Microtubule (MT) stabilization is regulated by the small guanosine triphosphate (GTP)-binding protein Rho and its effector, mammalian homolog of Diaphanous (mDia), in migrating cells, but factors responsible for localized stabilization at the leading edge are unknown. We report that integrin-mediated activation of focal adhesion kinase (FAK) at the leading edge is required for MT stabilization by the Rho-mDia signaling pathway in mouse fibroblasts. MT stabilization also involved FAK-regulated localization of a lipid raft marker, ganglioside GM1, to the leading edge. The integrin-FAK signaling pathway may facilitate Rho-mDia signaling through GM1, or through a specialized membrane domain containing GM1, to stabilize MTs in the leading edge of migrating cells.  相似文献

Diversionary winter feeding of browsing ungulates is an increasingly common management practice although evidence for its efficacy to reduce excessive browsing remains ambiguous. Moreover, comparative estimates on changes in browsing pressure (proportion of available shoots browsed) following long-term winter feeding are currently lacking. We quantified spatiotemporal changes in browsing pressure of moose (Alces alces L.) on commercial and non-commercial tree species around 30 feeding stations after 15–20 years of winter feeding. The results were compared with browsing pressure indices recorded at the same feeding stations 10 years previously. We expected leader stem and lateral twig browsing to have increased over time at a fine spatial scale (≤200 m from feeding station) and to have increased in spatial extent away from feeding stations. Furthermore, we tested whether moose browsing patterns conformed to central-place foraging theory. Despite 2–3-fold higher faecal pellet group numbers in the vicinity of feeding stations, leader stem browsing increased only on the commercially valuable Norway spruce (Picea abies (L.) Karst), a species normally avoided by moose. Lateral twig browsing largely decreased within 200 m of feeding stations while at a broader scale (≤1 km from feeding stations), leader stem browsing was high on most tree species (ca. 60% of available stems browsed) as expected, Peak browsing on lateral twigs of Scots pine (Pinus sylvestris L.) occurred further from feeding stations after 15–20 years of winter feeding than 10 years earlier. Browsing pressure on active feeding stations (n = 18) was comparable to that on inactive feeding stations (n = 12), suggesting the occurrence of rebrowsing. The ability of central-place foraging theory to explain fine-scale browsing patterns around feeding stations decreased as diversionary feeding continued over time. Long-term diversionary winter feeding of moose in unproductive boreal forests has serious implications for the intensity of fine-scale browsing pressure, which may lead to resource depletion close to feeding stations followed by high browsing pressure at distances further away from feeding stations. Our study emphasizes the importance of considering the longevity of a winter feeding programme at its inception.  相似文献

Elevated nitrogen (N) deposition in humid tropical regions may aggravate phosphorus (P) deficiency in forest on old weathered soil found in these regions. From January 2007 to August 2009, we studied the responses of soil microbial biomass and community composition to P addition (in two monthly portions at level of 15 g P m?2 yr?1) in three tropical forests in southern China. The forests were an old-growth forest and two disturbed forests (mixed species and pine dominated). The objective was to test the hypothesis that P addition would increase microbial biomass and change the composition of the microbial community, and that the old-growth forests would be more sensitive to P addition due to its higher soil N availability. Microbial biomass C (MBC) was estimated twice a year and the microbial community structure was quantified by phospholipid fatty acid (PLFA) analysis at the end of the experiment. Addition of P significantly increased the microbial biomass and altered the microbial community composition in the old-growth forest, suggesting that P availability is one of the limiting factors for microbial growth. This was also reflected by significant increases in soil respiration after P addition. In contrast, P addition had no effect on the microbial biomass and the microbial community composition in the pine forests. Also in the mixed forest, the microbial biomass did not significantly respond to P addition, but soil respiration and the ratio of fungal-to-bacteria was significantly increased.  相似文献

