Summary Cucumber mosaic virus (CMV) infections rank among the most devastating diseases in the commercial culture of tomato (Lycopersicon esculentum Mill.), for which suitable sources of natural resistance are not available. The concept of pathogen-derived resistance, however, offers an alternate approach to combat plant viral diseases by transformation of crops with nucleotide sequences derived from the viral genome. This report demonstrates the successful application of such a pathogen-derived resistance gene comprising the CMV coat protein (CP) gene, to generate protection to CMV infections in cultivated tomato. Transformation of an inbred tomato line with the CMV CP gene isolated from a subgroup I strain, engendered high levels of protection to various CMV strains, including a virulent strain causing lethal necrosis and a typical subgroup II strain. Moreover, when challenged by natural infection through aphid vectors in open field, levels of protection were largely maintained in hemizygous hybrids. In all, these results demonstrate that synthetic resistance genes based on the CMV CP gene make excellent sources of broad spectrum resistance to CMV infections for introgression into tomato breeding programs.  相似文献

Spread of plant disease in production chains of planting material is a process of great economic importance, but has received little attention from plant disease epidemiologists. Disease control in production chains is therefore often based on rules of thumb and expert judgement by regulatory bodies, rather than on an explicit analysis and evaluation of the epidemiological and economic consequences of alternative strategies. This paper puts forward the idea that individual-based models may be used as a framework to simulate the spread of disease-causing organisms in plant production chains. The “individuals” in this context are the trading units (e.g., batches, lots) of a production chain. The quarantine disease “potato brown rot”, caused by the bacterium Ralstonia solanacearum, is used as an illustrative example. The model simulates the spread of potato brown rot over all potato growing farms and fields in the Netherlands over a chosen time frame. It addresses the relevant infection pathways for this disease in potato production and is spatially explicit.

Model outputs of simulations based on the control strategy as applied in the Netherlands until 2004 are presented. The effects of minor adjustments to this strategy are investigated. The simulations show an irregular pattern of brown rot dynamics in the potato production chain, as is observed in practice. Simulations quantify the relative importance of different infection pathways, and elucidate the effect of testing frequency on these pathways and on the over-all brown rot incidence. The study shows that individual-based modelling (IBM) provides a powerful platform for modelling the epidemiology and impact of diseases in plant production chains. IBM can be effectively used for the analysis, evaluation and design of cost-effective disease management policies.  相似文献

In human beings, rFVIIa has been used successfully to promote hemostasis in patients with various bleeding disorders. Patients with hemophilia, thrombocytopenia, and thrombocytopathy have been effectively and safely treated as well as patients with severe trauma or surgery resulting in profuse bleeding and patients with impaired liver function. Given these successes, the possibility of using rFVIIa in veterinary medicine is appealing. The clinical indications and uses in veterinary medicine would be similar, but there are currently two major obstacles: immunogenicity and cost. If these problems can be addressed, rFVIIa may become a valuable and indispensable therapy, especially in the emergency and critical care settings. The use of rFVIIa could allow for successful treatment of otherwise intractable bleeding as well as decrease the number of blood transfusions needed.  相似文献

The transient elevated plasma growth hormone (GH) levels that occur at a young age in giant breed dogs may have consequences in adult life. The aim of this study was to investigate whether excess juvenile GH has consequences for cardiac function and morphology. To simulate the naturally occurring juvenile hypersomatotropism in giant breed dogs, elevated plasma GH and insulin-like growth factor-I (IGF-I) concentrations were induced in six miniature poodles (GH dogs) by daily administration of supraphysiological doses of GH starting at 12 weeks of age. Eight miniature poodles of the same age that received vehicle only served as controls. Cardiac anatomy and function were evaluated by echocardiography. After euthanasia at 21 weeks of age, the hearts were examined for weight, myocyte dimensions and collagen fraction. The hearts of the GH dogs had larger atria (+22%), a thicker left ventricular wall (+21%), greater weight (+84%), and their cardiomyocytes were 15% longer, 25% thicker, and 92% greater in volume than those of control dogs. The mean collagen fraction was also higher in the GH dogs (5.6%) than in the controls (3.1%). In conclusion, excess GH in juvenile miniature poodles resulted in myocardial hypertrophy and increased collagen content. These findings are consistent with observations in acromegalic human patients and in rats treated with GH.  相似文献

