A wild non-progenitor species from wheat (Triticum aestivum L.) tertiary gene pool, Aegilops peregrina (Hack.) Maire & Weiller accession pau3519 (UUSS), was used for introgression of leaf rust (caused by Puccinia triticina) and stripe rust (caused by Puccinia striiformis f. sp. tritici) resistance in bread wheat. The accession was crossed and backcrossed with hexaploid wheat line Chinese Spring PhI to develop two homozygous BC2F6 wheat-Ae. peregrina introgression lines (ILs), viz., IL pau16058 and IL pau16061, through induced homoeologous recombination. Homozygous lines were screened against six Puccinia triticina and two Puccinia striiformis f. sp. tritici pathotypes at the seedling stage and a mixture of prevalent pathotypes of both species at the adult plant stage. IL pau16061 showed resistance to leaf rust only, whereas IL pau16058 was resistant to both leaf and stripe rust pathotypes throughout plant life. Molecular profiling of these ILs with simple sequence repeat (SSR) markers indicated that alien introgressions were mainly terminal and very few were interstitial. Identification of linked markers with advanced genomic technologies will aid in marker-assisted pyramiding of alien genes in cultivated wheat background. 相似文献

The ability to differentiate thoracic masses of mediastinal and pulmonary origins is often confounded by their complex spatial relationship. The objectives of this retrospective, observational cross‐sectional study were to assess radiographic differentiation of mediastinal versus pulmonary masses, and to determine if there are any correlations with specific radiographic findings. Thoracic radiographs of 75 dogs and cats with mediastinal and/or pulmonary masses identified on CT were reviewed. Radiographic studies were anonymized, randomized, and reviewed twice by three reviewers. Reviewers categorized the origin of each mass(es) as mediastinal, pulmonary, or both. On the second review, the presence or absence of 21 different radiographic findings was recorded for each mass. Agreement between the radiographic and CT categorization of mass origin, as well as inter‐ and intraobserver agreement, was calculated. Overall agreement between radiographs and CT was moderate for both mediastinal (68.6%) and pulmonary masses (63%). Overall, interobserver agreement was moderate (κ = 0.50‐0.74), with moderate to strong intraobserver agreement (κ = 0.58‐0.93). Masses within the mediastinum were significantly more likely to displace other mediastinal structures. Alternatively, masses lateral to midline and in the caudal thorax were found to be significantly positively correlated with a pulmonary origin. The results of this study highlight the limitations of radiography for differentiation of mediastinal and pulmonary masses, with mass location and displacement of other mediastinal structures potentially useful for radiographic findings that may help improve accuracy. 相似文献

Fifteen male Murrah buffalo calves (15–18 months, 227.98?±?4.44 kg body weight) were distributed randomly in to three equal groups and fed solely on either berseem (G1), oats (G2), or chicory fodder (G3). A digestibility trial followed by methane measurement using SF6 tracer technique was conducted. No significant difference was observed in nutrient intake; however, crude protein (CP) intake was lower in G2 (0.35 kg) than G1 (0.7) and in G3 (0.71) and non-fibrous carbohydrates (NFC), and neutral detergent insoluble CP (NDICP) intake was significantly (p?<?0.05) higher in G3 (1.54 and 0.31 kg) followed by G2 (1.27 and 0.2 kg) and G1 (1.06 and 0.18 kg). The digestible dry matter, organic matter, neutral detergent fiber, and ether extract intake was similar in all the groups, whereas the digestible CP and NFC intake was lower in G2 compared to G1 and G3. Chicory- and berseem-fed groups emitted 12.2 and 5.2 % less methane than oats-fed group. However, no significant difference was observed in the absolute methane loss and methane loss as percentage of energy intake (p?>?0.05) among the groups. There was positive correlation between nutrient intake and total methane production. However, an inverse relationship was observed between total digestible carbohydrate intake and methane production (g/kg dry matter intake). The following regression equations were developed to estimate methane production: methane (g/kg BW) = 128.8553 + (167.7456 × dNDFI) + (216.32 × dCPI) ? 1. Poaching of peacocks, the national bird of India, is illegal. People kill this beautiful pheasant bird for tail feathers and mix the meat with chicken or turkey. Differentiation of the meat of these species is essential in order to address the ambiguity about the origin of the sample. 2. The present study was carried out to investigate the use of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) of mitochondrial 12S rRNA gene for identification of these species. 3. Peacock mitochondrial 12S rRNA partial gene was amplified using universal primers, cloned and characterised. It was found to be 446 nucleotides long. 4. Sequence analysis revealed 86.8 and 84.1% similarity with reported turkey and chicken sequences, respectively. Sequence and phylogenetic analysis showed that the peacock is much closer to the turkey than the chicken. 5. PCR-RFLP of 446 bp amplicon using commonly available restriction enzymes AluI and Sau3AI produced a differential pattern for identifying these poultry species unambiguously. 相似文献

