. In Exp. 1, 12 pigs (27.0 +/- 1.5 kg of BW) were fitted with an ileal T-cannula and were used in a 2-period change-over design, providing 6 observations per diet. Each period included 18 d: a 12-d acclimation period followed by 2-d feces, 3-d digesta, and 1-d venous blood collection periods. In Exp. 1, guar gum and cellulose slowed the passage rate of digesta through the small intestine by 26 and 18%, respectively (P < 0.05). Guar gum increased total tract retention time of the digesta by 14% (P < 0.05). Guar gum and cellulose increased the viscosity of ileal digesta by 72 and 76%, respectively (P < 0.05). Cellulose reduced ileal energy and CP digestibility (P < 0.05), but guar gum only tended to decrease ileal energy digestibility (P < 0.10). Guar gum and cellulose reduced total tract energy and CP digestibility (P < 0.05). At 60 min after feeding, guar gum decreased plasma glucose by 10% (P < 0.10). Guar gum interacted with cellulose to reduce plasma ghrelin before and after feeding (P < 0.05). Guar gum and cellulose interacted to increase ileal bifidobacteria and enterobacteria (P < 0.05); however, guar gum, but not cellulose, increased ileal clostridia (P < 0.05). In Exp. 2, 20 individually housed grower pigs (5 pigs per diet) had free access to the 4 diets used in Exp. 1 for 14 d. Guar gum and cellulose decreased ADG and reduced ADFI on d 0 to 14 (P < 0.05). In summary, increasing purified NSP in the diet reduced the passage rate of digesta, energy and protein digestibility, and feed intake, but increased ileal bifidobacteria and enterobacteria populations. The effects of cellulose were similar to those of guar gum. In conclusion, monitoring of dietary NSP is a critical factor to achieve predictable digestible nutrient intake and intestinal bacterial populations.  相似文献

ABSTRACT: Equine herpesvirus-1 (EHV-1) infection remains a significant problem despite the widespread use of vaccines. The inability to generate a protective immune response to EHV-1 vaccination or infection is thought to be due to immunomodulatory properties of the virus, and the ORF1 and ORF2 gene products have been hypothesized as potential candidates with immunoregulatory properties. A pony infection study was performed to define immune responses to EHV-1, and to determine if an EHV-1 ORF1/2 deletion mutant (ΔORF1/2) would have different disease and immunoregulatory effects compared to wild type EHV-1 (WT). Infection with either virus led to cytokine responses that coincided with the course of clinical disease, particularly the biphasic pyrexia, which correlates with respiratory disease and viremia, respectively. Similarly, both viruses caused suppression of proliferative T-cell responses on day 7 post infection (pi). The ΔORF1/ORF2 virus caused significantly shorter primary pyrexia and significantly reduced nasal shedding, and an attenuated decrease in PBMC IL-8 as well as increased Tbet responses compared to WT-infected ponies. In conclusion, our findings are (i) that infection of ponies with EHV-1 leads to modulation of immune responses, which are correlated with disease pathogenesis, and (ii) that the ORF1/2 genes are of importance for disease outcome and modulation of cytokine responses.  相似文献

Real-time quantitative PCR (RT-qPCR) is a critical tool used to evaluate changes in gene expression. The precision of this tool is reliant upon the selection of reference genes whose expression remains unaltered in culture conditions and following stimulation. Stably expressed reference genes are used to normalize data so observed changes in expression are not due to artifacts but rather reflect physiological changes. In this study, we examined the expression stability of the porcine genes glyceraldehyde 3-phosphate dehydrogenase (GAPDH), succinate dehydrogenase complex subunit A (SDHA), eukaryotic elongation factor 1 gamma-like protein (eEF1), ribosomal protein L19 (RPL19), beta-actin (ACTB) and ATP synthase mitochondrial F0 complex (ATP5G1) in peripheral blood mononuclear cells (PBMCs), monocytes, monocyte-derived dendritic cells (MoDCs), blood isolated dendritic cells (BDCs) and T cells with or without stimulation with lipolysaccharide (LPS). An M value was used as a measure of gene stability as determined using geNORM software. Recommendations for the use of reference genes include using GAPDH and B-actin in PBMCs: RPL19 and SDHA in T cells; RPL19 and B-actin in monocytes; RPL-19 and SDHA in BDCs: and RPL-19 and ATP5GA in MoDCs.  相似文献