Colibacillosis results from infection with avian pathogenic Escherichia coli bacteria. Healthy broilers are resistant to inhaled E. coli, but previous infection with vaccine or virulent strains of Infectious Bronchitis Virus (IBV) predisposes birds for severe colibacillosis. The aim of this study was to investigate how IBV affects the course of events upon infection with E. coli. Broilers were inoculated with IBV H120 vaccine virus or virulent M41 and challenged 5 days later with E. coli 506. A PBS and E. coli group without previous virus inoculation were included. Sections of trachea, lung and airsacs were stained for CD4, CD8, gammadelta-TCR, alphabeta1-TCR, and for macrophages (KUL-01) and both pathogens. Changes in the mucociliary barrier of trachea, lung and airsacs did not predispose for bacterial superinfection. The disease in the lungs of the E. coli group and both IBV/E. coli groups was similar. Lesions in the airsacs were more pronounced and of longer duration in the IBV/E. coli groups. The immunocytological changes differed substantially between the E. coli group and both IBV/E. coli groups. In trachea, lungs and airsacs the CD4+ and CD8+ populations were significantly larger than in the E. coli and PBS groups. In the lungs and the airsacs the macrophages were more numerous in the IBV/E. coli and the E. coli groups than in the PBS group. The presence of high numbers of T cells and macrophages in IBV infected birds most likely induced an altered immune response, which is responsible for the enhanced clinical signs of colibacillosis.  相似文献

We view agrobiodiversity as a social-ecological phenomenon and, therefore, an example of nature–gender relations within agrarian change, including social, economic, political and technical changes in agriculture and rural areas. As a result of the industrialization of agriculture, nature–gender relations in the field of agrobiodiversity have become characterized by separation processes such as conservation versus use or subsistence versus commodity production. We argue that the sustainable development paradigm, as currently implemented in European Common Agricultural Policy through the concept of multifunctionality, does not necessarily overcome separation tendencies and lead towards integration, despite its claim to bring together different ecological, economic and social needs. In our paper we critically reflect this observation and develop a theory-based analytical framework at the interface of nature and gender relations. For analytical purposes we distinguish between three different agrarian structures (pre-industrialized, industrialized and multifunctional) and focus on the development of two separation tendencies within them and their effects on agrobiodiversity. Concerning nature, we discuss the effects of separating agrobiodiversity conservation and use. With regard to gender, we discuss the separation of subsistence and commodity production. Against this background, we claim for new rural economic rationalities characterized by processes whose qualitative, material and value dimensions maintain agrobiodiversity.  相似文献

Every application of antibacterial drugs in veterinary medicine may encourage selection for resistant bacteria. In Germany no valid data are available which would be suitable for a species specific estimation of drug consumption especially regarding food producing animals. Therefore, a representative monitoring of consumption of antibacterial drugs in food producing animals should be implemented. As a first step, a feasibility project was conducted to identify the technical preconditions and develop a concept for a regular monitoring system within Germany as a country with a non-central federal state system. The data were collected via the forms obligatory by German law concerning the treatment of animals and the delivery of animal drugs to the animal owners by the veterinarian. 24 veterinary practices and 65 farmers were visited, and all applications of antibiotics to farm animals during the course of one year (September 1, 2006 to August 31, 2007) were entered into a central database. A total of 95,584 records were collected and analysed statistically. Consumption of antibiotics was calculated in kg, but also the number of applications was analysed. The consumption of tetracyclines in kg reached 54.3% of all antimicrobial substances applied to pigs, but only 25.7% of all doses applied to pigs were tetracyclines. For the farms' data, the number of daily doses per animal year (DD(ay)) was estimated based on the number of daily doses recorded and on the number of animals kept in the farm. Correct and detailed data regarding the structures of the farms as well as of veterinary practices are necessary to estimate the consumption of antibiotics reliably. The proposed system is able to function as a monitoring system for antibiotic use in Germany, when the monitoring data are linked to the agricultural data (farm sizes) accounting for differences between German regional agricultural and animal husbandry structures. Furthermore, the results of the antibiotic use analyses may serve as basis to assess the results of the sales data of the pharmaceutical industry. Results are comparable to the outcome of respective systems in other European countries, e.g. the Netherlands and Denmark, and therefore it will contribute to a better understanding and development of strategies for the control of antimicrobial resistances on the European level.  相似文献

Septoria tritici blotch caused by the fungus Zymoseptoria tritici (formerly Mycosphaerella graminicola) is one of the most yield-reducing diseases worldwide. Effective disease management involves the use of resistant cultivars and application of fungicides. In this study, the population structure and genetic diversity of 183 Z. tritici isolates from Denmark, Sweden, Finland and the Baltic countries were analysed by molecular markers. In population structure analysis, isolates from Denmark and Sweden were grouped together, whereas isolates from the Baltics and Finland were grouped together. Analysis of genetic diversity and ?-values confirmed the division of Nordic and Baltic regions. Danish isolates sampled from different regions and different varieties were not genetically different. However, significant genetic differences were detected between isolates sampled from different years in Denmark and for isolates sampled from specific cultivars in different years. Additionally, the frequency of several known point mutations in the gene cyp51, conferring decreased sensitivity to DMI fungicides, was investigated. Several of the examined mutations were detected at a lower frequency in Baltic isolates compared to Danish and Swedish isolates. Analysis of the Danish population revealed a significant increase in specific mutations over the years. Lastly, some mutations were significantly more frequent in isolates derived from certain varieties. By using different resistance sources in breeding programmes and application of a wide range of fungicides, a sustainable and efficient disease management can be obtained.  相似文献