Twelve of 46 female buffaloes with abdominal disorders were diagnosed with omasal impaction. They had been fed finely chopped machine-prepared straw. They were characterised by anorexia, an absence of defecation, abdominal distension, ruminal hypomotility or atony and a suspension of rumination. Omasal impaction was confirmed upon left flank laparorumenotomy on the basis of the size of the omasum and the consistency of its contents. After ruminal evacuation, a long flexible pipe was introduced through the reticulo-omasal orifice and the omasal contents were flushed back into the rumen with water under moderate pressure. Hyponatraemia, hypochloraemia, hypokalaemia and hypophosphataemia were consistent features in most cases. However, two buffaloes that later died had lower levels of plasma chloride, no reticulo-omasal orifice tone and were in an advanced stage of pregnancy. The level of total protein in peritoneal fluid was higher than normal, but the total white cell count was within the normal range. All the animals started passing faeces 36 to 48 hours after surgery. The presence of reticulo-omasal orifice tone and a plasma chloride level above 75 mmol/l were indicators of a good prognosis. 相似文献

Gluten strength is an important characteristic, determining the end product quality of durum wheat semolina. To identify the genetic basis of gluten strength in North Dakota durum cultivars, a doubled haploid population was developed from the cross of a weak gluten cultivar ‘Rugby’ and a strong gluten cultivar ‘Maier’. A framework linkage map consisting of 228 markers was constructed and used with phenotypic data on gluten strength (measured by sedimentation volume) to conduct single- and two-locus QTL analyses. Only one consistent QTL (QGs.ndsu-1B) contributing up to 90% of the phenotypic or 93% of the genotypic variation was detected on 1BS. No QTL × QTL or QTL × environment interactions were observed. The QGs.ndsu-1B was flanked by two DArT markers which were converted to STS markers and used along with SSR and EST-SSRs to develop a map of 1BS. QTL analysis delineated QGs.ndsu-1B in a 7.3 cM region flanked by an STS marker (STS-wPt2395) and a SSR marker (wmc85). The adapted background of this material and availability of PCR-based markers closely associated with this locus represent invaluable resources for marker-assisted introgression of gluten strength into other durum wheat varieties. Well-aligned macroscopic fibers composed solely of single-walled carbon nanotubes (SWNTs) were produced by conventional spinning. Fuming sulfuric acid charges SWNTs and promotes their ordering into an aligned phase of individual mobile SWNTs surrounded by acid anions. This ordered dispersion was extruded via solution spinning into continuous lengths of macroscopic neat SWNT fibers. Synergism of an oil of Azadirachta indica, a powdered extract of Allium sativum bulbs and an oleoresin of Zingiber officinale rhizomes by piperonyl butoxide and MGK-264 was studied against the snails Lymnaea acuminata and Indoplanorbis exustus. The active components of these plant-derived molluscicides, respectively azadirachtin, allicin and [6]gingerol, were also combined with these synergists. Both piperonyl butoxide and MGK-264 enhanced the toxicity of all of the test compounds. Sarcophine-diol (SD), one of the structural modifications of sarcophine, has shown chemopreventive effects on 12-dimethylbenz(a)anthracene-initiated and 12-O-tetradecanoylphorbol-13-acetate-promoted skin tumor development in female CD-1 mice. The objective of this study was to determine the chemopreventive effects of SD on UVB-induced skin tumor development in hairless SKH-1 mice, a model more relevant to human skin cancer, and to determine the possible mechanisms of action. Carcinogenesis was initiated and promoted by UVB radiation. Female hairless SKH-1 mice were divided into two groups having 27 mice in each group: control and SD treatment. The control group was topically treated with 100 μL acetone and SD treatment group was topically treated with SD (30 μg/100 μL in acetone) 1 hour before each UVB radiation for 32 weeks. Tumor counts were recorded on a weekly basis for 30 weeks. Effects of SD on the expression of caspases were investigated to elucidate the possible mechanism of action. The proteins from epidermal homogenates of experimental mice were used for SDS-PAGE and Western blotting using specific antibodies against caspase-3, caspase-8 and caspase-9 respectively. TUNEL assay was used for determining DNA fragmented apoptotic cells in situ. Results showed that at the end of experiment, tumor multiplicity in control and SD treatment groups was 25.8 and 16.5 tumors per mouse respectively. Furthermore, Topical treatment of SD induced DNA fragmented apoptotic cells by upgrading the expressions of cleaved caspase-3 and caspase-8. This study clearly suggested that SD could be an effective chemopreventive agent for UVB-induced skin cancer by inducing caspase dependent apoptosis. 相似文献
