The IgG response of goats experimentally infected with RH Toxoplasma gondii has been analysed using an indirect ELISA and Western-blot analysis. Specific IgG antibodies were first detected at 14 days post-inoculation (p.i.), reaching a peak by day 35 p.i. and showing slight fluctuations until the end of the experiment (91 p.i.). Specific IgG showed a reactivity over a whole range of peptides (125-24 kDa approximately), but the highest reactivity was observed against a group of antigens with a molecular weight between 34 and 28 kDa, in particular against a 30 kDa fraction which is considered to represent the major surface protein of T. gondii named p30 or SAG-1. 相似文献

Sixty hybrid Yorkshire-Landrace penned sows, 30 with eutocic farrowing and 30 experiencing a dystocic parturition, were studied to evaluate the obstetric and neonatal outcomes to low doses of oxytocin administered at advanced stages of parturition. Animals in each group were randomly subdivided into 2 subgroups: 15 eutocic and 15 dystocic sows received oxytocin 0.083 IU/kg (equivalent to 1 IU/12 kg body weight), administered intramuscularly after the delivery of the 5th piglet; the other 15 eutocic and 15 dystocic sows received saline solution intramuscularly at the same time. Oxytocin decreased the number of intrapartum deaths by approximately 50% (P = 0.002). No piglet was born dead from the saline- and oxytocin-treated eutocic sows. The highest viability score was observed among piglets born to eutocic sows treated with oxytocin. In summary, this dose schedule would help to decrease the number of stillbirths in both eutocic and dystocic farrowing sows. 相似文献

Two feeding trials were conducted to determine the digestibility of a casein-based semi-purified diet and the effects of different protein levels on growth and protein use of spotted sand bass Paralabrax maculatofasciatus juveniles. For trial I, a semipurified diet with vitamin-free casein as the sole source of protein was fed three times a day to apparent satiation, for a period of 20 d. Feces were collected by siphoning each tank. The digestibility of the experimental diet was high: 97% for protein, 89% for lipids, and 84% for gross energy, whereas that of organic matter was 78%. For trial II, seven diets were formulated using vitamin-free casein at graded levels (25, 30, 35, 40, 45, 50, and 55% protein). Triplicate tanks for each dietary treatment were stocked with fish and fed by hand three times a day to apparent satiation for 6 wk. Perfomance of fish fed the different diets was evaluated for survival, percent weight gain, specific growth rate, feed conversion ratio, and protein efficiency ratio. Survival was 100% for all treatments. Growth of spotted sand bass juveniles increased as the dietary protein increased, but no evidence of reaching a plateau was found. The daily feed intake values showed an inverse relation to the protein content of the diets. The feed conversion ratio did not differ among diets containing 40% protein or greater. The results indicate that spotted sand bass juveniles with 2.5-g mean weight need at least 55% dietary protein for best growth when casein is the sole protein source. However, in terms of feed conversion ratio, the requirement apparently could be lower. 相似文献

A partial characterization of polyphenol oxidase (PPO) activity in raspberry fruits is described. Two early cultivars harvested in May/June (Heritage and Autumm Bliss) and two late cultivars harvested in October-November (Ceva and Rubi) were analyzed for PPO activity. Stable and highly active PPO extracts were obtained using insoluble poly(vinylpyrrolidone) (PVP) and Triton X-100 in sodium phosphate, pH 7.0 buffer. Polyacrylamide gel electrophoresis of raspberry extracts under nondenaturing conditions resolved in one band (R(f)()(1) = 0.25). Raspberry PPO activity has pH optima of 8.0 and 5.5, both with catechol (0.1 M). Maximum activity was with D-catechin (catecholase activity), followed by p-coumaric acid (cresolase activity). Heritage raspberry also showed PPO activity toward 4-methylcatechol. Ceva and Autumm Bliss raspberries showed the higher PPO activity using catechol as substrate. 相似文献

Rabies remains an important public health issue in Bolivia, South America. Public concern and fears are most focussed on dogs as the source of rabies. The objective of the present study was to assess immunity of an inactivated suckling mouse brain vaccine against canine rabies used for the official vaccination campaigns under field conditions in an endemic area of rabies in Bolivia. A total of 236 vaccinated and 44 unvaccinated dogs in Santa Cruz de la Sierra, selected using stratified random sampling, were investigated in order to obtain owned dog characteristics and antibody titres against rabies in April 2007. The proportion of vaccinated dogs with an antibody titre exceeded the protection threshold value of 0.5 EU/ml was 58% [95% confidence intervals (CI): 52-65], indicating that vaccination is likely to elicit an antibody response (odds ratio 6.3, 95% CI: 1.2-11.5). The range of geometric mean of antibody titre for vaccinated dogs (0.89 EU/ml; 95% CI: 0.75-1.04) was considered to meet the minimal acceptable level indicating an adequate immune response to the vaccine. However, the titre level was not satisfactory in comparison with the results from other field investigations with inactivated tissue culture vaccines. It is recommended for public health authorities to (1) consider modernizing their vaccine manufacturing method because the level of immunity induced by the current vaccine is comparably low, (2) conduct frequent vaccination campaigns to maintain high levels of vaccination coverage, and (3) actively manage the domestic dog population in the study area, which is largely responsible for rabies maintenance. 相似文献

