Summary Somaclonal variation is a tool that can be used by plant breeders. The review examines where this tool can be applied most effectively and the factors that limit or improve its chances of success. The main factors that influence the variation generated from tissue culture are (1) the degree of departure from organised growth, (2) the genotype, (3) growth regulators and (4) tissue source. Despite an increasing understanding of how these factors work it is still not possible to predict the outcome of a somaclonal breeding programme. New varieties have been produced by somaclonal variation, but in a large number of cases improved variants have not been selected because (1) the variation was all negative, (2) positive changes were also altered in negative ways, (3) the changes were not novel, or (4) the changes were not stable after selfing or crossing. Somaclonal variation is cheaper than other methods of genetic manipulation. At the present time, it is also more universally applicable and does not require containment procedures. It has been most successful in crops with limited genetic systems and/or narrow genetic bases, where it can provide a rapid source of variability for crop improvement. 相似文献

Fusarium culmorum is a phytopathogenic, toxigenic fungus causing seedling diseases, foot rot and head blight of cereals. For estimating competition effects in mixtures of two single-spore isolates, two winter rye single crosses were tested with either four isolates individually or four 1 : 1 mixtures of the same isolates in six field environments. Two isolates (FC46, FC64) were highly aggressive deoxynivalenol (DON) and 3-acetyl DON-producers, the other two (FC30, FC71) were medium aggressive nivalenol-producers. Rye heads were inoculated during flowering with conidia of pairs of isolates expressing similar (FC46 + FC64, FC30 + FC71) or contrary (FC46 + FC71, FC30 + FC64) levels of aggressiveness and similar or different concentrations and chemotypes of mycotoxins, respectively. Head blight rating and yield components relative to the non-inoculated plots were recorded as aggressiveness traits. Additionally, mycotoxin concentrations were measured in the rye grain. Random pathogen samples were re-isolated from heads at the onset of symptom development and analysed by molecular markers (RAPD–PCR) in one environment. Aggressiveness of the isolate mixtures was significantly lower than that of the isolates applied individually on both rye genotypes. Similarly, mycotoxin concentrations were significantly lower in the mixtures in seven out of eleven comparisons. Among the re-isolates, the component genotypes of a mixture significantly deviated from the inoculated 1 : 1 ratio when a particular isolate (FC46) was present in the mixture. This isolate displayed a superior competitive ability irrespective of the aggressiveness or mycotoxin profile of the mixing partner illustrating that pathogenic fitness is caused by additional factors that have not, as yet, been identified. 相似文献

Regions within the mitochondrial gene encoding for the nicotinamide dehydrogenase subunit 4 (nad4) were characterized to evaluate the extent of genetic variation within and among Xiphinema diversicaudatum and Xiphinema simile populations. Four different sequence variants of nad4 were determined among eight populations of X. diversicaudatum and three variants among three populations of X. simile. Nucleotide variation was detected in 28 of 411 bp (2.43 to 4.87 %) in X. diversicaudatum and in three of 395 bp (0.25 to 0.76 %) in X. simile. The in-vivo toxicity data of 33 O,O-diethyl O-(1-alkyl-3-R″-1,2,4-triazol-5-yl) phosphorothioates, reported in Part I of this series, were subjected to multiregression analyses for the following organisms: Leptinotarsa decemlineata (larvae), Macrosiphum euphorbiae (aphids), Culex pipiens (adults), Spodoptera littoralis (larvae) and the rat. The activities were found to depend mainly on the steric parameters of R″, these being best expressed by the values L, B1 and B4 of the computer program STERIMOL. In agricultural pathosystems resistant cultivars are typically only temporarily effective, as widespread growth of said cultivars drives selection for pathogen genotypes capable of infecting them. A gene-for-gene interaction between Z. tritici and wheat has been demonstrated for one cultivar; however results of studies into the relevance of these interactions in the field remain inconsistent. Because genetic drift does not appear to occur between Z. tritici populations that are not widely geographically separated, according to neutral genetic theory if adaptation to different host cultivars is occurring, reduced genetic variation, and some differentiation between populations sourced from different cultivars should be observed. Selectively neutral microsatellite markers were used to genotype 260 isolates of Z. tritici taken from two naturally infected randomized block trials of four different cultivars, representing a spectrum of resistance to Z. tritici from susceptible to resistant. By calculating genetic parameters such as overall heterozygosity and F ST from this genotypic data, the presented study aimed to determine if genetic drift or host selection is impacting on the genetic structure of the Irish Z. tritici population. Results indicated that diversity was distributed almost entirely within, rather than among populations, with little or no differentiation, and almost no clone isolates were present in the dataset. However this result was not reflected in the accessory chromosomes, where evidence of minor but significant genetic structure was found. This lack of structure in the core chromosomes and weak structure in the accessory chromosomes confirms that forces of genetic drift and selection are minor compared to sexual reproduction, in concurrence with multiple previous studies on other populations worldwide. 相似文献

The sorption of pesticides to soil particles has implications for their distribution and fate in the soil environment. A batch equilibrium technique was used to investigate sorption of the fungicide AEC623892 to intact and hydrogen-peroxide-treated whole soils and several particle-size fractions isolated from them. Sorption of AEC623892 to the soil as a whole was low. K(f oc) values measured in the whole soils were 169.2 and 41.9 ml g(-1) for Soil A and Soil B respectively. The highest values of K(f oc) were measured in soil particle-size fractions <53 microm (266.5 ml g(-1) in the 2-20 microm fraction of Soil A; 471.9 ml g(-1) in the 20-53 microm fraction of Soil B). Sorption was most irreversible in the 2-20 microm fractions. Overall, treatment of soil particle-size fractions with hydrogen peroxide resulted in lower values of K(f oc) (112.3 ml g(-1) in Soil A whole soil and 30.9 ml g(-1) in Soil B whole soil). In both soils, the maximum sorption among hydrogen-peroxide-treated samples was observed in the <2 microm fraction (166.6 and 311.0 ml g(-1) for Soil A and Soil B, respectively). Investigation of the mineralogical composition of the soils suggested that the clay mineralogy (dominated by kaolinite and illite) is less likely to account for the differences in sorption observed than differences in the characteristics of the soil organic matter. Thermal analysis of the different soil fractions indicated that hydrogen peroxide treatment preferentially removed aliphatic fractions of organic matter, but had less effect on lignin-like, aromatic fractions. 相似文献

The simultaneous HPLC determination of clofentezine, fenoxycarb and hexythiazox in apples, pears and their pulps is described. The method is based on a clean-up procedure carried out on a Seppak C18 cartridge followed by HPLC analysis on an RP-18 column using acetonitrile + water as mobile phase. Average recoveries were 85.0% for clofentezine, 84.8% for fenoxycarb and 75.6% for hexythiazox on apple matrix; recoveries from pear matrix were in the same range. UV detection limits at 240 nm were 0.07 mg kg?1 for clofentezine, 0.064 mg kg?1 for fenoxycarb and 0.08 mg kg?1 for hexythiazox. 相似文献
