Here, two Punica species, viz., P. protopunica Balf. fil., reported as native to Socotra, and P. granatum L., were compared for the first time. Analysis of one P. protopunica and eleven P. granatum accessions was performed using three molecular markers, i.e., sequence related amplified polymorphism (SRAP), target region amplification polymorphism (TRAP), and intron targeted amplified polymorphism (ITAP), along with analysis of pgWD40 sequences, a gene involved in anthocyanin biosynthesis. All markers revealed the relationship between the two species and placed them at 33% similarity. SRAP, TRAP, and ITAP generated a total of 299, 260, and 160 bands, respectively. Of these, 78, 74, and 41 bands were specific for P. protopunica, and 92, 85, and 57 bands, respectively, were shared between both species. Sequence analysis of pgWD40~870 bp amplicons exhibited 100% identity among P. granatum accessions and 98% identity to that of P. protopunica. Phylogenetic analysis of WD40 sequences from monocot and dicot species, including both Punica species confirmed the relation between P. protopunica and P. granatum, supporting earlier reports that P. protopunica could be an ancestral species of P. granatum. Furthermore, the genetic diversity among and within P. granatum accessions from Egypt (3), Mexico (5), and Yemen (3) was assessed. Molecular marker-based relationships among region-bulked accessions was approximately the same (~90% similarity), whereas the degree of genetic variation was altered within each region. Specific bands (alleles) for accessions of each region along with those shared among them were identified. Thus, these bands could be used for pomegranate genotyping and breeding programs.  相似文献

A new collection of several Red Sea sponges was investigated for the discovery of potential breast cancer migration inhibitors. Extracts of the Verongid sponges Pseudoceratina arabica and Suberea mollis were selected. Bioassay-directed fractionation of both sponges, using the wound-healing assay, resulted into the isolation of several new and known brominated alkaloids. Active fractions of the sponge Pseudoceratina arabica afforded five new alkaloids, ceratinines A–E (2–6), together with the known alkaloids moloka’iamine (1), hydroxymoloka’iamine (7) and moloka’iakitamide (8). The active fraction of the sponge Suberea mollis afforded the three known alkaloids subereamolline A (9), aerothionin (10) and homoaerothionin (11). Ceratinine B (3) possesses an unprecedented 5,7-dibrominated dihydroindole moiety with an epoxy ring on the side chain of a fully substituted aromatic moiety. Ceratinines D (5) and E (6) possess a terminal formamide moiety at the ethylamine side chain. Subereamolline A (9) potently inhibited the migration and invasion of the highly metastatic human breast cancer cells MDA-MB-231 at the nanomolar doses. Subereamolline A and related brominated alkaloids are novel scaffolds appropriate for further future use for the control of metastatic breast cancer.  相似文献

A greenhouse experiment was conducted to assess the effects of magnetic field, arbuscular mycorrhizal fungi (AMF), and phosphorus (P) concentration in the nutrient solution (0, 5, 10, 20, or 40 mg L?1) on the mobility and accumulation of P in soil and plant tissues of basil (Ocimum basilicum L.). The experiment was designed as a factorial combination and treatments were arranged in a completely randomized design with four replicates. Magnetic field increased water-soluble P in the soil and P concentration in plant shoot by 30.0% and 13.0%, respectively, in comparison to the control. The application of magnetic field and inoculation of AMF at 10 mg P L?1 increased the P translocation efficiency by 23.3% and 17.8%, respectively. Overall, our results demonstrated that the use of magnetic field and AMF could be an effective tool for enhancing of uptake and movement efficiency of P even at low concentrations.  相似文献

Objective

To determine the absorption characteristics of fentanyl and buprenorphine administered transdermally in swine.

Study design

A randomized comparative experimental trial.

Animals

Twenty-four Yorkshire gilts weighing 27.8 ± 2.2 kg (mean ± standard deviation).