The study developed models for predicting the post-fire tree survival in Catalonia. The models are appropriate for forest planning purposes. Two types of models were developed: a stand-level model to predict the degree of damage caused by a forest fire, and tree-level models to predict the probability of a tree to survive a forest fire. The models were based on forest inventory and fire data. The inventory data on forest stands were obtained from the second (1989–1990) and third (2000–2001) Spanish national forest inventories, and the fire data consisted of the perimeters of forest fires larger than 20 ha that occurred in Catalonia between the 2nd and 3rd measurement of the inventory plots. The models were based on easily measurable forest characteristics, and they permit the forest manager to predict the effect of stand structure and species composition on the expected damage. According to the stand level fire damage model, the relative damage decreases when the stand basal area or mean tree diameter increases. Conversely, the relative stand damage increases when there is a large variation in tree size, when the stand is located on a steep slope, and when it is dominated by pine. According to the tree level survival models, trees in stands with a high basal area, a large mean tree size and a small variability in tree diameters have a high survival probability. Large trees in dominant positions have the highest probability of surviving a fire. The aim of this study was to define the dendrometric and dasometric parameters of five Mediterranean bushy plant species, to make their environmental assessment and to determine correlations with LIDAR data. The species studied were: Quercus coccifera, Rosmarinus officinalis, Ulex parviflorus, Cistus albidus, and Erica multiflora. We calculated the global form factor for each species so as to be able to determine the volume of the plants by measuring their base diameter and length. The model volume closest to the real volume was the cylinder, which had form factor values of between 0.8 and 2.33. We also studied the evolution of the moisture content of the vegetation after cutting. The initial values were from 20 to 30%, with a dry density of 1 t/m3. We defined an occupation factor to determine the relationship between the real volume occupied by the plants in an area and a model volume (hemisphere, paraboloid, cone, and cylinder). The occupation factor was similar for the five species studied in dm3 of material per m3 of (model) apparent volume: 3.5 dm3/m3 for the cylinder; 12.5 dm3/m3 for the cone; 8.5 dm3/m3 for the paraboloid; and 14.5 dm3/m3 for the hemisphere. These factors enabled us to calculate the biomass contained in an apparent volume of bushes (including materials and hollows) by density. The prediction models tested in this study will thus serve to determine the biomass of a bushy forest area when the Canopy Height Model (CHM) is calculated from LIDAR data. 相似文献

Summary The meiotic pairing behaviour at metaphase I of a Triticum aestivum×Triticum monococcum hybrid has been studied by means of the C-banding technique to ascertain the homology between the chromosomes in the A genome of the two species. The technique allowed the A and B genome chromosomes and the 2D, 3D and 5D chromosomes to be identified. Differences in the level of chromosome pairing in the A genome were noted. The T. monococcum 4A chromosome did not pair with any of the T. aestivum chromosomes in any of the metaphase I cells analysed. Two reciprocal translocations between the 2B and 2D chromosomes on one side and the 2A and 3D on the other side have been identified. The usefulness of the C-banding technique in the study of chromosome homology among species related to wheat is discussed. 相似文献

Approaches are needed to broaden the genetic base and improve earliness and yield potential of large-seeded beans under sustainable cropping systems. The objective of this research was to develop adapted dwarf bean populations having a commercial seed quality and yield suitable for the production in the South of Europe. The original base populations were produced from crosses between genotypes within each Mesoamerica, Nueva Granada and Peru races, and between Peru and Nueva Granada, and Mesoamerica and Nueva Granada races. Visual mass selection for plant performance was practised in the F2 and F3 generations. In the F4 and F5, single plants were harvested under two cropping systems (sole cropping and intercropping with maize). From F4, selection was based on precocity, combined with seed yield and seed commercial type. The F4:7 selected lines from each original population were compared with their parents and five checks at four environments and two cropping systems. Differences among environments, populations, parents and checks were observed for all traits. Under intercropping with maize, there was a 50% reduction in seed yield. Yield of parents and checks belonging to Andean South American races, intraracial (Nueva Granada × Nueva Granada) and interracial (Nueva Granada × Peru) populations, was higher than that of those of Middle American origin. Intraracial crosses within large-seeded Andean South American (Peru race) and Middle American gene pools (Mesoamerica race) did not produce lines yielding more than the highest yielding parent. Only two large-seeded lines selected from crosses between small- and large-seeded gene pools out-yielded the best parent and check cultivar. 相似文献

Grapefruit growers in the tropics require information about existing and new citrus cultivars with high productivity potential.
The objective of this study was to determine the growth, yield, and fruit quality performance of seven pigmented and four
white grapefruit cultivars under the dry tropic conditions of Colima, Mexico. The trees were budded on sour orange (Citrus aurantium L.) rootstock and planted at a distance of 8 × 4 m. ‘Oroblanco’ and ‘Marsh Gardner’ white-fleshed grapefruit cultivars and
‘Chandler’, a pink-fleshed pummelo, were the largest trees with the greatest height (5.0–5.6 m), canopy diameter (6.2–6.3 m),
trunk diameter (21.9–23.3 cm), and canopy volume (109–123 m3). Lower height (4.3–4.8 m) and canopy volume (73–96 m3), but with similar canopy diameter to the previously mentioned cultivars, were recorded for the remaining pigmented cultivars.
‘Chandler’ pummelo and four pigmented grapefruit cultivars (‘Shambar’, ‘Río Red’, ‘Ray Ruby’, and ‘Redblush #3’) had yearly
productions of 34.8, 34.9, 34.1, 32.7, and 30.6 ton ha−1, respectively. The most productive white grapefruit cultivar was ‘Marsh Gardner’ (30.5 ton ha−1). Grapefruit cultivars having the largest fruit size showed a higher inverse relationship between fruit weight and yield
than those with small fruit. Most genotypes had higher values of fruit weight, juice content, and maturity index than those
required by the local market. The most promising grapefruit cultivars based on their acceptable growth, yield superior to
30 ton ha−1, and acceptable fruit color were ‘Río Red’, ‘Shambar’, ‘Ray Ruby’, and ‘Redblush #3’. 相似文献
