F1-adenosine triphosphatase (ATPase) is an ATP-driven rotary molecular motor in which the central gamma subunit rotates inside a cylinder made of three alpha and three beta subunits alternately arranged. The rotor shaft, an antiparallel alpha-helical coiled coil of the amino and carboxyl termini of the gamma subunit, deeply penetrates the central cavity of the stator cylinder. We truncated the shaft step by step until the remaining rotor head would be outside the cavity and simply sat on the concave entrance of the stator orifice. All truncation mutants rotated in the correct direction, implying torque generation, although the average rotary speeds were low and short mutants exhibited moments of irregular motion. Neither a fixed pivot nor a rigid axle was needed for rotation of F1-ATPase.  相似文献

Genomic imprinting causes parental origin-specific monoallelic gene expression through differential DNA methylation established in the parental germ line. However, the mechanisms underlying how specific sequences are selectively methylated are not fully understood. We have found that the components of the PIWI-interacting RNA (piRNA) pathway are required for de novo methylation of the differentially methylated region (DMR) of the imprinted mouse Rasgrf1 locus, but not other paternally imprinted loci. A retrotransposon sequence within a noncoding RNA spanning the DMR was targeted by piRNAs generated from a different locus. A direct repeat in the DMR, which is required for the methylation and imprinting of Rasgrf1, served as a promoter for this RNA. We propose a model in which piRNAs and a target RNA direct the sequence-specific methylation of Rasgrf1.  相似文献

Degenerative disorders of motor neurons include a range of progressive fatal diseases such as amyotrophic lateral sclerosis (ALS), spinal-bulbar muscular atrophy (SBMA), and spinal muscular atrophy (SMA). Although the causative genetic alterations are known for some cases, the molecular basis of many SMA and SBMA-like syndromes and most ALS cases is unknown. Here we show that missense point mutations in the cytoplasmic dynein heavy chain result in progressive motor neuron degeneration in heterozygous mice, and in homozygotes this is accompanied by the formation of Lewy-like inclusion bodies, thus resembling key features of human pathology. These mutations exclusively perturb neuron-specific functions of dynein.  相似文献

Naturally occurring Foxp3+CD4+ regulatory T cells (Tregs) are essential for maintaining immunological self-tolerance and immune homeostasis. Here, we show that a specific deficiency of cytotoxic T lymphocyte antigen 4 (CTLA-4) in Tregs results in spontaneous development of systemic lymphoproliferation, fatal T cell-mediated autoimmune disease, and hyperproduction of immunoglobulin E in mice, and it also produces potent tumor immunity. Treg-specific CTLA-4 deficiency impairs in vivo and in vitro suppressive function of Tregs-in particular, Treg-mediated down-regulation of CD80 and CD86 expression on dendritic cells. Thus, natural Tregs may critically require CTLA-4 to suppress immune responses by affecting the potency of antigen-presenting cells to activate other T cells.  相似文献

Functional brain organization of macaque monkeys and humans was directly compared by functional magnetic resonance imaging. Subjects of both species performed a modified Wisconsin Card Sorting Test that required behavioral flexibility in the form of cognitive set shifting. Equivalent visual stimuli and task sequence were used for the two species. We found transient activation related to cognitive set shifting in focal regions of prefrontal cortex in both monkeys and humans. These functional homologs were located in cytoarchitectonically equivalent regions in the posterior part of ventrolateral prefrontal cortex. This comparative imaging provides insights into the evolution of cognition in primates.  相似文献

The purpose of this study was to investigate the strength properties of glued laminated timber composed of edge-glued laminae and to investigate the influence of edge gluing on the strength properties. Glued laminated timber composed of multiple-grade laminae (symmetrical composition, strength grade E95-F270, 10 laminations) was produced from karamatsu (Larix kaempferi) edge-glued laminae according to the Japanese Agricultural Standard. The bending, tensile, and compressive strengths of the glued laminated timber were measured. The average bending, tensile, and compressive strengths were 33.4, 24.5, and 35.9 MPa, respectively, and these values are almost equal to those of glued laminated timber composed of karamatsu single-piece laminae. It was determined that finger-joints and knots in the edge-glued laminae played a significant role in the failure of specimens. However, the use of glued edge-joints did not appear to affect the failure of specimens.  相似文献