Methods

Animals were randomly assigned to different doses of transdermal patches (TPs) of fentanyl (50 μg hour?1, 75 μg hour?1 and 100 μg hour?1) or buprenorphine (35 μg hour?1 and 70 μg hour?1), once or twice. Thirteen blood samples were obtained for each TP applied. Plasma concentrations were determined, and the area under the curve, peak serum concentration (Cmax) and time to Cmax were calculated.

Results

Fentanyl: Cmax was observed at different time points: for the first TP application: 30 hours for 50 μg hour?1, 6 hours for 75 μg hour?1 and 100 μg hour?1 patches; and for the second TP application: 30 hours for 50 μg hour?1 and 36 hours for 75 μg hour?1 patches. Buprenorphine: serum concentrations were not detected for the 35 μg hour?1 patch; Cmax was observed at different times for the 70 μg hour?1 patch: 18 hours (n = 1), 24 hours (n = 3), 30 hours (n = 1) and 42 hours (n = 1) after application of the first patch and 12 hours after the second patch.

Conclusions and clinical relevance

A relevant serum concentration obtained with fentanyl TP dosed at 75 μg hour?1 or 100 μg hour?1suggests that TPs could represent an analgesia option for laboratory pigs weighing 25–30 kg. As concentrations of buprenorphine were variable, this study does not support the use of buprenorphine TPs in pigs. Consecutive fentanyl or buprenorphine TPs did not provide reliable serum concentrations. Further pharmacokinetic studies and analgesiometric tests in swine are needed to confirm the clinical adequacy of TPs.  相似文献

Many-grained mutants occurring spontaneously among their less well-endowed field mates may have appeared to early farmers as fortunate twists of fate foreboding wealth and abundance. In domesticated barley, the number of kernel rows in spike can be tripled by recessive mutant alleles at the Six-rowed spike 1 (vrs1) locus that abolish the suppression of lateral spikelet fertility. In another barley row-type, so called intermedium-spike (int), lateral floret size is often intermediate between six-and two-rowed types. Phenotypic and sequence analyses of our intermedium-spike collection revealed that other genes can increase the size of florets and even stimulate occasional grain setting in lateral spikelets. Here, we show that a complete six-rowed phenotype occurs in a diverse panel of intermedium-spike barley carrying wildtype Vrs1 in the presence of the Int-c.a allele of the intermedium spike-c (int-c) gene, previously considered only as a modifier of lateral spikelet fertility. Int-c.a-type alleles had arisen before domestication and are associated with the enlargement of lateral florets in wild barley, suggesting that natural selection/evolution acts towards reduced lateral floret size. Since Int-c.a cannot overcome the suppression of lateral florets in the genomic background of wild barleys, we infer the existence of other gene loci, at which novel alleles or allelic combinations were selected for after domestication, to increase grain number of barley independently of Vrs1.  相似文献

Bioassay-guided partition of the extract of the Red Sea sponge Pseudoceratina arabica and HPLC purification of the active fraction gave a psammaplysin dimer, psammaceratin A (1), along with psammaplysin A (2). The dimer comprises two units of psammaplysin A (2) connected via the terminal amines with an unprecedented (2Z,3Z)-2,3-bis(aminomethylene)succinamide moiety, and it represents the first dimer to be identified among the psammaplysin family. Data from 1D- and 2D-NMR and HRMS supported the chemical structures of the compounds. Psammaceratin A (1) and psammaplysin A (2) exhibited significant growth inhibition of HCT 116, HeLa, and MBA-MB-231 cells down to 3.1 μM.  相似文献

