Backgrounds: Most of the hepatitis C virus (HCV) infections elicit poor immune responses and 75% to 85% of cases become chronic; therefore, the development of an effective vaccine against HCV is of paramount importance. In this study, we aimed to evaluate co-administration of HCV non-Structural Protein 2 and IL-12 DNA vaccines in C57BL/6 mice. Methods: A plasmid encoding full-length HCV NS2 protein (non-structural protein 2) was generated and used to vaccinate mice. Negative control (an empty expression vector) was also employed to evaluate the background response. To investigate immune responses against vaccine, C57BL/6 mice received three doses of the vaccine with a two-week interval. Cellular immunity was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay for lymphocyte proliferation, lactate dehydrogenase release for cytotoxic T lymphocyte (CTL) activity and cytokine assay. Results: The findings demonstrated that immunization of mice with plasmid expressing HCV NS2 induced CTL response, interferon gamma production, and lymphocyte proliferation compared to negative control. The results also demonstrated that co-administration of IL-12 with the HCV NS2 plasmid induced significantly better immune response in C57BL/6 mice. Conclusion: DNA vaccine encoding HCV NS2 is an effective candidate that can trigger CTL-based immune response against HCV. In addition, the results suggested that combining the DNA vaccine approach with immune stimulatory cytokines may significantly enhance antigen-specific immune responses. Textiles, especially those made of natural fibers, are suitable medium for the growth of microorganisms which causes disease transmission, stink, colorful spots, and reduction in fabric strength. This research focuses on the antimicrobial finishing of cotton fabrics using colloidal solution of silver nanoparticles. Due to the difficulties of adding a new step to the finishing process of cotton textiles, efforts have been made to combine the antimicrobial treatment with the conventional finishing processes. For this purpose two chemical finishes of Fixapret ECO as a crosslinking agent and Cellofix ME as a resin former have been used in anti crease finishing of cotton fabric and their effects were evaluated. The properties of the samples have been investigated by measuring the resistant of samples against bacteria, crease recovery angle, abrasion, and washing fastness. The results showed that treated samples by pad-dry method have the best antibacterial effect with a direct relation between the increase in drying temperature and antibacterial properties. However, the washing and abrasion fastness were not at the acceptable level. Co-application of the colloidal solution of silver nanoparticles with the crease resistant materials improved both fastness properties while at the same time limited the direct contact between the nanoparticles and the bacteria so the antibacterial efficiency was reduced. Subsequently, it was concluded that the antibacterial finishing method should be selected according to the end uses. In addition, antibacterial treatment could be one of the multi-purpose finishes for cotton fabric.  相似文献

Background:

Ovarian tissue cryopreservation is an alternative strategy to preserve the fertility of women predicted to undergo premature ovarian failure. This study was designed to evaluate the expression of folliculogenesis-related genes, including factor in the germline alpha (FIGLA), growth differentiation factor-9 (GDF-9), follicle-stimulating hormone receptor (FSHR), and KIT LIGAND after vitrification/warming of human ovarian tissue.

Methods:

Human ovarian tissue samples were collected from five transsexual women. In the laboratory, the ovarian medullary part was removed by a surgical blade, and the cortical tissue was cut into small pieces. Some pieces were vitrified and warmed and the others were considered as non-vitrified group (control). Follicular normality was assessed with morphological observation by a light microscope, and the expression of FIGLA, KIT LIGAND, GDF-9,, and FSHR genes was examined using real-time RT-PCR in both the vitrified and non-vitrified groups.

Results:

Overall, 85% of the follicles preserved their normal morphologic feature after warming. The percentage of normal follicles and the expression of FIGLA, KIT LIGAND, GDF-9, and FSHR genes were similar in both vitrified and non-vitrified groups (P > 0.05).

Conclusion:

Vitrification/warming of human ovarian tissue had no remarkable effect on the expression of folliculogenesis-related genes. Key Words: Vitrification, Gene expression, Humans  相似文献

The effects of anethol, the major component of anise oil, on the Ca2+-dependent excitability and afterhyperpolarization (AHP) in snail neurons were examined using intracellular recording. Anethol (0.5%) significantly broadened the spike, reduced the firing frequency and enhanced the AHP amplitude. In contrast, anethol (2%) significantly increased the firing frequency and decreased the AHP. Blockade of Ca2+ channels after anethol application depolarized the membrane potential and significantly reduced the firing rate. Furthermore, in the presence of anethol (0.5%) a significant decrease in the AHP was observed by Ca2+ channels blockage. Here, anethol-induced functional modification of Ca2+ and Ca2+-activated K+ channels is suggested.  相似文献

