In this study, in-house isolated laccase isoforms, i.e., Lac-I and Lac-II of the basidiomycete Pycnoporus sanguineus (CS43), were evaluated in relation to their Remazol Brilliant Blue R (RBBR) dye degradation capacity. A modified Dhouib medium additionally supplemented with 3% ethanol as a secondary inducer was used to propagate P. sanguineus CS43 for enhanced production of laccase under liquid state fermentation. The crude laccase extract was purified by passing through ion exchange diethylaminoethanol (DEAE)-Sepharose and gel filtration-based Sephadex G-200 column chromatography. The purified laccase fractions were subjected to the electrophoresis, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed two laccase isoforms Lac-I and Lac-II with 66 and 68 kDa, respectively. To explore the industrial applicability, for RBBR dye, degradation efficiencies ranged from 82 to 88% after 3 h of incubation for both; Lac-I and Lac-II at both concentrations were recorded. However, with 8 U/mL, the degradation ranged between 70 to 80% during the first 5 min of incubation. Enhanced degradation of RBBR dye was obtained in the presence of violuric acid and N-hydroxypthalamide as laccase mediators. Finally, using RBBR as a substrate kinetic characterization of both Lac-I and Lac-II isoforms was performed that revealed Km (0.243 and 0.117 mM for Lac-I and Lac-II) and Vmax (1.233 and 1.012 mM/Sec for Lac-I and Lac-II) values, respectively. 相似文献

Soil physico-chemical properties, biomass production, and root density are considered key factors indicating soil health in an agroecosystem. The soil physico-chemical changes and plant growth (e.g., shoot biomass production and root density) in a 6-year cultivation of plant species used as green manure in a sandy soil from Tropical ecosystem, North-eastern Brazil, were investigated between July and December 2019.
Material and methods
We characterized soil physical and chemical properties, shoot biomass production, and root density under ten plant species used as green manure: Brachiaria decumbens Stapf. cv. Basilisk, Canavalia ensiformis (L.) DC, Crotalaria juncea L., Crotalaria ochroleuca G. Don, Crotalaria spectabilis Roth, Lablab purpureus (L.) Sweet, Mucuna pruriens (L.) DC, Neonotonia wightii (Wight & Arn.) J.A. Lackey, Pennisetum glaucum L., and Stilozobium aterrimum Piper and Tracy.
Results and discussion
The highest values of soil pH, exchangeable cations, CEC, and soil available water capacity were found on the plots where Poaceae plants were cultivated, whereas for H++Al3+, C.E.C., soil available water, and soil available water capacity were found on the plots where Fabaceae plants were cultivated. On the plots where C. ensiformis and N. wightii were cultivated, we found the highest shoot dry biomass and root density, respectively. The results highlight the importance to consider plant species from both Poaceae and Fabaceae family used as green manure as soil conditioner (by promoting soil fertility, nutrient cycling, and hydraulic properties into plant root zone), and thus creating a positive plant-soil feedback.
Conclusions
Our findings suggest that (1) a consecutive green manure practice without any input of fertilizers after 6 years changed positively both soil physical and chemical properties, and improve plant growth (e.g., shoot dry biomass and root density) in tropical savanna climate conditions; and (2) by altering soil fertility, both Poaceae and Fabaceae plants used as green manure may create a sustainable cycle into the soil profile thus promoting soil health.