The hydrogeologic conditions of groundwater can be examined by carefully studying the patterns of fluctuations in groundwater levels. These fluctuations are spatially and temporally influenced by many complicated factors, including rainfall, topography, land use, and hydraulic properties of soils and bedrock (i.e., aquifers). In this article we report a methodology based on the Bayesian logistic mixture model to simultaneously cluster profiles of groundwater level changes over time and estimate the relationships between the characteristics of each cluster and environmental variables. We apply the proposed method to analyze groundwater level profiles from 37 monitoring wells in Seoul, South Korea, and we find four clusters of wells. Using the estimated relationship between the clusters and the environmental variables, we discern the hydrogeologic conditions of each cluster, thus gaining insight into the recharge and subsurface flow of bedrock groundwater in an urban setting and the vulnerability of groundwater to the inflow of potential pollutants from ground surface. The knowledge of tree species effects on soil C and N pools is scarce, particularly for European deciduous tree species. We studied forest floor and mineral soil carbon and nitrogen under six common European tree species in a common garden design replicated at six sites in Denmark. Three decades after planting the six tree species had different profiles in terms of litterfall, forest floor and mineral soil C and N attributes. Three groups were identified: (1) ash, maple and lime, (2) beech and oak, and (3) spruce. There were significant differences in forest floor and soil C and N contents and C/N ratios, also among the five deciduous tree species. The influence of tree species was most pronounced in the forest floor, where C and N contents increased in the order ash = lime = maple < oak = beech ? spruce. Tree species influenced mineral soil only in some of the sampled soil layers within 30 cm depth. Species with low forest floor C and N content had more C and N in the mineral soil. This opposite trend probably offset the differences in forest floor C and N with no significant difference between tree species in C and N contents of the whole soil profile. The effect of tree species on forest floor C and N content was primarily attributed to large differences in turnover rates as indicated by fractional annual loss of forest floor C and N. The C/N ratio of foliar litterfall was a good indicator of forest floor C and N contents, fractional annual loss of forest floor C and N, and mineral soil N status. Forest floor and litterfall C/N ratios were not related, whereas the C/N ratio of mineral soil (0–30 cm) better indicated N status under deciduous species on rich soil. The results suggest that European deciduous tree species differ in C and N sequestration rates within forest floor and mineral soil, respectively, but there is little evidence of major differences in the combined forest floor and mineral soil after three decades.  相似文献

Different food production methods may result in differences in the content of secondary metabolites such as polyphenolic compounds. The present study compared conventionally (CPD) and organically produced (OPD) diets in a human crossover intervention study (n = 16) with respect to the intake and excretion of five selected flavonoids and effect on markers of oxidative defense. The urinary excretion of quercetin and kaempferol was higher after 22 days of intake of the OPD when compared to the CPD (P < 0.05). The excretions of flavonoids in urine as a percentage of intake (0.6-4%) were similar after both interventions. Most markers of antioxidative defense did not differ between the diets, but intake of OPD resulted in an increased protein oxidation and a decreased total plasma antioxidant capacity compared to baseline (P < 0.05). Some varietal difference was seen in the study, and because selection of more resistant varieties is of central importance to organic farming, it cannot be excluded that the observed effects originate from these differences. The food production method affected the content of the major flavonoid, quercetin, in foods and also affected urinary flavonoids and markers of oxidation in humans.  相似文献

Input-output fluxes of nitrogen (N) and other ecosystem data from 64 European forest ecosystem studies have been compiled in a database (ECOFEE). Sites with high N deposition (up to 64 kg N ha–1yr–1) were characterized by high input of ammonia/ammonium. The deposition of oxidized N was usually only 10 to 15 kg N ha–1yr–1 Of all the sites included, 60 % leached more than 5 kg N ha–1yr–1. Elevated nitrate leaching appeared at inputs above 10 kg N ha–1yr–1. At several sites with inputs of 15–25 kg N ha–1yr–1 nitrate leaching approached the N input, whereas ammonium dominated sites with high input still retained c. 50 % of the input.  相似文献

During routine analysis of commercial fish on halogenated pollutants, an unknown tribromo component (TriBHD) was initially detected as an abundant peak in sample extracts from the Mediterranean Sea. The molecular formula was established to be C16H19Br3O by gas chromatography with electron ionization high-resolution mass spectrometry (GC/EI-HRMS). GC/EI-MS data were virtually identical with a polybrominated hexahydroxanthene derivative (PBHD) previously isolated from an Australian sponge species known to occur in the Mediterranean Sea as well. A tetrabromo isomer (TetraBHD) was also found in the fish samples. The concentrations of TriBHD and other halogenated compounds in commercial fish (sea bass, gilt head bream, anchovy, sardine, and salmon) were estimated with GC/electron capture detection (ECD). Using the ECD response of trans-nonachlor, the concentration of TriBHD reached up to 90 ng/g lipid weight and accounted for up to >90% of the concentration of p,p'-DDE, which was the most abundant peak in the most samples investigated. On the basis of the GC/ECD response, TetraBHD amounted for approximately 1/7 of TriBHD in all fish samples investigated. The sample with the highest content was a green-lipped mussel from New Zealand (236 ng/g lipid weight). The halogenated natural products TBA, Q1, and MHC-1 were also present in most of the samples. We assume that the bulk of the residues in fish from aquaculture may originate from algae and sponges living in proximity of the fish farms. Detection of TriBHD and TetraBHD in blubber of a monk seal (Monachus monachus) suggests that both HNPs may reach the top predators of food webs and thus also humans.  相似文献