Little information is available regarding the effect of sewage sludge biochar on soil properties and crop yield. Thus, our objective was to evaluate the effect of sewage sludge (S) and its biochar (B) on maize shoot yield, nutrients and heavy metals uptake in two calcareous soils. The amendments were applied at the rates of 0, 10, 20 and 40 Mg ha?1. Moreover, NK treatment was included to compare the effects of S and B with conventional fertilization. At harvest time, plant shoots and soil samples were collected for yield, nutrients uptake and chemical analyses. The highest shoot dry matter was obtained in the S treatment. The B application in the clay loam and loam soils resulted in 5.2% increment and 17.7% decrement of shoot dry matter relative to the control, respectively. Shoot dry matter in the NK treatment was significantly higher than in the control. B application decreased Fe, Zn, Mn, Cu and Pb uptake by maize shoot. DTPA-extractable Pb in B-amended soils was lower than in control, while an inverse trend was obtained for available Fe, Zn, Mn and Cu. Biochar application at the rate of 7.3 Mg ha?1 might be suggested for maize cultivation in clay loam soils.  相似文献

BACKGROUND: Phelebotumus papatsi is considered to be the main vector of zoonotic cutaneous leishmaniasis as well as sand fly fever in Iran. There are several measures for vector control, with emphasis on insecticides. The objective of this study was to determine the baseline susceptibility of this vector to the commonly used insecticides in an endemic focus of the disease in central Iran. P. papatasi collected from the field were used for susceptibility status. Its baseline susceptibility to DDT and pyrethroids was assessed on about 6866 specimens collected from Badrood rural district, Esfahan Province, Iran, during the summer of 2010. The LT50 and LT90 values were measured according to the World Health Organisation (WHO) test using probit analysis and regression lines. RESULTS: Results of tests against female P. papatasi revealed LT50 values to DDT 4%, permethrin 0.75%, deltamethrin 0.1%, cyfluthrin 0.15% and lambdacyhalothrin 0.05% of 1104.97, 182.35, 26.79, 15.42 and 1.48 s respectively. The figures for male P. papatasi were 973.51, 59.5, 4.4, 2.65 and 1.5. CONCLUSION: The results of this study provide a guideline for implementation of different vector control measures. Furthermore, guidelines are needed for monitoring and evaluation of insecticide susceptibility tests against sand flies. Copyright © 2012 Society of Chemical Industry  相似文献

Potassium (K+) directly released from primary K‐bearing minerals can contribute to plant nutrition. The objective of this research was to assess short‐term K+ release and fixation on a range of intensively cropped calcareous soils. Potassium sorption and desorption properties and the contributions of exchangeable‐K+ (EK) and nonexchangeable‐K+ (NEK) pools to K+ dynamics of the soil‐solution system was measured using a modified quantity‐to‐intensity (Q : I) experiment. Release and fixation of K+ were varied among soils. The relation between the change in the amount of NEK during the experiment and the initial constrain was linear, and soil ability for K+ release and fixation (β) for all soils varied from 0.041 to 0.183, indicating that 4% to 18% of added K+ converted to NEK when fixation occurred. The equilibrium potential buffering capacity (PBC) for K+ derived from Q : I experiments had significant correlation (r = 0.75, p < 0.01) with β, indicating that PBC depends not only on exchange properties but also on release and fixation properties. The depleted soils showed higher β value than the other soils, indicating much of the added K+ was converted to NEK in case of positive constraint. The range of the amount of EK which was not in exchange equilibrium with Ca (Emin) in the experimental conditions was large and varied from 0.68 to 9.00 mmol kg–1. On average, Emin amounted to 64% of EK. This fraction of EK may not be available to the plant. The parameters obtained from these short‐term K+ release and fixation experiments can be used in plant nutrition.  相似文献

Abstract

Thirty-four sugar beet drought-tolerant half-sib families were screened in greenhouse experiment under saline (16?dSm?1) and normal conditions, and a subset of 10 tolerant genotypes were selected for evaluating of yield-related traits, Na+, K+, and α-amino-N nitrogen contents under saline field condition (12?dSm?1). Drought-tolerant genotypes showed a different tolerance level under salt stress. Significant correlation was observed between EP2s and STIEP2 in greenhouse with root yield (RY) in saline field condition. It should be possible that a simple measurement at 16?dSm?1 in the greenhouse can be used as main part of sugar beet salt-tolerant breeding program. In total, genotypes SBSI-DR I-HSF14-P.23 (# 16) and 436B-HSF9-P.27 (# 26) were recognized as salt- and drought-tolerant genotypes with low Na+ and K+ contents in root and high white sugar content (WSC) and RY in field. A larger panel of genotypes to validate this kind of association is necessary.  相似文献