The effect of simultaneous drought and heat stress on free amino acid levels was compared in wild type and transgenic soybean (Glycine max (L.) Merr cv Ibis) plants transformed with the cDNA coding for the last enzyme of Pro biosynthesis, l-Delta(1)-pyrroline-5-carboxylate reductase (EC 1.5.1.2), in sense and antisense directions. The most rapid increase in Pro content was found in the sense transformants that exhibited the least water loss, while the slowest elevation of Pro levels was detected in the antisense transformants that exhibited the greatest water loss during stress. Correspondingly, the level of the Pro precursors Glu and Arg was higher in sense transformants and lower in antisense ones compared to the wild type plants during the initial part of the stress. Interestingly, genetic manipulation of Pro levels also affected the stress-induced changes in the concentration of several other amino acids, which indicates the coordinated regulation of their metabolic pathways. 相似文献
One hundred and sixty-two Capsicum genotypes were evaluated for powdery mildew (Leveillula taurica) resistance, following inoculations with a suspension of 5 × 104 conidia mL−1 on 10-leaved to 12-leaved plants. Genotypes were graded into five resistance classes, based on the areas under the disease progress curves calculated from disease incidence (percentage infected leaves per plant) and severity (total number of colonies per plant). Results revealed a continuum from resistance to susceptibility, with the majority (70%) of C. annuum materials being classified as moderately to highly susceptible to L. taurica. Conversely, C. baccatum, C. chinense and C. frutescens were most often resistant, indicating that resistance to L. taurica among Capsicum species is found mainly outside the C. annuum taxon. Nevertheless, some resistant C. annuum material was identified that may be useful for resistance breeding. Eight genotypes were identified as immune to the pathogen: H-V-12 and 4638 (previously reported), and CNPH 36, 38, 50, 52, 279 and 288. Only H-V-12 and 4638 are C. annuum, while all others belong to the C. baccatum taxon. Latent period of disease on a set of commercial sweet pepper genotypes varied, indicating diverse levels of polygenic resistance. The latent period progressively reduced with plant maturity, from 14·3 days in plants at the mid-vegetative stage to 8·6 days in plants at the fruiting stage. Young plants of all commercial genotypes tested at the early vegetative stage were immune, irrespective of the reaction of the genotype at later stages, demonstrating widespread juvenile resistance to L. taurica in the Capsicum germplasm. Inoculation of plants of different botanical taxa with a local isolate indicated a wide host range. Some hosts, including tomato (Lycopersicon esculentum), artichoke (Cynara scolymus) and poinsettia (Euphorbia pulcherrima), produced large amounts of secondary inoculum. We use line transect detection functions together with generalized linear and additive models to estimate detection probability
when detection on the line (“g(0)”) may not be certain. The methods provide a flexible way of modeling detection probability
for independent observer surveys, and for investigating the effects of explanatory variables. Analysis of data from an aerial
survey of pack-ice seals produced g(0) estimates substantially below 1 for some observers (it varied from 0.80 to 0.98), demonstrated a fairly complex dependence
of detection probability on covariates, and showed negative correlation between observers’ search width and their g(0). In addition to illustrating the utility of generalized additive models for capturing the effect of covariates on detection
probability, the analysis suggests that detection functions may be sufficiently variable that use of g(0) correction factors obtained from other surveys would be inadvisable. We recommend that estimation of g(0) be considered for all aerial surveys; if g(0) is found to be very close to 1, estimation from subsequent surveys under the assumption that it is 1 may be reasonable,
but without any estimation of g(0), the assumption that it is 1 is a matter of faith. 相似文献
Bovine anaplasmosis, caused by the tick-borne rickettsia Anaplasma marginale, is endemic in South Africa and results in considerable economic loss to the cattle industry. This study was designed to characterize strains of A. marginale at the molecular level from cattle raised in communal and commercial farms in the north-eastern and south-western regions of the Free State Province, South Africa, that varied in rainfall and vegetation. Seroprevalence to A. marginale was determined in 755 cattle by an Anaplasma spp. competitive enzyme-linked immunosorbent assay and ranged from 44% to 98% and was similar in both regions. While Anaplasma centrale was not targeted in this study, A. marginale infections were identified by species-specific msp1alpha polymerase chain reaction in 129 of 215 of the samples studied. Similar genetic diversity of A. marginale strains was found in both the north-eastern and south-western regions. The sequences of 29 A. marginalemsp1alpha amplicons from South African strains revealed considerable genetic diversity providing 14 new repeat sequences. However, 42% of MSP1a repeat sequences were not unique to this region. These results indicated the presence of common genotypes between South African, American and European strains of A. marginale. Cattle movement between different parts of South Africa was suggested by the presence of identical A. marginale MSP1a genotypes in north-eastern and south-western regions of the Free State Province. Control strategies for anaplasmosis in South Africa should therefore be designed to be protective against genetically heterogeneous strains of A. marginale. 相似文献
Boron (B) is transported mainly in the xylem, but by using isotopic tracers it was possible to verify that B has significant mobility in the phloem of polyols-producing species. Thus, the objective of this study was to evaluate the mobility of 10B applied to cashew plants. The experiment was conducted from 05/10 to 07/10/2010 in a greenhouse of University of São Paulo State-UNESP, Ilha Solteira Campus, State of São Paulo, Brazil. 0.5 mg of B L?1 of soil was used in the substrate. A solution of 255 mg of B L?1 was used in recently mature and fully developed leaves. The total content of B and and percentage of boron in the plant derived from fertilizer (Bppf%) were evaluated in the old and new leaves stemming at 30 and 60 days after the application of 10B. As evidenced, B applied to leaves had restricted mobility in cashew tree. However, B applied in the substrate was absorbed and translocated to young leaves. 相似文献