The interactions between the pathogen Ralstonia solanacearum and potato Solanum tuberosum plants were studied to investigate the reactive oxygen species metabolic system and ascorbate (ASC)-glutathione (GSH) redox cycle in response to compost application. Single potato eyepieces were germinated and grown in pots containing sandy soil with or without compost at a rate of 7.5 g kg?1 soil. Non-compost- and compost-treated plants (CTP) were inoculated with R. solanacearum 25 days after planting and then analyzed after 10 days, unless otherwise stated. The present results revealed that pathogen infection caused a remarkable decrease in plant growth related parameters and productivity and an increase in disease incidence. However, under these conditions compost had substantially improved plant growth and decreased disease incidence and bacterial population. R. solanacearum resulted in significant enhancement in the activities of NADPH oxidase, lipoxygenase, the production rate of superoxide and hydroxyl radicals, levels of hydrogen peroxide, membrane lipid peroxidation, and protein oxidation indicating the induction of oxidative stress in potato roots. However, the pathogen-mediated enhancement in indices of oxidative stress was considerably decreased by compost application, which enhanced the activities of ascorbate peroxidase (APX, EC 1.11.1.11), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), dehydroascorbate reductase (DHAR, EC 1.8.5.1) and glutathione reductase (GR, EC 1.6.4.2) in infected potato plants, implying a better ROS-scavenging activity. Data also indicated that there were general increases in ASC and GSH content in infected compost treated plants, but non-compost treated ones significantly had lower levels of such redox metabolites. In addition, significantly higher ratios of ASC/DHA (dehydroascorbate) and GSH/GSSG (glutathione disulphide) were generally found in CTP than in non-compost treated-ones. The obtained results suggest that compost provides effective protection against the Ralstonia bacterial pathogen via up-regulation of the capacity of the ASC-GSH cycle and modulation of the cellular redox status, thereby eliminating ROS damage and sustaining membrane stability.  相似文献

Protein concentrates, were isolated from three by-products, defatted soya-bean, rice-bran and protelan. The conditions of maximum protein isolation differed according to the material used. Concentrates of 55.70%, 22.41% and 57.45% protein could be obtained from soya-bean, rice-bran and protelan; respectively. Hydrogen peroxide was found to increase the isolated nitrogen from protelan and rice-bran, however, in soya-bean it was without any positive effect. Essential amino acid contents were found corresponding to the amino acid of the FAO recommended amino acid composition except for methionine in all cases and lysine in protelan.  相似文献

The present study was undertaken to investigate the genetic nature of zinc (Zn) accumulation in the ear‐leaves of maize (Zea mays L.) plants. Fourteen inbred lines were evaluated. Out of which, five inbreds were chosen, two high Zn accumulators (Rg‐5 and Rd‐2), two low Zn accumulators (Rg‐8 and G‐307) and a moderate one (K‐64). These five parental inbreds were utilized to obtain all possible ten F1 hybrids (in one direction), 10 F2, 10 Bl, and 10 B2. All genotypes were planted and Zn was estimated at 50% tasseling stage. Zinc accumulation in the present genotypes was found to be genetically controlled and affected by additive genes. Significant values were obtained for the General Combining Abilities (GCA) but not for the Specific Combining Abilities (SCA) and the high Zn accumulating parents were the best general combiners. Four genes were found to be the minimum segregating factors in the (high x low) crosses for Zn accumulation.  相似文献

To develop a cost-effective process for the production of Bacillus thuringiensis-based insecticide, it is important to cultivate the bacterial strain in rich medium to obtain the highest yields of spore-crystal complexes. It was found that cultivation of the bacterium in medium with high concentrations of glucose (50–90 g l?1) resulted in much lower bacterial spores, crystal protein and lower toxicity, when tested against Spodoptera littoralis and Anagasta kuehniella larvae. The best results were obtained with glucose concentration of 20.0 g 1?1 as 7.1 × 1011 spores ml?1 and 3.4 g l?1 of crystal protein were achieved with LC50 of 40.1 and 50.2 mg kg?1 meal against S. littoralis and A. kuehniella, respectively. However, >21% of the consumed glucose was diverted into by-product synthesis at the expense of spore-crystal protein mixture. Only 78.3% of consumed glucose was converted into spores and crystal protein. Among by-products formed, acetic acid and β-hydroxybutyric acid (PHB) were produced during the phase of active growth and glutamic acid and succinic acid during the phase of active toxin production.  相似文献
