Studies on laboratory animals have shown that viruses vary as to whether or not they are transmissible by the gametes or are capable of passing through the zona pellucida and infecting the embryo.
Methods of studying early embryos for the presence of infectious agents include electron microscopy, immunocytochemistry and cell cultivation.
Determination that early bovine embryos do not become infected by certain agents might allow for easing of restrictions in the current import and export regulations for cattle embryos.
Embryo transfer could be used as a means of controlling or eliminating disease in a herd or flock if the causal agent does not infect the early embryo via the gametes or by penetrating the zona pellucida.

Granular vulvovaginitis (GVV) was experimentally produced in female kids by topical application of M. agalactiae on the scarified vulvar mucosa. Grossly visible GVV lesions were seen in 25 out of 30 infected kids, yet all were positive for microscopic lesions. Microscope lesions that appeared by 7 days post infection (DPI) were comprised of stromal oedema, lymphocytic infiltration into the lamina propria and perivascular accumulation of a few lymphocytes. The lesions observed between 28 and 49 DPI were comparable to those of spontaneous cases (severe). The changes seen in kids euthanized between 56 to 70 DPI were suggestive of the chronic stage of the disease. M. agalactiae was reisolated from all the infected kids from 7 to 70 days after inoculation. The pathology and pathogenesis of this condition have been described and discussed. 相似文献

There is a lack of laboratory-based embryonic chicken toxicity studies with the ecologically relevant low dose/s of endosulfan that utilizes a more practical approach such as the chorioallontoic membrane (CAM) injection. In this investigation, 2μg AR grade α-endosulfan/egg (40% of LD50 for embryos) was injected through the CAM in 12-day-old chicken embryos and the activities of glucose-6-phosphatase (G6Pase, EC 3.1.3.9), fructose 1,6-diphosphatase (FDPase, EC 3.1.3.11), adenosine triphosphatase (ATPase, EC 3.6.1.3) and succinic dehydrogenase (SDH, EC 1.3.99.1) and DNA and RNA content in liver and brain tissues and acetyl cholinesterase (AChE, EC 3.1.1.7) in the latter were determined at 24, 48, and 72 h post-exposure. The wet weight of the embryos did not differ between groups. Following endosulfan exposure, except increase in the hepatic ATPase activity (P < 0.01), there was a significant decrease in the following parameters: G6Pase activity in both the liver and brain (P < 0.01), SDH activity in the brain (P < 0.01), brain overall DNA and RNA concentration (P < 0.05), brain AChE activity (P < 0.01). Exposure of 18-day-old embryos to 2-μg endosulfan for 24 h caused decrease (P < 0.01) in the lymphocyte count and IgG content. Histopathology of thymus and bursa of Fabricius revealed a reduction in the population of thymic follicles, smaller thymocytes with the clear vacuoles in cytoplasm and fewer bursocytes accompanied by infiltration of erythrocytes in lymphoid follicles of the endosulfan-treated embryos. It was inferred that in ovo injection of 0.041 μg/g egg weight of α-endosulfan suppress gluconeogenesis (main energy source in embryonic life), nerve transmission, and immunity. 相似文献

Hormonal asynchronies during oestrus, related to the presence of suprabasal plasma-progesterone (P4) concentrations and a delayed ovulation, interfere with the fertility of repeat-breeder heifers (RBH). Since tubal dysfunction can occur in connection with hormonal asynchronies and constrained availability of fertile spermatozoa at the time of ovulation, the present study tested the hypothesis that frequent sperm deposition from onset of oestrus to ovulation may improve pregnancy rates in RBH. Five RBH and five virgin heifers (VH; controls) were repeatedly artificially inseminated (AI) at 6 h intervals from onset of oestrus to spontaneous ovulation. Hormone analyses revealed suprabasal P4 concentrations and a delay in the occurrence of the luteinising hormone (LH) surge, but a normal cortisol profile in RBH. Compared with controls, RBH presented longer interval from onset of oestrus to ovulation, and therefore, received more AIs. Pregnancy rates in RBH reached control levels (60%; NS), indicating that the hypothesis might be correct. Pregnancy rates in VH were below the expected range, presumably attributed to a deleterious influence of the frequent handling. The study suggests that pregnancy rates can be improved in RBH by frequent AI in relation to spontaneous ovulation. However, this practice of repeated manipulations, while seeming not to show adverse effects, lacks practicality for routine use. 相似文献

DAS-ELISA proved to be reliable enough to detect a latent infection by Tomato spotted wilt virus (TSWV) in asymptomatic stock plants of chrysanthemum. A high density of Frankliniella occidentalis, the predominant vector, in the presence of latently infected stock plants resulted in a high incidence of disease in the chrysanthemum
production field. The incidence of disease was low when the vector thrips were not abundant in spite of the presence of latently
infected stock plants. These results suggest that an infestation of the vector thrips causes severe secondary spread of TSWV
originating from latently infected stock plants in chrysanthemum production fields.

