Rectal swab samples were collected from 60 red squirrels (Sciurus vulgaris) from July 2006 to April 2007 in Southern Italy. Samples were tested for Campylobacter jejuni and C. coli by culture methods and suspected colonies were then confirmed by polymerase chain reaction. C. jejuni was detected in 5/60 (8.3%) samples examined but infection status was not related to age or sex and C. coli was not isolated. This is believed to be the first report of C. jejuni infection in the red squirrel.  相似文献

We report the accurate determination of the allelic configurations of a total of eight new citrus tetraploid hybrids by means of SSR analysis, coupled with capillary electrophoresis, and PCR based dosage effects. Tetraploid hybrids were spontaneously obtained from different interploid crosses (2x × 4x) between diploid ‘Femminello’ lemon and the allotetraploid somatic hybrid (2n = 4x = 36) ‘Key’ lime + ‘Valencia’ orange, and between diploid ‘Wilking’ and ‘Fortune’ mandarins and an autotetraploid ‘Dancy’ mandarin (2n = 4x = 36). To understand the opportunity to employ them in further backcross programs, the cytological mechanisms underlying their ploidy level were unambiguously determined using six SSR primers. PCR conditions were optimized and skewness in template/product ratios were verified. Tetraploid allelic configurations were determined from PCR based dosage effects using electropherogram peak heights to estimate the copy number per allele. In all the tetraploid hybrids we found out that diploginy (2n eggs) has occurred, contributing the extra haploid genome in the tetraploids. According to the marker genotypes, it was further inferred that the 2n eggs in ‘Femminello’ lemon resulted from first division restitution (FDR), while in ‘Wilking’ and ‘Fortune’ mandarins 2n eggs occurred in second division restitution (SDR). These new genotypes, with their improved genetic female background, can be therefore considered very valuable in our citrus genetic improvement program as pollen donors in backcrosses suitable to eliminate negative traits.  相似文献

Stylosanthes capitata and S. guianensis are important forage legumes for tropical areas. The only available estimates of S. capitata and S. guianensis outcrossing rates were based on morphological markers, and the genus is considered as being mainly self-pollinated. Here we describe an estimation of the outcrossing rate in S. capitata and S. guianensis using microsatellite markers. The outcrossing rates were estimated in S. capitata and S. guianensis open-pollinated populations of 20 progenies consisting of ten individuals each. The multi locus outcrossing rate for S. capitata was estimated using 10 polymorphic loci, whereas five microsatellites were used for S. guianensis. The multi locus outcrossing rates for S. capitata and S. guianensis were 31 and 26%, respectively, suggesting a mixed mating system with predominance of autogamy. Comparison of single locus and multi locus estimates of outcrossing rates indicated that little inbreeding other than selfing occurred. The estimated Wright’s fixation index of the parental generation was lower than expected based on the multi locus outcrossing rate, possibly resulting from the use of some heterozygous breeding genotypes for the study. The data on the outcrossing rate described here are potentially useful for breeding programs and for maintenance of germplasm collections of these Stylosanthes species.  相似文献

This study was carried out in order to assess the influence of biochar applications on the estimation of colorimetric‐based enzymatic assays and to verify the effectiveness of the most common methods. Since most methods used to determine enzymatic activities in the soil are based on colorimetry, biochar may absorb substrates and/or coloured products thereby distorting the analytical result. Biochar was added to two soils, with different textures and cation exchangeable capacities, at a rate of 2% (w/w), and seven enzyme activities were determined following standard methods. The biochar amendment lowered the spectrophotometer reading of the activity of FDAase and dehydrogenase in the sandy soil. In the three enzymatic activities based on p‐nitrophenol production (β‐glucosidase, phosphatase and arylsulphatase), the addition of biochar did not change the enzyme assays. The biochar led to an overestimation in terms of the protease and urease activities in the sandy soil. In the clay loamy soil, biochar did not change the response of any of the enzyme activities tested. A biochar dose of up to 2% only guarantees the effectiveness of the most common spectrophotometric methods for not excessively sandy soils.  相似文献

Purpose

Biochar has unique properties which make it a powerful tool to increase soil fertility and to contribute to the decrease of the amount of atmospheric carbon dioxide through the mechanisms of C sequestration in soils. Chemical and physical biochar characteristics depend upon the technique used for its production and the biomass nature. For this reason, biochar characterization is very important in order to address its use either for agricultural or environmental purposes.