This report describes a unique case of presumed migration of Parelaphastrongylus tenuis through the spinal cord into the eye of a llama where it survived and matured within the ocular environment. Medium-chain fatty acids (MCFAs) are found at higher levels in milk lipids of many animal species and in the oil fraction of several plants, including coconuts, palm kernels and certain Cuphea species. Medium-chain triglycerides (MCTs) and fatty acids are efficiently absorbed and metabolized and are therefore used for piglet nutrition. They may provide instant energy and also have physiological benefits beyond their energetic value contributing to several findings of improved performance in piglet-feeding trials. MCTs are effectively hydrolyzed by gastric and pancreatic lipases in the newborn and suckling young, allowing rapid provision of energy for both enterocytes and intermediary hepatic metabolism. MCFAs affect the composition of the intestinal microbiota and have inhibitory effects on bacterial concentrations in the digesta, mainly on Salmonella and coliforms. However, most studies have been performed in vitro up to now and in vivo data in pigs are still scarce. Effects on the gut-associated and general immune function have been described in several animal species, but they have been less studied in pigs. The addition of up to 8% of a non-esterified MCFA mixture in feed has been described, but due to the sensory properties this can have a negative impact on feed intake. This may be overcome by using MCTs, allowing dietary inclusion rates up to 15%. Feeding sows with diets containing 15% MCTs resulted in a lower mortality of newborns and better development, particularly of underweight piglets. In conclusion, MCFAs and MCTs offer advantages for the improvement of energy supply and performance of piglets and may stabilize the intestinal microbiota, expanding the spectrum of feed additives supporting piglet health in the post-weaning period.  相似文献

Human probes identifying the cellular homologs of the v-ets gene, Hu-ets-1 and Hu-ets-2, and two panels of rodent-human cell hybrids were used to study specific translocations occurring in acute leukemias. The human ets-1 gene was found to translocate from chromosome 11 to 4 in the t(4;11)(q21;23), a translocation characteristic of a subtype of leukemia that represents the expansion of a myeloid/lymphoid precursor cell. Similarly, the human ets-2 gene was found to translocate from chromosome 21 to chromosome 8 in the t(8;21)(q22;q22), a nonrandom translocation commonly found in patients with acute myeloid leukemia with morphology M2 (AML-M2). Both translocations are associated with expression different from the expression in normal lymphoid cells of ets genes, raising the possibility that these genes play a role in the pathogenesis of these leukemias.  相似文献

Our current understanding of the mechanisms driving spatiotemporal yield variability in rice systems is insufficient for effective management at the sub-field scale. The overall objective of this study was to evaluate the potential of precision management for rice production. The spatiotemporal properties of multiyear yield monitor data from four rice fields, representing varying soil types and locations within the primary rice growing region in California, were quantified and characterized. The role of water management, land-leveling, and the spatial distribution of soil properties in driving yield heterogeneity was explored. Mean yield and coefficient of variation at the sampling points within each field ranged from 9.2 to 12.1 Mg ha?1 and from 7.1 to 14.5 %, respectively. Using a k-means clustering and randomization method, temporally stable yield patterns were identified in three of the four fields. Redistribution of dissolved organic carbon, nitrogen, potassium and salts by lateral flood water movement was observed across all fields, but was only related to yield variability via exacerbating areas with high soil salinity. The effects of cold water temperature and land-leveling on yield variability were not observed. Soil electrical conductivity and/or plant available phosphorus were identified as the underlying causes of the within-field yield patterns using classification and regression trees. Our results demonstrate that while the high temporal yield variability in some rice fields does not permit precision management, in other fields exhibiting stable yield patterns with identifiable causes, precision management and modified water management may improve the profitability and resource-use efficiency of rice production systems.  相似文献
