A phenological study was conducted to determine the impact of harvest maturity on the immune-modulating properties of Echinacea purpurea. The aerial parts of this plant were collected during seven stages of development and were assayed for a common botanical marker for this species, cichoric acid. Plants of selected development stages were also assayed for total polysaccharides and compared for their immune-modulating effects on the THP-1 monocyte/macrophage cell line by means of a gene expression study. Although the concentration of cichoric acid did not change significantly during the course of the study, stage 1 (advanced vegetative) had the highest concentration of total polysaccharides and exhibited the most potent induction activity on immune-modulating cytokines such as interferon-gamma and tumor necrosis factor-alpha. These findings suggest that the use of gene expression may be an effective tool not only to standardize botanical extracts but also to optimize harvest time.  相似文献

In this work, a single flow injection multicommuted system using solid-surface fluorescence spectroscopy has been explored for the determination of diphenylamine in apples and pears. The native fluorescence signal of diphenylamine retained on the solid support (C18 silica gel) was used for its determination (lambda(exc)/lambda(em) 291/372 nm). The sample treatment consists of a liquid-liquid extraction with acetonitrile followed by a cleanup step using primary-secondary amines. The use of a continuous flow assembly implementing multicommutation, based on a set of three-way solenoid valves controlled by appropriate software, provides the automatic control of sample, carrier, and eluting solution with remarkable advantages in relation to conventional flow injection approaches. Using an optimized sampling time, the proposed method was linear in the range 0.25-5 mg kg(-1) with a detection limit of 0.06 mg kg(-1) and RSD (percent) values better than 3%. The proposed method was successfully applied to the analysis of diphenylamine in different apple and pear samples fortified at different concentrations, and recoveries between 78 and 104% were found. The results obtained illustrate the usefulness of the proposed method for the screening and evaluation of postharvest treatment of crops possibly containing diphenylamine.  相似文献

A field experiment was carried out to evaluate the effectiveness of mycorrhizal inoculation with three arbuscular mycorrhizal (AM) fungi (Glomus intraradices Schenck & Smith, Glomus deserticola (Trappe, Bloss. & Menge), and Glomus mosseae (Nicol & Gerd.) Gerd. & Trappe) and the addition of composted sewage sludge (SS) with respect to the establishment of Retama sphaerocarpa L. seedlings, in a semiarid Mediterranean area. Associated changes in soil chemical (nutrient content and labile carbon fractions), biochemical (enzyme activities), and physical (aggregate stability) parameters were observed. Six months after planting, both the addition of composted SS and the mycorrhizal‐inoculation treatments had increased total N content, available‐P content, and aggregate stability of the soil. Values of water‐soluble C and water‐soluble carbohydrates were increased only in the mycorrhizal‐inoculation treatments. Rhizosphere soil from the mycorrhizal‐inoculation treatments had significantly higher enzyme activities (dehydrogenase, protease‐BAA, acid phosphatase, and β‐glucosidase) than the control soil. In the short‐term, mycorrhizal inoculation with AM fungi was the most effective treatment for enhancement of shoot biomass, particularly with G. mosseae (about 146% higher with respect to control plants). The addition of the composted SS alone was sufficient to restore soil structural stability but was not effective with respect to improving the performance of R. sphaerocarpa plants.  相似文献

The umbu tree (Spondias tuberosa) is a xerophytic woody species endemic to the northeast of Brazil. The present work reports a three-year in situ evaluation of major yield components in a random set of trees in Petrolina city, Pernambuco State, Brazil and the application of repeatability coefficient in the context of genetic resources utilization. Five traits were assessed: total fruit yield per plant (FY); total number of fruits per plant (NF); average fruit weight (FW); average pulp weight (PW) and average fruit diameter (FD). The values observed for FY ranged from 4.2–184 kg with mean of 61.5 kg. The values for NF ranged from 257 to 12,981 fruits/tree with mean 3,993 fruits. For FW, the range was from 10.82–23.36 g with mean of 16.03 g. The values for PW ranged from 7 to 17 g with mean of 11.2 g, while FD values varied from 2.5 to 3.5 cm with mean of 3.0 cm. Six methods were employed to estimate repeatability coefficient (r) for all parameters. The r ranged from 0.68–0.76 for NF; 0.87–0.89 for FW; 0.65–0.75 for FY; 0.64–0.78 for PW and 0.70–0.84 for FD. Narrower ranges across methods were observed to r-values greater than 0.80, as observed for FW and FD. Three to four years of measurements will be necessary for FY, NF, and PW and one year for FW to obtain a precision of 90% to provide a reliable identification of individuals for in situ or ex situ conservation.  相似文献

Coupled nitrification-denitrification and potential denitrification were measured as 15N2O and 15N2 evolution rates in ammonium sulphate-treated rice soils with or without Terrazole [5-ethoxy-3 (trichloromethyl) 1,2,3 Thiadizole] under laboratory and field conditions. The greatest coupled nitrification-denitrification activity was found after drying and rewetting the soil, with maximum values of 322 ng N cm–2 h–1 in the laboratory and 90.8 ng N cm–2 h–1 in the field. These 15N2O + 15N2 evolution rates were about 10 times lower than potential denitrification in these soils. These results and the observed decrease in 15N2O + 15N2 evolution rate in soils treated with Terrazole (60% under laboratory conditions and 52% under field conditions) indicate that denitrification was limited by coupled nitrification-denitrification activity. Oxygen and previous addition of ammonium sulphate appear to control the rate of 15N2O + 15N2 evolution in ammonium sulphate-fertilised soils.  相似文献

To verify the presence of glycolaldehyde in wine resulting from reduction of glyoxal after malolactic fermentation, sterilized solutions of synthetic cultures were inoculated with a lactic bacterium of the type Oenococcus oeni. Fermentation was also carried out on solutions with glyoxal added. The resulting glycolaldehyde concentrations turned out to be associated with the amounts of glyoxal present, and glyoxal was seen to decrease as glycolaldehyde increased. In addition, it was observed that glyoxal principally forms from breakdown of sugars and that reduction to glycolaldehyde is mainly promoted by bacterial activity. Finally, the ability of glycolaldehyde to induce browning of (+)-catechin in a model wine system was verified and turned out to be about 10 times higher than that of ascorbic acid.  相似文献

Coevolution of mammals and their gut microbiota has profoundly affected their radiation into myriad habitats. We used shotgun sequencing of microbial community DNA and targeted sequencing of bacterial 16S ribosomal RNA genes to gain an understanding of how microbial communities adapt to extremes of diet. We sampled fecal DNA from 33 mammalian species and 18 humans who kept detailed diet records, and we found that the adaptation of the microbiota to diet is similar across different mammalian lineages. Functional repertoires of microbiome genes, such as those encoding carbohydrate-active enzymes and proteases, can be predicted from bacterial species assemblages. These results illustrate the value of characterizing vertebrate gut microbiomes to understand host evolutionary histories at a supraorganismal level.  相似文献

The isolation of broadly neutralizing antibodies against influenza A viruses has been a long-sought goal for therapeutic approaches and vaccine design. Using a single-cell culture method for screening large numbers of human plasma cells, we isolated a neutralizing monoclonal antibody that recognized the hemagglutinin (HA) glycoprotein of all 16 subtypes and neutralized both group 1 and group 2 influenza A viruses. Passive transfer of this antibody conferred protection to mice and ferrets. Complexes with HAs from the group 1 H1 and the group 2 H3 subtypes analyzed by x-ray crystallography showed that the antibody bound to a conserved epitope in the F subdomain. This antibody may be used for passive protection and to inform vaccine design because of its broad specificity and neutralization potency.  相似文献
