In the summer of 2011 Schmallenberg virus (SBV), a Culicoides-borne orthobunyavirus, emerged in Germany and The Netherlands and subsequently spread across much of Europe. To draw inferences about the transmission of SBV we have developed two models to describe its spread within and between farms. The within-farm model was fitted to seroprevalence data for cattle and sheep farms in Belgium and The Netherlands, with parameters estimated using approximate Bayesian computation. Despite the short duration of viraemia in cattle and sheep (mean of 3–4 days) the within-farm seroprevalence can reach high levels (mean within-herd seroprevalence >80%), largely because the probability of transmission from host to vector is high (14%) and SBV is able to replicate quickly (0.03 per day-degree) and at relatively low temperatures (threshold for replication: 12.3 °C). Parameter estimates from the within-farm model were then used in a separate between-farm model to simulate the regional spread of SBV. This showed that the rapid spread of SBV at a regional level is primarily a consequence of the high probability of transmission from host to vector and the temperature requirements for virus replication. Our results, obtained for a region of the UK in a typical year with regard to animal movements, indicate that there is no need to invoke additional transmission mechanisms to explain the observed patterns of rapid spread of SBV in Europe. Moreover, the imposition of movement restrictions, even a total movement ban, has little effect on the spread of SBV at this scale.  相似文献

Activation of N-methyl-d-aspartate subtype glutamate receptors (NMDARs) is required for long-term potentiation (LTP) and long-term depression (LTD) of excitatory synaptic transmission at hippocampal CA1 synapses, the proposed cellular substrates of learning and memory. However, little is known about how activation of NMDARs leads to these two opposing forms of synaptic plasticity. Using hippocampal slice preparations, we showed that selectively blocking NMDARs that contain the NR2B subunit abolishes the induction of LTD but not LTP. In contrast, preferential inhibition of NR2A-containing NMDARs prevents the induction of LTP without affecting LTD production. These results demonstrate that distinct NMDAR subunits are critical factors that determine the polarity of synaptic plasticity.  相似文献

Glycoprotein G of the vesicular stomatitis virus triggers membrane fusion via a low pH-induced structural rearrangement. Despite the equilibrium between the pre- and postfusion states, the structure of the prefusion form, determined to 3.0 angstrom resolution, shows that the fusogenic transition entails an extensive structural reorganization of G. Comparison with the structure of the postfusion form suggests a pathway for the conformational change. In the prefusion form, G has the shape of a tripod with the fusion loops exposed, which point toward the viral membrane, and with the antigenic sites located at the distal end of the molecule. A large number of G glycoproteins, perhaps organized as in the crystals, act cooperatively to induce membrane merging.  相似文献

Lymphatic metastasis contributes to mortality from solid tumors. Whether metastasizing cancer cells reach lymph nodes via intratumor lymphatic vessels is unknown. Here, we examine functional lymphatics associated with mouse tumors expressing normal or elevated levels of vascular endothelial growth factor-C (VEGF-C), a molecule that stimulates lymphangiogenesis. Although VEGF-C overexpression increased lymphatic surface area in the tumor margin and lymphatic metastasis, these tumors contained no functional lymphatics, as assessed by four independent functional assays and immunohistochemical staining. These findings suggest that the functional lymphatics in the tumor margin alone are sufficient for lymphatic metastasis and should be targeted therapeutically.  相似文献

Aggregate stability often exhibits a large inter-annual and seasonal variability which occurs regardless of residue treatments and is often larger than the differences between soils or cropping systems. Variations in soil moisture and seasonal stimulation of microbial activity are frequently cited as the major causes. The goal of this paper was to evaluate the effects of drying-rewetting cycles on aggregate stability and on its main microbially mediated agents from a mechanistic point of view. The 3-5 mm aggregates of a silty soil were incubated at 20 °C for 63 days with the following treatments and their combinations: (i) with or without straw input and (ii) with or without exposure to four dry-wet cycles. Microbial activity was followed by measuring the soil respiration. We estimated the microbial agents of aggregate stability measuring hot-water extractable carbohydrate-C, microbial biomass carbon and ergosterol content. We measured the water drop penetration time to estimate the hydrophobicity and aggregate stability according to Le Bissonnais [1996. Aggregate stability and assessment of soil crustability and erodibility: I. Theory and methodology. European Journal of Soil Science 47, 425-437] to distinguish three breakdown mechanisms: slaking, mechanical breakdown and microcracking. The addition of straw stimulated microbial activity and increased the resistance to the three tests of aggregate stability, enhancing the internal cohesion and hydrophobicity of aggregates. All the estimated microbial agents of aggregate stability responded positively to the addition of organic matter and were highly correlated with aggregate stability. Fungal biomass correlated better with aggregate stability than total microbial biomass did, showing the prominent role of fungi by its triple contribution: physical entanglement, production of extracellular polysaccharides and of hydrophobic substances. Dry-wet cycles had less impact on aggregate stability than the addition of straw, but their effects were more pronounced when microbial activity was stimulated demonstrating a positive interaction.  相似文献

The legal method (polarimetric measurement) for the determination of sucrose content and the wet chemical analysis for the quality control of sugar beet uses lead acetate. Because heavy metals are pollutants, the law could forbid their use in the future. Therefore, near-infrared spectroscopy (NIRS) was evaluated as a procedure to replace these methods. However, there are alternatives to lead clarification, such as the use of aluminum salts, which have been applied at many sugar companies. The real advantage of NIRS is in speed and ease of analysis. The aim of this study was to determine simultaneously the concentration of several components which define the industrial quality of beets. The first objective was the determination of sucrose content, which determines the sugar beet price. The standard error of prediction (SEP) was low: 0.11 g of sucrose/100 g of fresh beet. NIRS was also able to determine other beet quality parameters: brix, marc, glucose, nitrogen, sodium, potassium, sugar in molasses (i.e. sucrose in molasses), and juice purity. The results concerning brix, marc, sugar in molasses, and juice purity were satisfactory. NIRS accuracy was lower for the other parameters. Nevertheless, RPD (ratio standard deviation of concentration/SEP) and RER (ratio concentration range/SEP ratio) show that NIRS might be used for the sample screening on nitrogen, potassium, sodium, and glucose content.  相似文献

Rice cultivation has frequently been suggested to provide an important wetland habitat for waterbirds. However, in contrast to most other regions of the Mediterranean, the Camargue still has a substantial amount of surface area of natural marshes. Thus, we compared the abundance, species richness, and community composition of waterbirds in ricefields and natural marshes of the Camargue, during a one-year study. Based on surveys conducted at 4–7 day intervals in 1997–1998, our results suggest that natural marshes had substantially greater abundance of waterbirds, with ca. 99% of the individuals having been observed in natural marshes. Estimates of species richness and associated parameters further indicated that ricefields were clearly less rich than natural marshes. Although our results were rather striking for the Camargue, they may not be applicable to other regions because of the relatively high availability of natural marshes and differences in management of ricefields.  相似文献
