A radioassay was used to demonstrate the high affinitiy of a crude rat uterine extract for tritiated estradiol-17B. Although no other chemical procedures were employed to further characterize the extract, it may contain the estrogen binding protein isolated by others. The isolate was antigenic in rabbits and administration of immune serum to female rats significantly lowered the in vitro retention of tritiated estradiol-17B in the rat uterus. It is thought that this immune serum could be successfully used to study the estrogen binding activity of rat uterus. 相似文献

Vaccines protect chickens against H5 highly pathogenic avian influenza in the face of genetic changes in field viruses over multiple years 总被引:7,自引:0,他引:7
Inactivated whole avian influenza (AI) virus vaccines, baculovirus-derived AI haemagglutinin vaccine and recombinant fowlpoxvirus-AI haemagglutinin vaccine were tested for the ability to protect chickens against multiple highly pathogenic (HP) H5 AI viruses. The vaccine and challenge viruses, or their haemagglutinin protein components, were obtained from field AI viruses of diverse backgrounds and included strains obtained from four continents, six host species, and isolated over a 38-year-period. The vaccines protected against clinical signs and death, and reduced the number of chickens shedding virus and the titre of the virus shed following a HP H5 AI virus challenge. Immunization with these vaccines should decrease AI virus shedding from the respiratory and digestive tracts of AI virus exposed chickens and reduce bird-to-bird transmission. Although most consistent reduction in respiratory shedding was afforded when vaccine was more similar to the challenge virus, the genetic drift of avian influenza virus did not interfere with general protection as has been reported for human influenza viruses. 相似文献

Rice endosperm protein was modified to enhance solubility and emulsifying properties by controlled enzymatic hydrolysis. The optimum degree of hydrolysis (DH) was determined for acid, neutral, and alkaline type proteases. Solubility and emulsifying properties of the hydrolysates were compared and correlated with DH and surface hydrophobicity. DH was positively associated with solubility of resulting protein hydrolysate regardless of the hydrolyzing enzyme, but enzyme specificity and DH interactively determined the emulsifying properties of the protein hydrolysate. The optimum DH was 6–10% for good emulsifying properties of rice protein, depending on enzyme specificity. High hydrophobic and sulfhydryl disulfide (SH-SS) interactions contributed to protein insolubility even at high DH. The exposure of buried hydrophobic regions of protein that accompanied high-temperature enzyme inactivation promoted aggregation and cross-linking of partially hydrolyzed proteins, thus decreasing the solubility and emulsifying properties of the resulting hydrolysate. Due to the highly insoluble nature of rice protein, surface hydrophobicity was not a reliable indicator for predicting protein solubility and emulsifying properties. The antimicrobial activity and mode of action of chitosan were evaluated against Streptococcus iniae, a pathogenic Gram‐positive bacterium of fish worldwide. Cell proliferation kinetics were examined following exposure to varying concentrations of chitosan. The action of chitosan on S. iniae was also investigated by measuring agglutination activity, conductivity, and extracellular and intracellular bacterial adenosine triphosphate (ATP) levels. Chitosan exhibited antibacterial activity against S. iniae at concentrations of 0.1% and above and was lethal at a concentration of 0.4% and higher. The mechanism of antibacterial activity of chitosan at the inhibitory level of bacterial growth appears to hinge upon the interaction between chitosan and the oppositely charged bacterial surface. This interplay causes agglutination, which was readily observed grossly and microscopically. After interacting with the cell surface via adsorption, an efflux of intracellular ATP was documented, which suggests that chitosan disrupts the bacterial cell causing leakage of cytosolic contents and ultimately cell death. Results suggest chitosan may be worth evaluating as a natural alternative to antibiotic against S. iniae infection of fish. 相似文献

- 1. Despite the extensive literature on the ecology, systematics and culture of oysters worldwide, an assessment of their diversity, distribution and conservation status for the Atlantic and Caribbean coasts (i.e. depth <50 m) of South America is lacking. Such information is crucial because of the increasing coastal development that threatens most nearshore habitats throughout the region.
- 2. The available information on oysters on Atlantic and Caribbean coasts is reviewed with a focus on identifying regional conservation priorities based on ecological and socio‐economic importance, as well as the magnitude of current or potential threats faced by oyster populations. The current status of α‐ taxonomy within the Ostreidae was also examined.
- 3. Ten species of native Ostreidae (plus three introduced species) inhabit the coastal waters of the Atlantic and Caribbean coasts of South America.
- 4. Oyster species were ranked according to their biological/ecological and socio‐economic value and conservation status within 10 distinct ecoregions. Crassostrea gasar in the Eastern Brazil ecoregion, C. rhizophorae in the Central Caribbean ecoregion and Ostrea puelchana in the North Patagonian Gulfs ecoregion should receive the highest priority for immediate conservation action due to extensive loss of mangrove habitat in the two former regions and evidence of decline of one of the most important populations for the latter. The need for a standardized methodology to assess the status of oyster populations throughout the ecoregions is identified.
- 5. On a local scale, the allocation of territorial use rights for fisheries under a collaborative/voluntary community framework is strongly advocated to fulfil management, conservation and poverty alleviation goals in these developing countries.

Bird migration has long been hypothesized as the main mechanism for long‐distance dispersal of flaviviruses, but the role of migratory birds in flaviviruses spillover is not well documented. In this study, we investigated the eco‐epidemiology of West Nile virus (WNV) and Usutu virus (USUV) in trans‐Saharan passerines during their spring stopover in southern Tunisian oases. To do, we combined oral swab analysis and serological tools to assess whether migratory birds could be reaching these stopover sites while infectious or have been previously exposed to viruses. All sampled birds tested negative for oral swab analysis. However, anti‐WNV and anti‐USUV antibodies were detected in 32% and 1% of tested birds, respectively. Among WNV‐seropositive species, the Golden oriole (Oriolus oriolus) showed the highest anti‐WNV occurrence probability. In this species, anti‐WNV occurrence was twice larger in males than females. Inter‐specific and intraspecific morphological, physiological and behavioural differences could explain these results. Although our findings did not show evidence for passerines migrating while infectious, they did not exclude an existing enzootic WNV transmission cycle in Tunisian oases. Further investigations including larger samples of migratory birds are needed for a better understanding of this issue. 相似文献