Variation of Parthenium hysterophorus in response to soil quality: implications for invasiveness 总被引:1,自引:0,他引:1
Summary The performance of Parthenium hysterophorus , native to the tropical Americas and invasive to several countries, was evaluated for response to soil quality. Phenological (six stages) and quantitative growth variables [relative growth rates in height (Rh ) and diameter (Rd )] were measured every fortnight. Based on harvest data, the variables, root:shoot (R:S) ratio, specific leaf area (SLA), relative growth rate in biomass (Rw ), net assimilation rate (NAR) and dry matter allocation to plant components, were determined. High clay content in soils prolonged the rosette stage, enhanced Rh and Rd and hampered root growth, but promoted biomass allocation to shoots. The extreme degree of plant mortality was observed, with only 33.3% individuals surviving to reproduce in the soils with highest clay contents. This appears to be one of the most important findings that certainly has a major bearing on the range of adaptation of the weed. Seed mass declined whereas seed production increased in relatively coarser soils with the exception of nearly pure sand. In soils rich in clay, plants produced a smaller number of larger seeds. These data suggest that variation in fundamental functional traits would enable P. hysterophorus to adjust to a variety of habitat conditions. 相似文献

Laboratory evaluation of the insect growth regulator lufenuron againstHelicoverpa armigera on cotton
An insect growth regulator (IGR), lufenuron (Match 5EC), was tested for its toxicity toHelicoverpa armigera on cotton. Potency of the IGR against the larval stage of the pest was demonstrated with respect to larval instars; the LC90 values of 1st, 2nd, 3rd, 4th and 5th instar larvae were 5.63, 7.89, 8.03, 11.39 and 14.76 mg a.i.l
−1, respectively. However, different larval instars did not differ significantly with respect to LC50 and LC10. IGR-treated larvae had swollen heads and were significantly smaller (1.5–2.3 mm) than the untreated control (2.9 mm). Larval
weight was significantly reduced from 190 mg in the control to 50–70 mg in the lufenuron treatment. IGR treatment in the larval
stage significantly affected both pupal length and pupal weight. Pupal duration of the test insect was significantly extended
by IGR treatment. Pupal deformities, including an inability to shed the last larval skin and formation of larval-pupal intermediates,
occurred following treatment. A significant reduction in adult emergence was recorded. In addition, abnormalities in the form
of development of cavities in the forewings of adult were evident. A significant decline in fecundity was noted in the studies.

Aciculosporium take (Ascomycota; Clavicipitaceae) is a causal agent of witches' broom of
bamboo plants. The symptoms of this disease are believed to be induced by plant
hormones, particularly auxins. Indole-3-acetic acid (IAA) was identified in
cultures of this fungus in an l-tryptophan-supplemented liquid medium.
IAA production was confirmed on 30 isolates of A. take from various
hosts and locations at levels up to 1 mg/l. The biosynthetic pathway of
IAA in A. take culture was examined by analyzing intermediate products
and by feeding experiments. The results showed that the indole-3-pyruvic acid
pathway (l-tryptophan → indole-3-pyruvic acid → indole
acetaldehyde → IAA) was the dominant pathway in A. take.

Natural occurrence of a geminivirus causing severe leaf curl disease on sunn hemp (Crotalaria juncea) was recorded in India. The association of a geminivirus with the disease was demonstrated by whitefly transmission tests and polymerase chain reaction (PCR) amplification of DNA fragments of expected sizes with three pairs of degenerate geminivirus primers. The PCR-amplified viral DNA fragments were further characterized by Southern hybridization with a geminivirus probe consisting of the cloned coat protein (CP) gene of Indian tomato leaf curl virus (ITLCV). Restriction fragment length polymorphism analysis of a PCR-amplified CP fragment revealed that the geminivirus from sunn hemp was different than ITLCV. 相似文献