Materials and methods

Three different biochars obtained from an industrial gasification process were selected in order to establish their chemical and physical peculiarities for a possible use in agronomical practices. They were obtained by charring residues from the wine-making industry (marc) and from poplar and conifer forests. Routine analyses such as pH measurements, elemental composition, and ash and metal contents were performed together with the evaluation of the cross-polarization magic angle spinning (CPMAS) 13C NMR spectra of all the biochar samples. Finally, relaxometry properties of water-saturated biochars were retrieved in order to obtain information on pore size distribution.

Results and discussion

All the biochars revealed basic pH values due to their large content of alkaline metals. The quality of CPMAS 13C NMR spectra, which showed the typical signal pattern for charred systems, was not affected by the presence of paramagnetic centers. Although paramagnetism was negligible for the acquisition of solid state spectra, it was effective in some of the relaxometry experiments. For this reason, no useful information could be retrieved about water dynamics in marc char. Conversely, both relaxograms and nuclear magnetic resonance dispersion profiles of poplar and conifer chars indicated that poplar char is richer in small-sized pores, while larger pores appear to be characteristic for the conifer char.

Conclusions

This study showed the potential of relaxometry in revealing chemical?Cphysical information on industrially produced biochar. This knowledge is of paramount importance to properly direct biochar agronomical uses.  相似文献

Summary Physiological components of kernel development — LAG period, effective filling period duration (EFPD) and grain filling rate (GFR) — ear moisture release (U), ear size (row number and kernels per row), days from emergence to silking and number of leaves, were examinated on 45 F1 hybrids (10×10 diallel cross) in order to study their genetic relationships with yield. Combining ability analysis revealed that all trait variability derived mainly from g.c.a. effects. LAG period and EFPD were the traits most affected by genotype-environment interaction.Covariation analysis (path method) based on mean phenotypic values and on g.c.a. effects yielded similar information. It is shown that GFR and EFPD are both related to plant yield, but GFR made the most important contribution. On the contrary, a significant relationship between yield and LAG was not detected. Ear size components were also positively related to yield and had negative effects on GFR. These results indicate that, for our material, the dry matter accumulation rate is the main limiting factor of yield.Considering s.c.a. effects, kernel number per row made the most important contribution.  相似文献

Two vinification methods involving different degrees of antioxidant protection of Falanghina must during prefermentative steps, and referred as HAMP (high antioxidant must protection) and LAMP (low antioxidant must protection), were compared in terms of fermentation performances of four different yeast strains, composition of the volatile fraction of wines at the end of alcoholic fermentation, and shelf life of wines during storage. The use of HAMP technology resulted in wines with lower volatile acidity and higher concentrations of medium-chain fatty acid ethyl esters, acetates, and volatile fatty acids. For two of the four strains a lower concentration of isoamyl alcohol was also observed. HAMP wines also revealed increased shelf life because of the higher concentration of odor active esters at the end of storage and better preservation of varietal aromas.  相似文献

In this paper, we estimate the distribution of population by exposure to multiple airborne pollutants, taking into account the spatio-temporal variability of daily air quality and the high-resolution spatial spread of human population around Europe. In particular, we consider monitoring network data for five pollutants, namely carbon monoxide, nitrogen dioxide, ozone, coarse and fine particulate matters. The spatial information contained in the large dataset of daily continental air quality is exploited using a multivariate spatio-temporal model capable to cover cross correlation among pollutants, covariates, and missing data as well as spatial and temporal variability and correlation. At the same time, the model is simple enough to be feasible for the large dataset of daily continental air quality over three years. Maximum likelihood estimation is performed using the EM algorithm, and kriging-like spatial estimates are used to compute high-resolution exposure distribution. Moreover, a novel semi-parametric bootstrap technique is used to assess the exposure distribution uncertainty. In this way, we compare the daily population exposure of 33 European countries and three important metropolitan areas in years 2009–2011 using a single flexible model. This article reviews three general themes pertaining to the transformation of Eastern European agriculture from a command system to a market oriented system. The first theme deals with the diverse character of Eastern European agriculture. In a context in which the agricultures of this region are often considered homogenous, acknowledgement of the varied dimension of this sector is a key element in both analytical and political terms. The second theme pertains to the market. The historical and theoretical dispute over the role of the market has not only been central in the socio-economic reorganization of Eastern European countries and agricultures but is also a common element shared with the West. The third and final theme refers to the changing social stratification of rural regions. The emergence of new and powerful social groups and the demise of others represent fundamental aspects in the understanding of possible trajectories for development. The article concludes with a review of the contributions contained in this special issue of Agriculture and Human Values.Alessandro Bonanno is Associate Professor of Rural Sociology at the University of Missouri-Columbia.