Many observations have been made on cloned embryos and on adult clones by somatic cell nuclear transfer (SCNT), but it is still unclear whether the progeny of cloned animals is presenting normal epigenetic status. Here, in order to accumulate the information for evaluating the normality of cloned cattle, we analyzed the DNA methylation status on satellite I region in blastocysts obtained from cloned cattle. Embryos were produced by artificial insemination (AI) to non‐cloned or cloned dams using semen from non‐cloned or cloned sires. After 7 days of AI, embryos at blastocyst stage were collected by uterine flushing. The DNA methylation levels in embryos obtained by using semen and/or oocytes from cloned cattle were similar to those in in vivo embryos from non‐cloned cattle. In contrast, the DNA methylation levels in SCNT embryos were significantly higher (P < 0.01) than those in in vivo embryos from non‐cloned and cloned cattle, approximately similar to those in somatic cells used as donor cells. Thus, this study provides useful information that epigenetic status may be normal in the progeny of cloned cattle, suggesting the normality of germline cells in cloned cattle.  相似文献

In sandy fields with vegetable cultivation, fertilizer leaching may occur and it should be well-controlled. The development of a direct soil water sampler is necessary to examine solute transport and fertilizer leaching in the vadose zone, since soil water reflects timely monitoring of data more accurately than groundwater. We developed a Suction-Controlled Flux Sampler to collect infiltration soil water in a sandy soil. In the present study, we monitored fertilizer leaching in an unsaturated sandy field during the rainy season, while evaluating the sampling performance of SCFS for the sampling of infiltration water. SCFS directly collected the infiltration water effectively over a period of several months in the sandy field and recorded the Water-Collecting Efficiency from 92 to 115% under various infiltration conditions during a period of 50 d. WCE was affected by the rainfall intensity as well as by previous rainfall, which enhanced WCE. The results obtained from the use of SCFS and several sensors demonstrated that the amount of leached water remained low as long as irrigation was applied according to the cultivation manual. However, an unexpected heavy rainfall event led to fertilizer leaching. The fertilizer leaching trend was effectively monitored by several sensors inserted into the soils, while detailed analysis of the components was performed after collection by using SCFS. Direct access to infiltration water enabled to examine the infiltration process and detailed variations in the amounts of discharged anions. The sensor-equipped monitoring system together with SCFS is suitable for precise management of fertilizer and irrigation application.  相似文献

As a defense response to attacks by herbivores such as the smaller tea tortrix ( Adoxophyes honmai Yasuda), tea ( Camellia sinensis ) leaves emit numerous volatiles such as (Z)-3-hexen-1-ol, linalool, α-farnesene, benzyl nitrile, indole, nerolidol, and ocimenes in higher concentration. Attack of Kanzawa spider mites ( Tetranychus kanzawai Kishida), another major pest insect of tea crops, induced the emission of α-farnesene and ocimenes from tea leaves. The exogenous application of jasmonic acid to tea leaves induced a volatile blend that was similar, although not identical, to that induced by the smaller tea tortrix. Most of these herbivore-induced plant volatiles (HIPV) were not stored in the tea leaves but emitted after the herbivore attack. Both the adaxial and abaxial epidermal layers of tea leaves emitted blends of similar composition. Furthermore, HIPV such as α-farnesene were emitted mostly from damaged but not from undamaged leaf regions. A principal component analysis of metabolites (m/z 70-1000) in undamaged tea leaves exposed or not to HIPV suggests that external signaling via HIPV may lead to more drastic changes in the metabolite spectrum of tea leaves than internal signaling via vascular connections, although total catechin contents were slightly but not significantly increased in the external signaling via HIPV.  相似文献

Kraft pulp bleaching effluent supplemented with glucose was decolorized by a newly isolated fungusGeotrichum candidum Dec 1 (Dec 1) that showed a wide decolorizing spectrum to synthetic dyes. When the glucose concentration in the effluent was 30 g/l, the color removal and the reduction of absorbable organic halogens were 78% and 43% after 7 days culture, respectively. The average molecular weight of colored substances measured by gel filtration chromatography was lowered to less than 3000 from 5600 after 7 days culture. As the contribution of extracellular enzymes such as peroxidase (DyP), manganese peroxidase, and laccase to the decolorization of the kraft pulp bleaching effluent was small, Dec 1 appears to have a different mechanism of decolorizing kraft pulp bleaching effluent when compared with enzymes used to decolorize synthetic dyes.  相似文献
